Tissue reactions after experimental luxation injuries in immature rat teeth.

Immature permanent teeth often develop complications after luxation injuries. The pathology and the degenerative processes of the complicati … m to be damaged immediately after the trauma. By the 3rd day after injury, however, displacement of odontoblasts into the dentinal tubules was observed in many teeth. Moreover, formation of irregular dentin was observed in some teeth after 7 days. These changes of the pulp tended to disappear in the later stages of the experiment.

Dental papilla cells synthesize but do not deposit fibronectin in culture.

The dental papilla cells play a major regulatory role during tooth morphogenesis, and they are the only mesenchymal cells capable of differentiating into odontoblasts secreting dentin. In this paper, we have extended our studies on the behavior of cultured dental papilla cells which have been disaggregated from 17-day mouse embryo teeth. Quite unexpectedly, we observed that these cells, which in vivo are embedded in a fibronectin-rich extracellular matrix, lose all surface-associated fibronectin when cultured as monolayers. Fibronectin was, however, detected intracellularly, and metabolic labe…

On the origin of ganoine: histological and ultrastructural data on the experimental regeneration of the scales of Calamoichthys calabaricus (Osteichthyes, Brachyopterygii, Polypteridae).

In order to understand the process of ganoine formation on the ganoid scales, scale regeneration has been studied to overcome the lack of a growth series of scale ontogeny. Seven stages of ganoid scale regeneration have been defined over a period of five months in the polypterid fish Calamoichthys calabaricus. The study has been carried out using transmission electron microscopic techniques. After wound healing and differentiation of the osseous basal plate, a layer of vascular dentin is deposited at the upper surface of the basal plate owing to the presence there of odontoblasts closely appli…

Orthodentine-osteodentine and osteocytic-anosteocytic transition inferred from Silurian stem chondrichthyan dermal jawbones with ever-white teeth overgrown by brown denticles.

BACKGROUND: The dentigerous jawbones of a 423-million-year-old stem chondrichthyan (ischnacanthid acanthodian) carry a characteristic combin … tubules through orthodentine and osteodentine indicates their production by the same population of odontoblasts. The teeth are added sequentially, while the same generation of denticles is added synchronously, both associated with bone apposition. The overgrowth of denticles at the growing front of a tooth row will terminate its tooth addition, and the timing …

Histochemical localization of putative stem cells in irreversible pulpitis.

OBJECTIVES: Our study aimed to observe the distribution of putative stem cells in irreversible pulpitis and to investigate the expression of specific molecules. SUBJECTS AND METHODS: Extracted third molar teeth were collected and divided into two groups: the normal pulp group and inflamed pulp group. Real-time PCR was applied to detect the expression of several embryonic and dentinogenic genes. The expression of mesenchymal cell markers (STRO-1, CD90, and CD146) and stromal cell-derived factor 1α (SDF-1α)/CXC chemokine receptor 4 (CXCR4) proteins was examined by immunohistochemical analysis.

Biocompatibility of a restorative resin-modified glass ionomer cement applied in very deep cavities prepared in human teeth.

This study evaluated whether a restorative resin-modified glass ionomer cement, Vitremer (VM), would be biocompatible with pulp tissue when used as a liner in very deep cavities prepared in young human permanent teeth. Two dental cements in current use as liner materials, Vitrebond (VB) and Dycal (DY), were compared to VM. Class V cavities were prepared in 36 sound premolars that were scheduled for extraction, and the cavity floor was lined with the restorative cement (VM) or a liner/base control cement (VB or DY). For VM specimens, the cavity floor was pretreated with a primer (polyacrylic ac

Histological and immunohistochemical analyses of the chronology of healing process after immediate tooth replantation in incisor rat teeth.

Dental tissues have special characteristics, and its regenerative capacity is noteworthy. However, understanding the circumstances that lead to regeneration is challenging. In this study, the chronology of the healing process after immediate replantation of rat incisor teeth was examined by histological and immunohistochemical analyses within a 60-day period. Thirty-six male Wistar rats had their maxillary right incisors extracted and replanted after 15 min in saline storage. The rats were sacrificed immediately 3, 7, 15, 28, and 60 days after replantation. The histological analysis showed rup

Ablation of systemic phosphate-regulating gene fibroblast growth factor 23 (Fgf23) compromises the dentoalveolar complex.

Fibroblast growth factor-23 (FGF23) is a hormone that modulates circulating phosphate (P(i)) levels by controlling P(i) reabsorption from the kidneys. When FGF23 levels are deficient, as in tumoral calcinosis patients, hyperphosphatemia ensues. We show here in a murine model that Fgf23 ablation disrupted morphology and protein expression within the dentoalveolar complex. Ectopic matrix formation in pulp chambers, odontoblast layer disruption, narrowing of periodontal ligament space, and alteration of cementum structure were observed in histological and electron microscopy sections. Because ser

Endoplasmic reticulum chaperone protein GRP-78 mediates endocytosis of dentin matrix protein 1.

