Diastematomyelia and spina bifida can be caused by the intraspinal grafting of somites in early avian embryos.

OBJECTIVE: In this experimental study, an embryological model was created to reproduce diastematomyelia and spina bifida and to investigate new aspects of the origin of spinal cord malformations. METHODS: A somite was implanted from a donor quail embryo into the neural tube of a 2-day-old chick embryo. The somite was chosen because the septum that characteristically separates the two hemicords consists exclusively of mesodermal derivatives. RESULTS: After 2 days of reincubation, diastematomyelia, spina bifida, or a normal embryo without a graft was observed. If the graft persisted in the neu…

Functional histology of the human thymus.

The thymus develops from a paired epithelial anlage in the neck. This review considers how ectoderm (vesicula cervicalis) and endoderm (third pharyngeal pouch) contribute to the epithelial stroma of the thymus. Stromal elements of mesodermal origin are capillaries, septae and perivascular spaces and single invading cells. These elements separate the thymus into pseudolobuli. The thymus epithelial space and the perivascular spaces are always separated from each other by a closed, flat epithelial cell layer, with a basal lamina which contributes to the blood-thymus barrier. From the 9th gestatio…

Protocol to generate microglia-containing cerebral organoids to model HIV neuroinflammation.

Cerebral organoids (COs) are valuable for studying neurodegenerative diseases and pathogens, but their limited microglia pose challenges. Here, we present a protocol to generate organoids with physiologically relevant microglia (CO-iMs). We describe steps for differentiating induced pluripotent stem cells (iPSCs) into mesoderm, seeding embryoid bodies (EBs), and harvesting hematopoietic progenitor cells (HPCs). We detail procedures for co-culturing HPCs and iPSCs to generate CO-iMs, followed by their infection and treatment. The model provides a platform to study viral infections and neuroinfl…

Associated congenital genitourinary and intestinal anomalies: A case report.

The female urogenital tract originates in the intermediate mesoderm, with the Müllerian ducts being critical for uterine, vaginal, and tubal formation. This report presents a 44-year-old patient with uterus didelphys, urethral and bladder duplication, complete longitudinal vaginal septum and intestinal malrotation, suggesting a complex, multi-stage embryonic disorder. The patient, asymptomatic from a gynecological viewpoint, presented with refractory urinary incontinence. Magnetic resonance imaging was crucial in characterizing the anatomy. The rarity and complexity of such cases underscore th…

Characterization of an induced pluripotent stem cell line (NCHi013-A) from a 5-year-old male with pulmonary atresia with intact ventricular septum and a biventricular repair.

Pulmonary atresia with intact ventricular septum (PA/IVS) is a rare congenital heart defect that causes a significant decrease of blood outflow from the heart and is fatal if left untreated. iPSC line NCHi013-A was produced from peripheral blood mononuclear cells from a male child with PA/IVS using Sendai virus reprogramming. NCHi013-A displayed normal stem cell morphology, expressed markers for pluripotency, and presented ability to differentiate into cells of endoderm, ectoderm, and mesoderm lineages. The iPSC line also maintained normal karyotype, was validated for cell identity, and tested…

Two induced pluripotent stem cell (iPSC) lines derived from patients affected by Waardenburg syndrome type 1 retain potential to activate neural crest markers.

Waardenburg syndrome type 1 (WS1), a rare genetic disease characterized by pigmentation defects and mild craniofacial anomalies often associated with congenital deafness is caused by heterozygous mutations in the PAX3 gene (2q36.1). We have generated two induced pluripotent stem cell lines (PCli029-A and PCli031-A) from two patients from the same family both carrying the same heterozygous deletion in PAX3 exon 1 (c.-70_85 + 366del). These cells are pluripotent as they can differentiate into ectoderm, mesoderm and endoderm. They also can activate the early neural crest marker SNAI2. These cells…

A rare case of craniofacial morphology with absent face and neck structures, with its review.

