Blast phase of chronic myeloid leukemia with concurrent BCR::ABL1 and SET::NUP214: A report of two cases.

Chronic myeloid leukemia (CML) is a myeloproliferative neoplasm defined by the presence of t(9;22)(q34;q11.2)/BCR::ABL1. Additional chromosomal abnormalities play an important role in the progression to CML. However, the additional fusion gene was rarely reported such as CBFB::MYH11. In this report, we described two cases of the co-occurrence of BCR::ABL1 and SET::NUP214 in CML-BP for the first time, which is associated with poor outcomes during tyrosine kinase inhibitor (TKI) treatment. Meanwhile, we retrospectively analyzed SET::NUP214 fusion transcript of the two cases at initial diagnosis

Assessment of health related quality of life in polish patients with primary biliary cirrhosis.

BACKGROUND: Most patients with primary biliary cirrhosis (PBC) have impaired health related quality of life (HRQoL), as assessed by PBC-specific HRQoL (PBC-40) and generic (SF-36) questionnaires. Data on the applicability of PBC-27, a shorter version of PBC-40, have been limited. AIMS: To assess HRQoL in Polish PBC patients, applying PBC-40, PBC-27 and SF-36 and to associate clinical or laboratory parameters with HRQoL factors. METHODS: A total of 205 PBC patients (188 females) were analyzed using PBC-40, PBC-27 and SF-36; 85 disease-free demographically matched (in terms of age, gender, eth

Sulfonylurea receptor 1 deficiency alleviates acute lung injury by activating PARP1 in epithelial cells.

Acute lung injury (ALI) is a critical disease with high mortality, yet its mechanisms remain incompletely elucidated. In this study, we investigated sulfonylurea receptor 1 (SUR1), the regulatory subunit of ATP-sensitive potassium (KATP) channels, in the progression of ALI. We observed elevated SUR1 protein levels in the bronchoalveolar lavage fluid (BALF) of patients with sepsis-associated ALI, as well as in BALF and lung tissues of ALI mouse models. Genetic deletion of SUR1, but not Kir6.2 (the pore-forming subunit of KATP channels), significantly alleviated ALI induced by cecal ligation and

FAST-CRISPR: Fusogenic Association and Secured Transfection of CRISPR/Cas9 Ribonucleoproteins Using Lipid-Silica Hybrid Nanoparticles for Therapeutic Genome Editing.

Clinical translation of CRISPR/Cas9 therapeutics is challenged by inefficient cytosolic delivery and toxicity issues associated with viral vectors and nanoparticle-based carriers. To overcome these concerns, herein we report a lipid-silica hybrid nanoparticle platform for fusogenic association and secured transfection of CRISPR/Cas9 (FAST-CRISPR), designed for rapid cytosolic delivery of CRISPR/Cas9 ribonucleoproteins, followed by efficient gene editing. Through direct fusion with the plasma membrane and bypassing conventional endocytic barriers, FAST-CRISPR nanoparticles displayed superior in

Mitochondrial protein COXFA4L3 (C15ORF48) confers resistance to DNA-damaging anticancer agents by repressing mitochondrial DNA damage responses.

DNA-damaging anticancer agents selectively kill highly proliferative cancer cells by inducing DNA damage, such as DNA double- or single-strand breaks, and by inhibiting DNA replication and transcription. These agents have been used for cancer chemotherapy for a long time; however, acquired resistance remains a serious problem that limits their effectiveness. Although mitochondria have their own DNA, which is distinct from nuclear DNA, the involvement of mitochondria and mitochondrial DNA (mtDNA) in resistance to DNA-damaging anticancer agents remains largely unknown. In this study, we found th

Close Contacts Unlocked: Nanopore-Stabilized Microvilli Bypass T Cell Receptor-Ligand-Dependent T Cell Activation.

The biophysical microenvironment critically shapes T cell activation, yet how nanoscale geometry regulates signaling remains poorly understood. Here, we demonstrate that microvilli insertion into nanopores robustly activates primary human T cells in the absence of TCR ligands, in a pore-size-dependent manner. Nanopores of ∼240 nm in diameter elicit strong ERK phosphorylation, Ca2+ influx, and NFAT nuclear translocation, reaching levels comparable to biochemical stimulation using antibodies against the TCR complex and CD28. Although TCR knockdown attenuates responses, residual CD69 expression u

Genetic haploinsufficiency of POM121 exacerbates ischemia reperfusion injury induced acute kidney injury by promoting p53/p21-mediated cellular senescence.

Acute kidney injury (AKI) is a severe clinical syndrome with limited therapeutic options. Here, we identified a previously unrecognized role of the nuclear pore complex protein POM121 in AKI. Analysis of the public transcriptomic dataset GSE153625, together with qPCR, Western blot, and immunohistochemical validation, demonstrated that POM121 expression was markedly increased in ischemia-reperfusion injury (IRI)-induced AKI. Due to embryonic lethality of homozygous POM121 knockout mice, heterozygous POM121⁺/⁻ mice were used for functional studies. POM121 haploinsufficiency significantly aggrava…

Complexes of vertebrate TMC1/2 and CIB2/3 proteins form hair-cell mechanotransduction cation channels.

Calcium and integrin-binding protein 2 (CIB2) and CIB3 bind to transmembrane channel-like 1 (TMC1) and TMC2, the pore-forming subunits of the inner-ear mechano-electrical transduction (MET) apparatus. These interactions have been proposed to be functionally relevant across mechanosensory organs and vertebrate species. Here we show that both CIB2 and CIB3 can form heteromeric complexes with TMC1 and TMC2 and are integral for MET function in mouse cochlea and vestibular end organs as well as in zebrafish inner ear and lateral line. Our AlphaFold 2 models suggest that vertebrate CIB proteins can

Role of cytoplasmic dynein and kinesins in adenovirus transport.

