Fe/N-doped hollow carbon nanozymes for synergistic chemo-photothermal-catalytic therapy via tumor microenvironment remodeling.

The limited penetration and efficacy of chemotherapeutic agents, constrained by the distinctive pathophysiological conditions of the tumor microenvironment (TME), remain a major therapeutic obstacle. In recent years, there has been growing interest in modulating the TME to overcome these barriers, which provides a promising way for enhancing cancer treatment. To address this challenge, a multifunctional nanoparticle system (DOX/FeNHC-PEG) was developed for oxidative stress-augmented triple-modal therapy. The system was constructed by doping Fe3+ ions and N atoms into hollow carbon nanostructur

Structure of the NAD+ Bound Erythrose-4-Phosphate Dehydrogenase (E4PDH) Reveals the Stabilizing Effect of Polyethylene Glycol on the Quaternary Structure.

Erythrose-4-phosphate dehydrogenase (E4PDH, EC 1.2.1.72) from Acinetobacter baumannii (AbE4PDH) is an essential multifunctional enzyme. E4PDH catalyzes the first step of the deoxyxylulose-5-phosphate (DXP) dependent Vitamin B6 biosynthetic pathway. It utilizes nicotinamide adenine diphosphate (NAD+) as a co-factor while exhibiting dual catalytic activity wherein it converts (i) D-Erythrose-4-phosphate to 4-phosphoerythronate and (ii) Glyceraldehyde-3-phosphate to Glyceraldehyde-1,3-bisphosphate. An alternate function of AbE4PDH is the capture of human transferrin (Tf) and lactoferrin (Lf) for

Physiological response and molecular regulatory mechanism reveal a positive role of nitric oxide and hydrogen sulfide applications in salt tolerance of Cyclocarya paliurus.

As a multifunctional tree species, Cyclocarya paliurus leaves are rich in bioactive substances with precious healthy values. To meet the huge requirement of C. paliurus leaf production, sites with some environmental stresses would be potential land for developing its plantations due to the limitation of land resources in China. Nitric oxide (NO) and hydrogen sulfide (H2S) are common gas messengers used to alleviate abiotic stress damage, whereas the mechanism of these messengers in regulating salt resistance of C. paliurus still remains unclear. We performed a comprehensive study to reveal the

Antioxidant and angiotensin-converting enzyme (ACE) inhibitory activity of thymosin alpha-1 (Thα1) peptide.

In this research, the antioxidant property of thymosin alpha-1 (Thα1) peptide was investigated through various antioxidant methods. Thα1 showed 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity (IC50 = 20 µM) and its 2,2-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) scavenging reached 45.33% at 80 µM (IC50 = 85 µM). In addition, hydroxyl and superoxide radical scavenging of Thα1 peptide exhibited a concentration-depended manner. The IC50 values of hydroxyl and superoxide radical scavenging were estimated to be 82 µM and 20 µM, respectively. The effect of Thα1 on eli

Antioxidative therapy in an ex vivo human cartilage trauma-model: attenuation of trauma-induced cell loss and ECM-destructive enzymes by N-acetyl cysteine.

OBJECTIVE: Mechanical trauma of articular cartilage results in cell loss and cytokine-driven inflammatory response. Subsequent accumulation of reactive oxygen (ROS) and nitrogen (RNS) species enhances the enzymatic degradation of the extracellular matrix (ECM). This study aims on the therapeutic potential of N-acetyl cysteine (NAC) in a human ex vivo cartilage trauma-model, focusing on cell- and chondroprotective features. DESIGN: Human full-thickness cartilage explants were subjected to a defined impact trauma (0.59 J) and treated with NAC. Efficiency of NAC administration was evaluated by f

Maturation of peroxisomes in differentiating human hepatoblastoma cells (HepG2): possible involvement of the peroxisome proliferator-activated receptor alpha (PPAR alpha).

We have studied the alterations of peroxisomes in the human hepatoblastoma cell line HepG2, induced to differentiate by long-term cultivation (20 days without passaging) using morphological and biochemical techniques as well as mRNA analysis. Ultrastructural studies revealed alterations in shape and size of peroxisomes, with significant increases in mean diameter and formation of small clusters exhibiting heterogeneous staining for catalase after 20 days in culture. These alterations of peroxisomes correspond to the changes described during the maturation process from prenatal to adult human h

MTHFD1L-associated branched-chain amino acid degradation transcriptional signature represents a conserved metabolic program in Epstein-Barr virus-associated malignancies.