The development of the head and the face requires an intimate interaction between two mesenchymal populations, a paraxial mesoderm and neural crest cells for the morphogenesis of the musculoskeletal components of the calvaria, the face and the branchial regions. The disruptions in these interactions can cause foetal fatalities or congenital craniofacial anomalies. We are describing a rarest case with a craniofacial malformation, who was born with complete absence of the facial skeleton and the neck structures, a set of well developed ears in their normal locations and eyelids at the junction b…

Placental mesenchymal dysplasia associated with hepatic and pulmonary hamartoma.

This report describes a 31-week stillborn female infant with placental mesenchymal dysplasia (PMD) in association with hepatic mesenchymal hamartoma (HMH) and pulmonary hamartoma. Placental mesenchymal dysplasia was initially misdiagnosed as a partial mole. However, histologically, no trophoblastic proliferation or inclusions were observed. Differential diagnosis of the hepatic mass with similar tumors is discussed. To our knowledge, this is the first case of lung hamartoma reported in a fetus and the first case related to PMD and HMH. A common anomalous development of the mesoderm, a reparati…

Congenital systematized basaloid follicular hamartoma with microphthalmia and hemimegalencephaly.

The lines of Blaschko are a cutaneous pattern of mosaicism present in a variety of skin disorders. Developmental abnormalities affecting other tissues derived from the embryonic ectoderm and mesoderm are sometimes associated. Here, we describe a 5-year-old boy with basaloid follicular hamartoma affecting the left side of the body in linear multiple bands, following Blaschko lines. Lesions were predominantly hypopigmented macules and streaks, but among these, we could observe brownish atrophic patches and brown papules. Furthermore, ipsilateral hemimegaloencephaly and microphthalmia were presen…

Derivation of the mammalian skull vault.

This review describes the evolutionary history of the mammalian skull vault as a basis for understanding its complex structure. Current information on the developmental tissue origins of the skull vault bones (mesoderm and neural crest) is assessed for mammals and other tetrapods. This information is discussed in the context of evolutionary changes in the proportions of the skull vault bones at the sarcopterygian-tetrapod transition. The dual tissue origin of the skull vault is considered in relation to the molecular mechanisms underlying osteogenic cell proliferation and differentiation in th…

Embryonic stem cell lines derived from human blastocysts.

Human blastocyst-derived, pluripotent cell lines are described that have normal karyotypes, express high levels of telomerase activity, and express cell surface markers that characterize primate embryonic stem cells but do not characterize other early lineages. After undifferentiated proliferation in vitro for 4 to 5 months, these cells still maintained the developmental potential to form trophoblast and derivatives of all three embryonic germ layers, including gut epithelium (endoderm); cartilage, bone, smooth muscle, and striated muscle (mesoderm); and neural epithelium, embryonic ganglia, a…

[Cantrell's syndrome].

Congenital diaphragmatic hernia is associated with a high mortality, especially when combined with other defects. The unusual repeated combination of the same congenital abnormalities by a number of patients and the fact that during embryonic life these organs develop from the mesoderm implies the existence of a specific syndrome. This article summarises the available clinical data and reports on three cases with Cantrell's syndrome, showing different pattern of congenital defects such as: 1) supraumbilical abdominal wall defects; 2) a defect of the lower sternum; 3) a deficiency of the anteri…

Construction and Expression Analysis of the bin 3xOllas Tool Line.

Precise detection of endogenous protein expression is essential for understanding gene function in vivo . We generated a Drosophila knock-in line inserting a 3×Ollas tag at the C-terminus of the endogenous biniou ( bin ) locus, enabling specific visualization of the Bin transcription factor. The bin 3xOllas allele faithfully recapitulates native expression in the visceral mesoderm and does not disrupt Bin function. Integrating single-cell RNA-sequencing data, we further identified Bin expression in adult reproductive tissues, including ovarian escort cells and testis hub and cyst cells. This a…

Derivation of an iPSC line using the Sendai virus from a newborn boy with Alagille Syndrome.

