Degradation of free and sulfur-dioxide-bound acetaldehyde by malolactic lactic acid bacteria in white wine.

AIMS: Acetaldehyde is the major carbonyl compound formed during winemaking and has implications for sensory and colour qualities of wines as well as for the use of the wine preservative SO(2). The current work investigated the degradation of acetaldehyde and SO(2)-bound acetaldehyde by two commercial Oenococcus oeni starters in white wine. METHODS AND RESULTS: Wines were produced by alcoholic fermentation with commercial yeast and adjusted to pH 3.3 and 3.6. While acetaldehyde was degraded rapidly and concurrently with malic acid at both pH values, SO(2)-bound acetaldehyde caused sluggish bac

Energy-linked potassium uptake by mitochondria from wild-type and poky strains of Neurospora crassa.

Mitochondria from Neurospora crassa, like mammalian mitochondria, carry out rapid, energy-linked K+ uptake and H+ release in the presence of valinomycin. The maximal rate of K+ uptake was about 1.0 mumol/mg of mitochondrial protein per min and was seen at valinomycin concentrations in the range of 100 to 200 mug per mg of mitochondrial protein and at K+ concentrations of 4 mM or above. Uptake could be supported either by substrate oxidation or by adenosine 5'-triphosphate (ATP), and was inhibited in the former case by antimycin or cyanide, in the latter case by oligomycin, and in both cases by

Growth and sporulation of Bacillus subtilis mutants blocked in the pyruvate dehydrogenase complex.

Two "ACE" mutants of Bacillus subtilis which require acetate for growth on glucose minimal medium have been isolated. They do not grow with acetoin, 2,3-butanediol, fatty acids, isoleucine, lipoic acid, malic acid, pyruvic acid, succinic acid, thiamine, or valine, but respond somewhat to glutamate or citrate. The mutants lack the activity of the pyruvate dehydrogenase complex; they excrete pyruvate and later acetoin. They grow in nutrient sporulation medium (NSMP) to one-half the normal turbidity and do not sporulate subsequently. When acetate is added to NSMP (at the optimal concentration of

The effect of illumination of the malic acid content and anion/cation balance of mustard leaves (Sinapis alba).

1. Mustard plants have been grown under conditions in which the length of artificial day could be controlled. 2. Leaf samples were analysed for malic acid and citric acid, and for a number of inorganic anions and cations. A simple method is described by which sap was obtained from 0.5g. samples of leaves. 3. In days of 16hr. or more, malic acid was accumulated; the chief cation accumulated was calcium. 4. When the day-length was reduced the malic acid content decreased considerably but the calcium content remained the same. There was little change in the pH value of the sap, the balance of ani

L-Malic acid from Cissus gongylodes induces cannabinoid-mediated antinociception in mice.

ETHNOPHARMACOLOGICAL RELEVANCE: Cissus gongylodes (Baker) Planch is a vine found in South America. Primarily used as food, its leaves and branches are consumed raw, cooked, roasted or flour by indigenous people (Kayapó, Kulina and Apinayé tribes) as muscle relaxant, to treat rheumatism, disinfection, gastritis, diabetes, kidney stones, joint pain and stomach ache. AIM OF THE STUDY: This study provides scientific validation for the traditional use of C. gongylodes and opens new perspectives for L-malic acid as a potential analgesic for acute pain. MATERIALS AND METHODS: The extract was evalua

Physiological and Molecular Responses of Anabaena laxa to Rac-Metalaxyl Fungicide: Exploring Metabolic and Oxidative Changes.

Rac-metalaxyl is a widely used fungicide for managing plant diseases; however, its environmental persistence and potential toxicity to aquatic microorganisms raise significant ecological concerns. Despite its widespread application, there is limited understanding of how cyanobacteria, which play vital ecological roles in aquatic systems, respond to such chemical stressors. This study addresses this gap by investigating the physiological and metabolic responses of the cyanobacterium Anabaena laxa to rac-metalaxyl exposure. Cultures were treated with 100 mg/L and 200 mg/L concentrations of the f

Intra- and extracellular real-time analysis of perfused fibroblasts using an NMR bioreactor: A pilot study.

INTRODUCTION: Metabolomic discrimination of different mitochondrial defects is challenging. We describe an NMR-based bioreactor allowing real-time intra- and extracellular metabolic investigation of perfused fibroblasts. OBJECTIVES: The objective of this study is (I) determining whether metabolic investigations of perfused fibroblasts overall and separated for intra- and extracellular contributions by real-time NMR allows for discrimination of different representative mitochondrial defects in a feasibility study and (II) gaining insight into physiological consequences of mitochondrial dysfunc

Sensitive and Reproducible Mass Spectrometry-Compatible RP-UHPLC Analysis of Tricarboxylic Acid Cycle and Related Metabolites in Biological Fluids: Application to Human Urine.

We describe a method for the analysis of organic acids, including those of the tricarboxylic acid cycle (TCA cycle), by mixed-mode reversed-phase chromatography, on a CSH Phenyl-Hexyl column, to accomplish mixed-mode anion-exchange separations, which results in increased retention for acids without the need for ion-pairing reagents or other mobile phase additives. The developed method exhibited good retention time reproducibility for over 650 injections or more than 5 days of continuous operation. Additionally, it showed excellent resolution of the critical pairs, isocitric acid and citric aci

The role of glucagon in regulating chicken hepatic malic enzyme and histidase messenger ribonucleic acid expression in response to an increase in dietary protein intake.

Increased dietary protein intake rapidly (3 h) decreases hepatic malic enzyme and increases hepatic histidase mRNA expression in broiler chicks. A series of experiments was conducted to determine the role that glucagon or a specific mixture of dietary amino acids might have in regulating the rapid changes in mRNA expression of these enzymes, when dietary protein intake is increased. Three hours after the injection of glucagon (240 microg/kg of BW) into the brachial vein of broiler chicks, hepatic malic enzyme mRNA expression was significantly lower and hepatic histidase mRNA expression was sig

An ultrastructural and ultrahistochemical study of the human placenta in maternal pre-eclampsia.

At the electronoptical level the placental villi of the pre-eclamptic woman are characterized by focal syncytial necrosis, loss and distortion of microvilli, dilatation of syncytial rough endoplasmic reticulum, decreased syncytial pinocytotic activity, a reduced number of syncytial secretory droplets, cytotrophoblastic hyperplasia, degeneration of occasional cytotrophoblastic cells, thickening of the trophoblastic basement membrane and the presence of small fetal capillaries with bulbous endothelial cells. Ultrahistochemical studies show reduced alkaline phosphatase and dehydrogenase activity
