A meta-analysis of false memory in healthy and pathological cognitive aging.

Although there is a consensus about age-related impairments in true memory, the relationship between aging and false memory remains less clear. Both the fuzzy-trace theory and the activation-monitoring theory postulate possible effects of cognitive aging on the processes of encoding, consolidation, and retrieval. Yet, quantitative analyses of cognitive aging, both healthy (younger vs. older adults) and pathological (older adults vs. mild cognitive impairment/Alzheimer's disease), on false memory have not been conducted. We meta-analyzed 150 articles with 414 independent effect sizes and found

Closed-Loop Slow-Wave tACS Improves Sleep-Dependent Long-Term Memory Generalization by Modulating Endogenous Oscillations.

Benefits in long-term memory retention and generalization have been shown to be related to sleep-dependent processes, which correlate with neural oscillations as measured by changes in electric potential. The specificity and causal role of these oscillations, however, are still poorly understood. Here, we investigated the potential for augmenting endogenous slow-wave (SW) oscillations in humans with closed-loop transcranial alternating current stimulation (tACS) with an aim toward enhancing the consolidation of recent experiences into long-term memory. Sixteen (three female) participants were

MeCP2 gates spatial learning-induced alternative splicing events in the mouse hippocampus.

Long-term memory formation is supported by functional and structural changes of neuronal networks, which rely on de novo gene transcription and protein synthesis. The modulation of the neuronal transcriptome in response to learning depends on transcriptional and post-transcriptional mechanisms. DNA methylation writers and readers regulate the activity-dependent genomic program required for memory consolidation. The most abundant DNA methylation reader, the Methyl CpG binding domain protein 2 (MeCP2), has been shown to regulate alternative splicing, but whether it establishes splicing events im…

Noradrenergic β-receptor antagonism in the basolateral amygdala impairs reconsolidation, but not extinction, of alcohol self-administration: Intra-BLA propranolol impairs reconsolidation of alcohol self-administration.

A critical barrier to recovery from alcohol addiction is relapse propensity. Alcohol cues can trigger relapse, and pharmacologically facilitating processes such as extinction, which decreases cue associations, may help prevent relapse. The noradrenergic system mediates extinction learning for alcohol; however, the neural locus of this effect is unknown. This study sought to determine whether the basolateral amygdala (BLA), a region critical for fear extinction, also mediates extinction of alcohol seeking. Hooded Wistar rats (N = 12-15 per experiment) were implanted with bilateral cannula targe

Effect of inhibition of reactivated alcohol-associated memories with propranolol on alcohol craving.

BACKGROUND: Alcohol craving and relapse occur after the reactivation of alcohol reward memory. Previous studies suggest that drug-associated memory undergoes reconsolidation once retrieved by drug-associated stimuli. This study hypothesized that propranolol administration during memory reconsolidation induced by conditioned stimulus (CS) would significantly attenuate alcohol craving. METHODS: A total of 40 patients with alcohol dependence who met the diagnostic criteria for alcohol dependence in DSMV were enrolled. The patients were randomized located into the memory retrievalpropranolol grou

Enhancement of motor consolidation by post-training transcranial direct current stimulation in older people.

Consolidation, by which performance increments after a training intervention are secured and sometimes generated, is reduced in elderly humans. The present study addressed the question whether transcranial direct current stimulation (tDCS) applied after motor training improves consolidation of explicit motor sequence learning in healthy older humans. In the first experiment, anodal or cathodal tDCS to the left primary motor cortex, anodal tDCS to premotor cortex, or sham tDCS was applied immediately after completion of a finger sequence learning task. Performance was retested at 8 and 22 hours

Strong and Weak Episodic Memories Are Shaped by Multiple Cycles of NREM Spindles and REM Alpha Bursts.

Prior research has built up the notion that non-rapid eye movement sleep (NREM) and rapid eye movement sleep (REM) are separately important for specific types of memory. However, growing evidence suggests that these sleep stages work in conjunction to jointly shape memory. The sequential hypothesis proposes that the ordered sequence of NREM followed by REM is critical for memory consolidation. Additionally, NREM spindles and REM alpha bursts have been independently implicated in episodic memory, yet it is unknown how these oscillatory events may shape episodic memory together. We hypothesised …

How Psychedelics Modulate Multiple Memory Mechanisms in Posttraumatic Stress Disorder.

Posttraumatic stress disorder (PTSD) is a psychiatric disorder with defining abnormalities in memory, and psychedelics may be promising cand … srupt maladaptive beliefs. More research is needed on the acute effects of psychedelics on episodic memory consolidation, retrieval, and reconsolidation and post-acute effects of psychedelics on all phases of episodic memory. We conclude by discussing how targeting multiple memory mechanisms could improve upon the current psychedelic therapy paradigm for PTSD, thereby …

Opposite actions of dopamine on aversive and appetitive memories in the crab.