We developed a well-characterized induced pluripotent stem cell (iPSC) line. This line was generated by reprogramming peripheral blood mononuclear cells from a male Alagille Syndrome baby using the Sendai viral method, which maintains genomic stability and pluripotent capacity. Markers of undifferentiated iPSCs were validated via immunocytochemistry. Trilineage potential was confirmed through immunofluorescent detection of lineage-specific markers (endoderm, mesoderm, ectoderm). Karyotypic analysis verified a normal diploid complement (46, XY). Additional assessments demonstrated Sendai virus …

Characterization of an induced pluripotent stem cell line (NCHi024-A) from a male infant with bicuspid aortic valve.

Bicuspid aortic valve is one of the most common congenital heart defects which have only two leaflets instead of three. NCHi024-A is an induced pluripotent stem cell (iPSC) line derived from peripheral blood mononuclear cells (PBMCs) of a male infant with a bicuspid aortic valve. The line exhibits normal iPSC morphology, normal karyotype, and donor-matched identity. Markers of undifferentiated iPSC and differentiation into ectoderm, mesoderm, and endoderm were validated through immunofluorescence staining. NCHi024-A was tested negative for transgenes and mycoplasma contamination. This iPSC lin…

Sacral myolipoma with involuntary contraction causing tethered cord syndrome: illustrative case.

BACKGROUND: Spinal lipomas sometimes involve various ectopic tissues originating from the ectoderm, mesoderm, and endoderm in the process of morphological development. OBSERVATIONS: A 29-year-old male patient with myolipoma of the conus medullaris at the S2 and S3 levels was described. The unusual finding, involuntary muscle contraction, was presented in an operative video and a literature review. In the present case, sacral myolipoma with involuntary contraction caused tethered cord syndrome in adulthood, and untethering surgery resolved continuous buttock and leg pain. LESSONS: This rare f…

CRISPR/Cas9 editing to generate a heterozygous COL2A1 p.G1170S human chondrodysplasia iPSC line, MCRIi019-A-2, in a control iPSC line, MCRIi019-A.

To develop an in vitro disease model of a human chondrodysplasia, we used CRISPR/Cas9 gene editing to generate a heterozygous COL2A1 exon 50 c.3508 GGT > TCA (p.G1170S) mutation in a control human iPSC line. Both the control and COL2A1 mutant lines displayed typical iPSC characteristics, including normal cell morphology, expression of pluripotency markers, the ability to differentiate into endoderm, ectoderm and mesoderm lineages and normal karyotype. These chondrodysplasia mutant and isogenic control cell lines can be used to explore disease mechanisms underlying type II collagenopathies and …

Mesothelium and Malignant Mesothelioma.

The mesothelium is an epithelial structure derived from the embryonic mesoderm. It plays an important role in the development of a number of different organs, including the heart, lungs, and intestines. In this publication, we discuss aspects of the development of the mesothelium, where mesothelial structures can be found, and review molecular and cellular characteristics associated with the mesothelium. Furthermore, we discuss the involvement of the mesothelium in a number of disease conditions, in particular in the pathogenesis of mesotheliomas with an emphasis on malignant pleural mesotheli…

Dermoid cyst of the parotid gland: report of a rare entity with literature review.

Dermoid cysts (DCs)are benign lesions and histologically composed of tissues originating from ectoderm and mesoderm, but not endoderm. Approximately 7 % of all DCs are seen in head and neck area. However, parotid gland is an extremely rare localization in which DCs develop, and only 17 cases have been reported in the literature to date. Correct preoperative diagnosis is difficult to be established due to the rarity and ambiguous radiological findings. We report a case of a 21-year old man. All previous reports reviewed and the pathogenesis as well as the histopathologic and radiologic features…