Epstein-Barr virus (EBV)-associated malignancies encompass multiple tumor types with distinct tissue origins, yet the extent to which they share conserved metabolic programs remains poorly understood. Here, we performed an integrated cross-cancer transcriptomic analysis of nasopharyngeal carcinoma, EBV-associated gastric cancer, and EBV-associated plasmablastic lymphoma to identify conserved metabolic programs, followed by independent cohort validation and single-cell transcriptomic characterization. We identified 103 consensus differentially expressed genes and 72 conserved gene-metabolic pat

FAN1 exo- not endo-nuclease pausing on disease-associated slipped-DNA repeats: A mechanism of repeat instability.

Ongoing inchworm-like CAG and CGG repeat expansions in brains, arising by aberrant processing of slipped DNAs, may drive Huntington's disease, fragile X syndrome, and autism. FAN1 nuclease modifies hyper-expansion rates by unknown means. We show that FAN1, through iterative cycles, binds, dimerizes, and cleaves slipped DNAs, yielding striking exo-nuclease pauses along slip-outs: 5'-C↓A↓GC↓A↓G-3' and 5'-C↓T↓G↓C↓T↓G-3'. CAG excision is slower than CTG and requires intra-strand A·A and T·T mismatches. Fully paired hairpins arrested excision, whereas disease-delaying CAA interruptions further slow

SMN and symmetric arginine dimethylation of RNA polymerase II C-terminal domain control termination.

The carboxy-terminal domain (CTD) of the RNA polymerase II (RNAP II) subunit POLR2A is a platform for modifications specifying the recruitment of factors that regulate transcription, mRNA processing, and chromatin remodelling. Here we show that a CTD arginine residue (R1810 in human) that is conserved across vertebrates is symmetrically dimethylated (me2s). This R1810me2s modification requires protein arginine methyltransferase 5 (PRMT5) and recruits the Tudor domain of the survival of motor neuron (SMN, also known as GEMIN1) protein, which is mutated in spinal muscular atrophy. SMN interacts

Ataxia with oculomotor apraxia type 2: a clinical and genetic study of 19 patients.

Ataxia with oculo-motor apraxia type 2 (AOA2) is a recently described autosomal recessive cerebellar ataxia (ARCA) caused by mutations in the senataxin gene (SETX). We analysed the phenotypic spectrum of 19 AOA2 patients with mutations in SETX, which seems to be the third most frequent form of ARCA in Algeria after Freidreich ataxia and Ataxia with vitamin E deficiency. In AOA2 patients, the mean age at onset for all families was in the second decade. Cerebellar ataxia was progressive, slowly leading to disability which was aggravated by axonal polyneuropathy present in almost all the patients

Bioinspired Zinc Macrocycles Confined in UiO-67: A Heterogeneous Synzyme for Sustainable CO2 Valorization.

Nature relies on multifunctional microenvironments to combine substrate recognition, activation, and catalysis with remarkable efficiency. Inspired by this principle, we report heterogeneous synzymes generated by the supramolecular immobilization of pseudopeptidic zinc macrocyclic complexes onto UiO-67 for CO2 activation and valorization. The zinc chiral pseudopeptidic macrocycles can be selectively prepared through solvent-directed macrocyclization, exploiting the conformational preorganization of the precursors to create enzyme-inspired catalytic motifs. Organic Nanozyme-Embedded Biotherapeutic Thermosensitive Hydrogel for Extracellular GSH Depletion and Postoperative Tumor Suppression.

Postoperative tumor recurrence remains a major challenge in solid tumor treatment, largely attributed to an immunosuppressive tumor microenvironment and the enrichment of extracellular glutathione (GSH) in the tumor bed, which supports tumor cell survival and proliferation. To address these issues, we designed a multifunctional hydrogel (named SEH) loaded with a novel cyanine nanozyme and Escherichia coli (E. coli). The cyanine nanozyme integrates efficient photothermal therapy (PTT), photodynamic therapy (PDT), and peroxidase-like enzyme activity. SEH is a temperature-sensitive agarose hydrog