Following receptor-mediated uptake into endocytic vesicles and subsequent escape, adenovirus particles are transported along microtubules. The microtubule motor proteins dynein and one or more kinesins are involved in this behavior. Dynein is implicated in adenovirus transport toward the nucleus. The kinesin Kif5B has now been found to move the adenovirus (AdV) toward microtubule plus ends, though a kinesin role in adenovirus-induced nuclear pore disruption has also been reported. In undifferentiated cells, dynein-mediated transport predominates early in infection, but motility becomes bidirec…

Influence of Scaffold Topography and Culture Duration on Fibroblast Morphology in Tissue Engineering.

Cell morphology is not only integral to its function within the body but also plays a critical role in cellular behavior and fate. In tissue engineering, cell-scaffold interactions play a critical role because scaffold physical and biochemical characteristics, such as pore size, fiber alignment, and surface architecture, directly influence cellular morphology and behavior. These interactions impact key biological processes, including adhesion, proliferation, migration, and differentiation of the cells, ultimately influencing tissue formation and regeneration. This study investigated how scaffo

Homozygous splicing mutation in NUP133 causes Galloway-Mowat syndrome.

OBJECTIVE: Galloway-Mowat syndrome (GAMOS) is a neural and renal disorder, characterized by microcephaly, brain anomalies, and early onset nephrotic syndrome. Biallelic mutations in WDR73 and the 4 subunit genes of the KEOPS complex are reported to cause GAMOS. Furthermore, an identical homozygous NUP107 (nucleoporin 107kDa) mutation was identified in 4 GAMOS-like families, although biallelic NUP107 mutations were originally identified in steroid-resistant nephrotic syndrome. NUP107 and NUP133 (nucleoporin 133kDa) are interacting subunits of the nuclear pore complex in the nuclear envelope dur…

Osteoprotegerin induces osteoclast pyroptosis via NF-κB signaling activation.

As a decoy receptor for RANKL, osteoprotegerin (OPG) suppresses osteoclast differentiation and can induce osteoclast death. Pyroptosis, an inflammatory programmed cell death, critically influences bone resorption, but its regulation remains unclear. This study investigated whether OPG induces pyroptosis in osteoclasts via the NF-κB pathway. Osteoclasts were treated with OPG and assessed for pyroptotic markers, NF-κB activation, morphology, and bone resorption. OPG dose-dependently decreased osteoclast viability, induced membrane rupture (LDH release, P < 0.01), and increased IL-18/IL-1β secret

Ion channel structure and function of the MERS coronavirus E protein.

Coronavirus envelope (E) proteins form drug-targeted ion channels that cause virulence to infected cells. The Middle East respiratory syndrome (MERS) virus has high mortality rates, but its E structure and function are unknown. We report the single-channel conductance and structure of membrane-bound MERS E protein. MERS E conducts K+ ions with a unitary conductance of 113 picosiemens, fivefold larger than the conductance of severe acute respiratory syndrome coronavirus 2 E. Solid-state nuclear magnetic resonance data indicate that the MERS E transmembrane domain forms a five-helix bundle that

TDP-43 Triggers Mitochondrial DNA Release via mPTP to Activate cGAS/STING in ALS.

Cytoplasmic accumulation of TDP-43 is a disease hallmark for many cases of amyotrophic lateral sclerosis (ALS), associated with a neuroinflammatory cytokine profile related to upregulation of nuclear factor κB (NF-κB) and type I interferon (IFN) pathways. Here we show that this inflammation is driven by the cytoplasmic DNA sensor cyclic guanosine monophosphate (GMP)-AMP synthase (cGAS) when TDP-43 invades mitochondria and releases DNA via the permeability transition pore. Pharmacologic inhibition or genetic deletion of cGAS and its downstream signaling partner STING prevents upregulation of NF

Effects of Cardiovascular Risk Factors on Cardiac STAT3.

Nuclear, mitochondrial and cytoplasmic signal transducer and activator of transcription 3 (STAT3) regulates many cellular processes, e.g., the transcription or opening of mitochondrial permeability transition pore, and its activity depends on the phosphorylation of Tyr705 and/or Ser727 sites. In the heterogeneous network of cardiac cells, STAT3 promotes cardiac muscle differentiation, vascular element formation and extracellular matrix homeostasis. Overwhelming evidence suggests that STAT3 is beneficial for the heart, plays a role in the prevention of age-related and postpartum heart failure,

A brief update on potential molecular mechanisms underlying antimicrobial and wound-healing potency of snake venom molecules.

Infectious diseases remain a significant cause of morbidity and mortality worldwide. A wide range of diverse, novel classes of natural antibiotics have been isolated from different snake species in the recent past. Snake venoms contain diverse groups of proteins with potent antibacterial activity against a wide range of human pathogens. Some snake venom molecules are pharmacologically attractive, as they possess promising broad-spectrum antibacterial activities. Furthermore, snake venom proteins (SVPs)/peptides also bind to integrins with high affinity, thereby inhibiting cell adhesion and acc

Effects of increasing intranuclear calcium levels via MCU inhibition on mouse and human PSC-derived cardiomyocyte differentiation and maturation.

Background Cardiovascular diseases remain the leading cause of death worldwide, accounting for approximately 19.41 million deaths annually. Human-induced pluripotent stem cell-derived cardiomyocytes (hiPSC-CMs) hold great promise for disease modeling and therapy. However, variability in differentiation efficiency and maturation state remains a significant challenge. Calcium signaling plays a pivotal role in cardiac differentiation and maturation, with mitochondrial calcium dynamics closely linked to transcriptional activation. Recent evidence suggests that closure of the mitochondrial permeabi
