We have used cell or tissue-specific promoters to express lymphocytic choriomeningitis virus (LCMV) proteins in selected cells in independent lines of transgenic mice. Upon adoptive transfers into these mice, MHC-restricted LCMV-specific cytotoxic T lymphocytes homed specifically to either the choroid plexus (SV40 promoter) or beta cells of the islets of Langerhans (rat insulin promoter). Polyribosilribitolphosphate (PRP), the capsular polysaccharide from Haemophilus influenzae type b, is a T-cell-independent type 2 antigen. In vitro culture of adult peripheral blood T cells with 15 micrograms/ml PRP leads to induction of interleukin-2 receptor (IL-2R) expression on up to 10% of T cells. A B lymphoblastoid cell line can provide a comitogenic, accessory signal for mitogen-treated T cells. The utilisation of glucose, glutamine, acetoacetate and D-3-hydroxybutyrate were investigated over 72 h of incubation of rat splenic lymphocytes, with and without concanavalin A. The present study was designed to determine the influence of signaled shock on splenic natural killer (NK) activity and nonspecific T-lymphocyte mitogenic responsiveness. We studied in vivo lymphocyte activation in patients who underwent cardiac operation. A large number of deoxyribonucleic acid-synthesizing cells characterized by 3H-thymidine uptake and morphologically atypical lymphocytes were found after operation in the peripheral blood samples of patients, the peak level occurring on the sixth or seventh postoperative day. We have studied the in vitro B-cell maturation induced by pokeweed mitogen of lymphocytes from seventeen patients with partial Di George syndrome. Absolute number of T and B lymphocytes were measured in the peripheral blood of 25 patients with biopsy proven HBsAg-positive Chronic Active Hepatitis (CAH), in 9 with Chronic Persistent Hepatitis (CPH), and in 25 HBsAg-negative healthy controls. In patients with CAH and cirrhosis total T lymphocytes were decreased; no difference were found between patients with CPH and normal controls. This data emphasize the importance of monitoring T-cells population in patients with HBsAg-positive CAH.
