Mammalian-evolved strains of the fungal pathogen Cryptococcus neoformans show adaptation via heritable transcript level differences specific to mouse MHC haplotype.

BACKGROUND: Organisms adapt to novel environments using changes to genome, gene expression, and protein functions. This study focused on changes that had occurred when a fungal pathogen previously encountered hosts that differed only at the major histocompatibility complex (MHC) region, loci that control immune recognition during the adaptive immune response. To investigate how this fungal pathogen adapted to the host environment, next generation sequencing data were examined from strains of Cryptococcus neoformans (C. neoformans) that had been previously passaged eight times through congenic …

Genetic basis of disease resistance to coccidiosis and necrotic enteritis in chickens.

The chicken has served as a valuable model organism for studying various biological systems, including the development and delineation of the various arms of the immune system. Central to more recent discoveries have been the characterization of the major histocompatibility complex (MHC) and the sequencing of the chicken genome. Even prior to those events, disease resistance in poultry had been linked to bird genetics, particularly in the context of the MHC and other alloantigen systems. Disease outbreaks pose significant challenges across the global poultry industry. Enteric challenges such a…

Association Analysis of the MHC in Lupus Nephritis.

Lupus nephritis (LN) is one of the most prevalent and serious complications of SLE, with significant effects on patient and renal survival. Although a large number of genetic variants associated with SLE have been identified, biomarkers that correlate with LN are extremely limited. In this study, we performed a comprehensive sequencing analysis of the whole MHC region in 1331 patients with LN and 1296 healthy controls and validated the independent associations in another 950 patients with LN and 1000 controls. We discovered five independent risk variants for LN within the MHC region, including

MHC genes and parasitism in Carassius gibelio, a diploid-triploid fish species with dual reproduction strategies.

BACKGROUND: The gibel carp is a fish species with dual reproduction modes, gynogenesis and sexual reproduction, coexisting in mixed diploid-polyploid populations. Following the Red Queen (RQ) assumption, asexual organisms are, due to their low genetic diversity, targets for parasite adaptation. Because MHC polymorphism is maintained by selection from parasites and sexual selection, MHC genes are considered as a suitable candidate for testing the RQ hypothesis. In this study, we investigated MHC variability and the selection pressure acting on MHC genes in sexual diploids and asexual triploids.

MHC haplotype involvement in avian resistance to an ectoparasite.

Research on immune function in evolutionary ecology has frequently focused on avian ectoparasites (e.g., mites and lice). However, host immunogenetics involved with bird resistance to ectoparasites has not been determined. The critical role of the major histocompatibility complex (MHC) in adaptive immunity and high genetic variation found within the MHC make this gene complex useful for exploring the immunogenetic basis for bird resistance to ectoparasites. The objective of this study was to determine if the avian MHC influenced resistance to a blood-feeding ectoparasite. Four congenic lines o…

Dendritic cells and the control of immunity: enhancing the efficiency of antigen presentation.

BACKGROUND: Relevant antigens often are known for diseases that involve the immune system. Yet purified antigens by themselves do not contro … the special mechanisms whereby DCs enhance the formation of ligands for T-cells, i.e., complexes of major histocompatibility complex (MHC) products and antigenic peptides. RESULTS: Once criteria were developed to identify and isolate DCs, several functional properties became evident. DCs are unusually potent in initiating T-cell mediated immunity in culture. In vivo, DCs are pos …

Immunogenetics of chronic liver diseases.

The genetic background of autoimmune diseases becomes more and more evident. Immunogenetics comprises the analysis of genes and their products located at the region 6p21 on the short arm of chromosome 6, which is also known as the major histocompatibility complex (MHC). MHC class I and II genes are highly polymorphic. The complement genes C2, C4A, C4B, and BF, which are also polymorphic, became known as MHC class III genes. In autoimmune hepatitis type 1, there is a dual association for white persons with either HLA-A1-B8-DR3 or HLA-DR4. In patients from Japan, autoimmune hepatitis type 1 is p…

[Erysipelas as a disease factor].

The infection with Erysipelas rhusiopathiae demonstrates that it is possible to characterize the significance of predisposing factors. The virulence of the agent is most important among the exogenous factors; it determines decisively the course of the disease: 1. paracute course as septicemia; 2. chronic course as polyarthritis; 3. subclinical course nearly without any symptoms. The immune status and the genotype of the host are predominant out of endogenous factors. The importance of immunity is known since a long time. A series of field observations supported the potential genetical influenc

Aberrant expression of class II HLA antigens by the target cell: cause or consequence of the autoimmune aggression?

Since the aberrant HLA-DR expression on thyroid follicular cells (TFC) has been found in glands already having an ongoing immune response, the possibility exists that this ectopic expression is not the primary event leading to the infiltration by autoreactive lymphocytes, but rather a consequence of that infiltration. We have explored that possibility in studies on the behaviour of TFC from normal and autoimmune glands with respect to their expression of HLA-DR antigens when they are cultured with or without autologous mononuclear cells from the intrathyroidal infiltrates or peripheral blood.

Multivariate analysis of HLA-disease associations.

A multivariate procedure is presented for the analysis of HLA-disease associations. This formulation explicitly accounts for the inner correlations of the HLA complex itself, both within loci (arising from the fact that multinomial allelic frequencies must sum to unity) and between loci (arising from a failure of independent assortment). The analysis yields a single test criterion for association of the HLA complex, as a whole, with the disease in question: this criterion has an approximately chi square distribution. The overall test may conveniently be partitioned into a variety of interestin

[Genetic control by the HLA region of the immune response to factor VIII in hemophilic patients].

In 10 sibships of haemophilic A patients, every one including 1 haemophilic brother with an antibody to factor VIII and 1 haemophilic brother without antibody, pattern of HLA segregation was analysed. All sibships were HLA haploidentical or different and not HLA identical. This fact suggested a linkage between the major histocompatibility complex and an "Ir" gene to factor VIII.

Hepatitis C virus and ethanol alter antigen presentation in liver cells.

Alcoholic patients have a high incidence of hepatitis C virus (HCV) infection. Alcohol consumption enhances the severity of the HCV disease course and worsens the outcome of chronic hepatitis C. The accumulation of virally infected cells in the liver is related to the HCV-induced inability of the immune system to recognize infected cells and to develop the immune responses. This review covers the effects of HCV proteins and ethanol on major histocompatibility complex (MHC) class I- and class II-restricted antigen presentation. Here, we discuss the liver which functions as an immune privilege o…

Renal tubular epithelial cells as immunoregulatory cells in renal allograft rejection.

Kidney transplantation is the best therapeutic option for patients with end-stage kidney disease. However, the long-term survival of kidney transplants still remains relatively poor even under potent immunosuppression. Thus, it is necessary to further review the pathogenesis of renal allograft failure. Here, we discuss the potential impact of activated resident tubular epithelial cells (TECs) on infiltrating leukocyte responses on renal allograft failure. Immunohistochemical staining or in situ hybridization of renal allograft biopsies shows that activated TECs produce inflammatory cytokines a

Role of IFN-gamma in allograft rejection.

Interferon (IFN)-gamma is a cytokine produced mostly by activated T cells and NK cells that has complex effects on immune and nonimmune cells. IFN-gamma plays important roles in inflammation, usually in synergy with other cytokines, such as IL-1beta and TNF-alpha. The uniqueness of IFN-gamma lies in its ability to induce major histocompatibility complex (MHC) expression in many tissues, making it particularly relevant to transplantation. The results of graft rejection in the absence of IFN-gamma show that IFN-gamma modulates but is not essential for the allogeneic responses, suppressing genera…