Dentin matrix protein 1 (DMP1), a phosphorylated protein present in the mineral phase of both vertebrates and invertebrates, is a key regulatory protein during biogenic formation of mineral deposits. Previously we showed that DMP1 is localized in the nuclear compartment of preosteoblasts and preodontoblasts. In the nucleus DMP1 might play an important role in the regulation of genes that control osteoblast or odontoblast differentiation. Here, we show that cellular uptake of DMP1 occurs through endocytosis. Interestingly, this process is initiated by DMP1 binding to the glucose-regulated prote…

Temporal and spatial localization of the dentin matrix proteins during dentin biomineralization.

Formation of bone and dentin are classical examples of matrix-mediated mineralization. The mineral phase is essentially the same in these two tissues and primarily consists of a carbonated hydroxyapatite, but the difference lies in the crystal size and shape. There are three components that are necessary for proper mineralization, namely the proper synthesis and secretion of the non-collagenous proteins (NCPs), self-assembly of the collagenous matrix, and delivery of calcium and phosphate ions to the extracellular matrix. Three major NCPs present in the dentin matrix are dentin matrix protein

Heterochronic quantitative microevolution: dental divergence in aboriginal Americans.

Studies of tooth crown morphology alone have proven valuable in defining human population differentiation. We test the hypothesis that quantitative comparisons of more complex whole tooth structure may prove informative in understanding human diversity. Three disparate populations in Native American genetic history were compared: Kodiak Island Western Eskimos, Peruvian Inca Amerindians, and Southeast Asians. Enamel depth (an increasing gradient extended from Southeast Asians to the Inca) and root parameters were the most discriminating variables. The observed microevolution appears to result f

Development, growth and eruption of permanent incisor teeth in Romney sheep.

The development of permanent incisor teeth of sheep was followed from the age of six weeks to 116 weeks. The teeth were observed through the … m, and the cell rests of Malassez appeared to continue to develop from the outer enamel epithelium. Odontoblasts, which arose from the pulp connective tissue, were initially cuboidal but became fusiform as the tooth matured. At the time of eruption, amelogenesis was almost complete and cementum covered the majority of the tooth surface. The cell rests and mela …

Wnt1's Differential Effects on Craniofacial Bone and Tooth Development.

The development of craniofacial bones and teeth relies heavily on the Wnt signaling pathway, yet the specific mechanisms and Wnt variants involved remain under continual investigation. Using publicly available single-cell sequencing data from the mouse incisor, we reveal Wnt1 expression across dental structures and investigate its role using a Col1a1-dependent Wnt1 transgenic mouse model. Inducing Wnt1 early on affects craniofacial bone without disturbing tooth development, but prolonged embryonic induction leads to postnatal mortality with osteopetrosis-like bone overgrowth and malformed teet

Chapter 3: Pathological Features of Coronal Caries.

Pathology is the science of how a tissue changes during the process of the disease. The pathology is of important knowledge for understanding subsequent treatment concepts of a disease. In the cariology field, pathological features of caries are often presented using tooth sections, whereby the sequence and spread can be monitored. It is optimal to describe such changes using thin undecalcified tooth sections as an overview can be given of both enamel demineralization and pulp-dentine reactions. Also, an optimal understanding is achieved if the clinical status of carious lesion activity is kno

Intrauterine exposure to omeprazole increases the risk of teeth morphological anomalies in the offspring of a murine model.

Conditions experienced in early life have long-lasting effects on offspring health. Despite this, little is known about how maternal exposure to drugs during pregnancy affects offspring teeth morphogenesis. In humans, omeprazole is a common drug used to mitigate Gastroesophageal Reflux Disease. Importantly, omeprazole is a non-specific proton-pump inhibitor, which may inhibit the proton pumps expressed in the developing tooth germ. To date, however, the effects of intrauterine life exposure to omeprazole on offspring tooth development remain unknown. In this study, we addressed this gap in a m

Bobby sox homolog regulates tooth root formation through modulation of dentin sialophosphoprotein.

Tooth root development occurs through the interaction of multiple growth factors and transcription factors expressed in Hertwig's epithelial root sheath (HERS) and dental mesenchyme. Previously, we demonstrated that bobby sox homolog (Bbx) regulates odontoblast differentiation of human dental pulp stem cells. Here, we generated Bbx knockout (Bbx-/- ) mice to address the functional role of Bbx in tooth formation. During tooth development, Bbx was expressed in both dental epithelium and mesenchyme. However, molar and incisor morphology in Bbx-/- mice at postnatal Day 0 (P0) exhibited no prominen

Fibrin glue mixed with platelet-rich fibrin as a scaffold seeded with dental bud cells for tooth regeneration.

Odontogenesis is a complex process with a series of epithelial-mesenchymal interactions and odontogenic molecular cascades. In tissue engineering of teeth from stem cells, platelet-rich fibrin (PRF), which is rich in growth factors and cytokines, may improve regeneration. Accordingly, PRF was added into fibrin glue to enrich the microenvironment with growth factors. Unerupted second molar tooth buds were harvested from miniature swine and cultured in vitro for 3 weeks to obtain dental bud cells (DBCs). Whole blood was collected for the preparation of PRF and fibrin glue before surgery. DBCs we