The understanding of how the reinforcement is represented in the central nervous system during memory formation is a current issue in neurob … ppetitive memories in the crab Chasmagnathus. We found that DA-receptor antagonists impair aversive memory consolidation, in agreement with previous reports in insects, while administration of DA facilitates memory formation after a weak training protocol. In contrast, DA treatment during appetitive training was found to impair formation of long-term appetitive memory …

Previous studies have suggested that the basolateral amygdala (BLA) and the ventral hippocampus (VH) are critical sites for predator-related … ry exposure on the formation of contextual fear responses and that GR-induced signaling facilitates memory consolidation within the BLA and VH. The results are relevant in understanding how life-threatening situations such as a predator encounter impact fear memory storage and open exciting perspectives to investigate GR-induced proteins as targets to deciphering and …

The hippocampal FTO-BDNF-TrkB pathway is required for novel object recognition memory reconsolidation in mice.

Memory reconsolidation refers to the process by which the consolidated memory was restored after reactivation (RA). Memory trace becomes labile after reactivation and inhibition of memory reconsolidation may disrupt or update the original memory trace, which provided a new strategy for the treatment of several psychiatric diseases, such as drug addiction and post-traumatic stress disorder. Fat mass and obesity-associated gene (FTO) is a novel demethylase of N6-methyladenosine (m6A) and it has been intensively involved in learning and memory. However, the role of FTO in memory reconsolidation h

Remote episodic memory deficits in patients with unilateral temporal lobe epilepsy and excisions.

The nature of remote memory impairment in patients with medial temporal lobe damage is the subject of some debate. While some investigators have found that retrograde amnesia in such patients is temporally graded, with relative sparing of remote memories (Squire and Alvarez, 1995), others contend that impairment is of very long duration and that remote memories are not necessarily spared (Sanders and Warrington, 1971; Nadel and Moscovitch, 1997). In this study, remote memory was assessed in 25 patients with unilateral temporal lobe epilepsy and 22 non-neurologically impaired controls using the

Cortical Serotonin Type 2A Receptor Activation Shields Episodic-like Memories from Retroactive Interference in Rodents.

The acquisition of temporally proximate information can impair the brain's ability to consolidate earlier experiences, resulting in retroactive interference (RI). Recognition-based behavioral paradigms are well-suited for investigating RI in rodents, particularly those involving sequential learning episodes. The medial prefrontal cortex (mPFC) integrates multimodal information relevant to the regulation of memory interference and is strongly modulated by the serotonergic system. Serotonin 2A receptors (5-HT2AR), which are densely expressed in the mPFC, have been shown to influence the retrieva

Sleep Spindle Characteristics and Relationship with Memory Ability in Patients with Obstructive Sleep Apnea-Hypopnea Syndrome.

Obstructive sleep apnea syndrome (OSAS) causes intermittent hypoxia and sleep disruption in the brain, resulting in cognitive dysfunction, but its pathogenesis is unclear. The sleep spindle wave is a transient neural event involved in sleep memory consolidation and synaptic plasticity. This study aimed to investigate the characteristics of sleep spindle activity and its relationship with memory ability in patients with OSAS. A total of 119 patients, who were divided into the OSAS group (n = 59, AHI ≥ 15) and control group (n = 60, AHI < 15) according to the Apnea Hypopnea Index (AHI), were enr…

Behavioral, neurological, and psychiatric frailty of autobiographical memory.

Autobiographical-episodic memory is considered to be the most complex of the five long-term memory systems. It is autonoetic, which means, self-reflective, relies on emotional colorization, and needs the features of place and time; it allows mental time traveling. Compared to the other four long-term memory systems-procedural memory, priming, perceptual, and semantic memory-it develops the latest in phylogeny and ontogeny, and is the most vulnerable of the five systems, being easily impaired by brain damage and psychiatric disorders. Furthermore, it is characterized by its fragility and pronen

Acquisition and consolidation of implicit motor learning with physical and mental practice across multiple days of anodal tDCS.

BACKGROUND: Acquisition and consolidation of a new motor skill occurs gradually over long time span. Motor imagery (MI) and brain stimulation have been showed as beneficial approaches that boost motor learning, but little is known about the extent of their combined effects. OBJECTIVE: Here, we aimed to investigate, for the first time, whether delivering multiple sessions of transcranial direct current stimulation (tDCS) over primary motor cortex during physical and MI practice might improve implicit motor sequence learning in a young population. METHODS: Participants practiced a serial react

Renin-angiotensin system: a novel target for brain health.

Emerging evidence highlights the brain renin-angiotensin system (RAS) as a key regulator of reward, memory, and stress. While these discoveries established the brain RAS as a promising therapeutic target for interventions in neurological and neuropsychiatric disorders, translational progress is hampered by the lack of an integrative mechanistic framework. Here, we consolidate accumulating evidence on the molecular and system-level roles of the brain RAS in reward, memory, and stress pathways, and its dual regulatory architecture. Pharmacological RAS modulation regulates domain-specific signali

Retrieval of autobiographical memory in Alzheimer's disease: relation to volumes of medial temporal lobe and other structures.

The representation of autobiographical memory is distributed over a network of brain structures, with the medial temporal lobe (MTL) at its epicenter. Some believe that, over time, all memories become independent of their MTL component ("consolidation theories"). Others have suggested that this is true only of semantic memory, while episodic aspects of autobiographical memories are dependent on the MTL for as long as they exist, such as multiple trace theory (MTT). In the present study, the volumes of 28 brain regions, including the MTL, and their relation to autobiographical memory were inves
