Primary visual cortex: awareness and blindsight.

The primary visual cortex (V1) is the principal telencephalic recipient of visual input in humans and monkeys. It is unique among cortical areas in that its destruction results in chronic blindness. However, certain patients with V1 damage, though lacking visual awareness, exhibit visually guided behavior: blindsight. This phenomenon, together with evidence from electrophysiological, neuroimaging, and psychophysical experiments, has led to speculation that V1 activity has a special or direct role in generating conscious perception. To explore this issue, this article reviews experiments that h

Brain systems for baroreflex suppression during stress in humans.

The arterial baroreflex is a key mechanism for the homeostatic control of blood pressure (BP). In animals and humans, psychological stressors suppress the capacity of the arterial baroreflex to control short-term fluctuations in BP, reflected by reduced baroreflex sensitivity (BRS). While animal studies have characterized the brain systems that link stressor processing to BRS suppression, comparable human studies are lacking. Here, we measured beat-to-beat BP and heart rate (HR) in 97 adults who performed a multisource interference task that evoked changes in spontaneous BRS, which were quanti

Objective assessment of cortical excitability in migraine with and without aura.

Recent progress in the genetics of migraine has refocused attention on cortical dysfunction as an important component of the pathophysiology of this disorder. In previous work, we have demonstrated functional changes in the visual cortex of migraine patients, using an objective transcranial magnetic stimulation technique, termed magnetic suppression of perceptual accuracy (MSPA). This study aimed to replicate previous findings in migraine with aura (MA) and to use the technique to examine migraine without aura (MoA). Eight MA patients, 14 MoA patients and 13 migraine-free controls participated

The neuropsychology of obsessive compulsive disorder: the importance of failures in cognitive and behavioural inhibition as candidate endophenotypic markers.

Obsessive compulsive disorder (OCD) is a highly debilitating neuropsychiatric condition with estimated lifetime prevalence of 2-3%, more than twice that of schizophrenia. However, in contrast to other neuropsychiatric conditions of a comparable or lesser prevalence, relatively little is understood about the aetiology, neural substrates and cognitive profile of OCD. Despite strong evidence for OCD being familial, with risk to first-degree relatives much greater than for the background population, its genetic underpinnings have not yet been adequately delineated. Although cognitive dysfunction i

Ethanol and neurotransmitter interactions--from molecular to integrative effects.

There is extensive evidence that ethanol interacts with a variety of neurotransmitters. Considerable research indicates that the major actions of ethanol involve enhancement of the effects of gamma-aminobutyric acid (GABA) at GABAA receptors and blockade of the NMDA subtype of excitatory amino acid (EAA) receptor. Ethanol increases GABAA receptor-mediated inhibition, but this does not occur in all brain regions, all cell types in the same region, nor at all GABAA receptor sites on the same neuron, nor across species in the same brain region. The molecular basis for the selectivity of the actio

Spinal modulation of the induction of central sensitization.

Peripheral tissue injury results in a change in the excitability of spinal dorsal horn neurons, central sensitization, and the behavioral correlate, hyperalgesia. It is proposed here that a dynamic balance exists between excitatory and inhibitory synaptic input to the spinal dorsal horn that functions to prevent central sensitization following brief, mild, noxious stimulation. Following more severe stimulation and injury, there is a loss of these inhibitory mechanisms that allow central sensitization to proceed. Single-unit recordings were made from L4-L5 deep dorsal horn neurons (wide dynamic

Impaired "natural reciprocal inhibition" in patients with spasticity due to incomplete spinal cord injury.

Experiments were performed to compare the ability of normal subjects and patients with spinal spasticity to suppress antagonist H reflexes during isometric ankle contractions. Soleus H reflex suppression was examined during tonic pretibial muscle contractions in which the torque levels were constant and during dynamic pretibial muscle contractions in which the torque followed a predetermined ramp. As well, subjects were instructed to alternately contract ankle plantarflexors and dorsiflexors at various frequencies to examine patterns of EMG activity during rhythmically alternating isometric co

Interbulbar reciprocal inhibition in frog olfaction.

Frogs were submitted in acute experiments to electrical stimulation of both ipsi- and contralateral homologous olfactory nerve bundles subserving the dorsal olfactory mucosae. Mitral layer responses on one side were recorded. When both stimulations were given at different onset times and equal intensity, the ipsilateral mitral layer response fell to a sharp minimum of 42% when the contralateral stimulation was given 25 ms before the ipsilateral one. When both stimuli were adjusted for maximal temporal inhibition and the intensity of the contralateral stimulation was varied, the amplitude of th

Roles of intracellular cAMP and protein kinase A in the actions of dopamine and neurotensin on midbrain dopamine neurons.

The role of intracellular cAMP and protein kinase A in dopamine-induced inhibition of dopamine neurons and the attenuation of this inhibition by neurotensin were studied in rat midbrain slices. Spontaneous activity of dopamine cells was recorded extracellularly from both the ventral tegmental area and the substantia nigra. Perfusion of the slices with 8-bromo-cAMP and forskolin significantly attenuated dopamine-induced inhibition, but neither blocked the inhibition completely. Neither SQ22536, an inhibitor of adenylate cyclase, nor H8, an inhibitor of protein kinase A, mimicked the inhibitory

[Complex frequency-selective properties of the fibers of the acoustic nerve in the pigeon].

Frequency-selectivity properties of 198 single auditory fibres in the pigeon were investigated by means of an automatic method. Spontaneous activity in 56 of them innervating hair cells with oscillatory membrane potential changes could be inhibited by single-frequency stimuli (one-tone inhibition). Inhibition areas lay above (45%), below (7%) or on both sides (48%) of their characteristic frequencies (CFs). Input-output functions at inhibitory frequencies were nonmonotonic, while they were always monotonic at best frequencies near CF. One-tone inhibition was not observed in the remaining 142 f

Excitatory and inhibitory effects of dopamine on synaptic transmission in the rat olfactory cortex slice.

A study has been undertaken of the effects of dopamine on excitatory transmission at the lateral olfactory tract (LOT)-superficial pyramidal cell synapse of the rat olfactory cortex slice by measuring the effects of bath-applied dopamine on the amplitudes and latencies of the surface field potentials evoked on submaximal LOT stimulation in a total of 32 preparations. In 7 (22%) slices, dopamine had no detectable effects on transmission. In the remaining preparations, dopamine (1-250 microM) depressed transmission in a concentration-dependent manner. This action was unaffected by nadolol (10 mi

Neostriatal evoked inhibition and effects of dopamine on globus pallidal neurons in rat slice preparations.

Spontaneous unit discharges were recorded extracellularly from globus pallidal (GP) neurons in rat slice preparations. The firing rates of GP neurons ranged from 2.0 to 24.0 spikes/s and their firing patterns were predominantly of two types: regular and irregular. Stimulation of the neostriatum evoked two distinct types of inhibition which were dependent on GP neuronal firing patterns, a brief inhibition (about 75 ms) followed by resetting rhythmic neuronal activities and a relatively long-term inhibition (about 100 ms). These inhibitions evoked by neostriatal stimulation were attenuated or co

An electrophysiological study of the accessory olfactory bulb in the rabbit--I. Analysis of electrically evoked potential fields.

Following electrical stimulation of the vomeronasal nerves, the primary olfactory nerves, the lateral olfactory tract and the corticomedial amygdala, we have made a study of evoked potentials in the rabbit accessory olfactory bulb. Vomeronasal nerve stimulation evoked a complex field potential consisting of a compound action potential followed by 4 negative waves (N1, N2, N3, N4). In contrast to the field potential elicited in the main olfactory bulb following primary olfactory nerve stimulation, there was either no evoked wave or only a weak positive component of the field in the accessory bu

[Change in recurrent inhibition when standing in normal subjects and spastics (author's transl)].

In normal subjects and patients with diseases of the upper motor neurone, recurrent inhibition directed to soleus motoneurones was studied in three different postures: sitting (control situation), standing when leaning backward on a wall, standing independently without the support of a wall. In normal subjects, this recurrent inhibition was not modified when standing with the support of a wall, while it was enhanced when standing without such a support.

Making the rich richer: Frontoparietal tDCS enhances transfer effects of a single-session distractor inhibition training on working memory in high capacity individuals but reduces them in low capacity individuals.

Working memory (WM) performance depends on the ability to extract relevant while inhibiting irrelevant information from entering the WM storage. This distractor inhibition ability can be trained and is known to induce transfer effects on WM performance. Here we asked whether transfer on WM can be boosted by transcranial direct current stimulation (tDCS) during a single-session distractor inhibition training. As WM performance is ascribed to the frontoparietal network, in which prefrontal areas are associated with inhibiting distractors and posterior parietal areas with storing information, we

Dorsal striatum D1-expressing neurons are involved with sensorimotor gating on prepulse inhibition test.

Prepulse inhibition (PPI) is a behavioral test in which the startle reflex response to a high-intensity stimulus (pulse) is inhibited by the prior presentation of a weak stimulus (prepulse). The classic neural circuitry that mediates startle response is localized in the brainstem; however, recent studies point to the contribution of structures involved in higher cognitive functions in regulating the sensorimotor gating, particularly forebrain regions innervated by dopaminergic nuclei. The aim of the present study was to verify the role of dorsal striatum (DS) and dopaminergic transmitting medi

The Differential Developmental Neurotoxicity of Valproic Acid on Anterior and Posterior Neural Induction of Human Pluripotent Stem Cells.

Valproic acid (VPA), widely used as an antiepileptic drug, exhibits developmental neurotoxicity when exposure occurs during early or late pregnancy, resulting in various conditions ranging from neural tube defects to autism spectrum disorders. However, toxicity during the very early stages of neural development has not been addressed. Therefore, we investigated the effects of VPA in a model where human pluripotent stem cells differentiate into anterior or posterior neural tissues. Exposure to VPA during the induction of neural stem cells induced different developmental toxic effects in a dose-
