PTBP1 controls oncogenic transcript processing and maintenance of acute myeloid leukemia.

RNA-binding proteins are critical regulators of gene expression in both normal physiology and cancer. Here we set out to systematically annotate the functions of RNA-associated proteins across multiple cancer types via domain-focused CRISPR screens targeting RNA-modifying enzymes and RNA-binding proteins. We utilized 3182 sgRNAs targeting 527 RNA-enzymatic and binding domains in 341 RNA-associated proteins and identified multiple RNA-binding proteins as dependencies in acute myeloid leukemia (AML), including the RNA splicing factor PTBP1 and the N6-methyladenosine reader RBM15, with a bias tow

BUB1 as a candidate non-oncogene addiction vulnerability in metastatic phaeochromocytoma/paraganglioma.

Phaeochromocytomas and paragangliomas (PPGLs) are rare neuroendocrine tumours with limited treatment options in the metastatic setting. We performed integrative transcriptomic analyses of TCGA and COMETE datasets and identified BUB1, a mitotic checkpoint kinase and prototypical non-oncogene addiction (NOA) factor, as significantly upregulated in metastatic PPGLs. We validated BUB1 overexpression in primary tumour tissues using RT-qPCR and subsequently conducted functional studies in the human phaeochromocytoma-derived hPheo1 cell line. Genetic silencing of BUB1 or its pharmacological inhibitio

Oncogenic driver and therapeutic target: Prolactin signalling axis in retroperitoneal sarcoma.

BACKGROUND: Retroperitoneal sarcoma (RPS) is a type of malignant tumour arising from mesenchymal tissues within the retroperitoneal space. RPSs tend to develop covertly and are often undiscovered when they have already grown significantly and invaded surrounding tissues and organs. These malignancies demonstrate high recurrence rates, present surgical challenges and exhibit limited responsiveness to radiotherapy and chemotherapy. Serum-derived molecules are known to play critical roles in tumourigenesis and tumour progression. However, the serum molecular profile of RPS patients remains unclea

NSD2 degradation remediates the oncogenic cistrome in t(4;14) multiple myeloma.

AbstractThe t(4;14) chromosomal translocation drives overexpression of the histone methyltransferase NSD2 and defines a high-risk segment of patients with multiple myeloma (MM). Herein, we report the discovery of NSD2-ligand-directed degrader (NSD2-LDD), a cereblon-recruiting and PWWP1-mediated LDD that selectively and potently eliminates full-length and PWWP1 domain-containing NSD2 protein isoforms. NSD2-LDD treatment induces the global loss of H3K36me2, leading to promoter-proximal spreading of H3K27me3 and rewiring of cis-regulatory interactions that reverse t(4;14) transcriptional programs

A Deep Dive Into Oncogenic Viruses Understanding Their Role in Cancer and Approaches for Prevention.

Oncogenic viruses, including Epstein-Barr virus (EBV), Human Papillomavirus (HPV), Kaposi's Sarcoma-Associated Herpesvirus (KSHV), Hepatitis B Virus (HBV), Merkel Cell Polyomavirus (MCPyV), Human T-cell Lymphotropic Virus (HTLV-1), and Hepatitis C Virus (HCV), are significant contributors to cancer development, each through distinct mechanisms. EBV is recognised for its association with cancers such as Burkitt lymphoma and nasopharyngeal carcinoma. High-risk types of HPV are primarily connected to cervical cancer. KSHV plays a crucial role in the development of Kaposi's sarcoma, especially amo

Bidirectional Regulation of c-Myc and TRB1 Establishes Oncogenic Reciprocal Regulation.

The transcription factor c-Myc is frequently upregulated in human cancers through multiple mechanisms, including chromosomal translocation and gene amplification. Since the human TRIB1 gene is located on chromosome 8q24 in close proximity to the MYC oncogene, the MYC and TRIB1 genes may be co-amplified in a large percentage of human cancers. We herein demonstrate molecular crosstalk between TRB1 and c-Myc. Bioinformatic analyses reveal a positive correlation between MYC and TRIB1 expression across multiple cancer types. In prostate cancer cells, the knockdown of c-Myc reduces TRB1 expression.

Tirzepatide alters oncogenic signaling pathways in colorectal cancer cells in vitro.

Obesity prevalence is rapidly increasing worldwide, necessitating diverse treatment approaches ranging from pharmacotherapy to surgical interventions. Tirzepatide, a recently approved dual GIP/GLP-1 receptor agonist, has shown therapeutic promise, but its impact on cancer-related pathways remains unclear. This in vitro study investigated the molecular effects of tirzepatide on colorectal cancer SW48 cells by assessing the expression of key regulatory genes, including NF-kB, p53, c-Myc, and CASP8, after treatment with varying tirzepatide concentrations compared to untreated controls. Results de

Disruption of constitutive CXCR4 oligomers impairs oncogenic properties in lymphoid neoplasms.

The chemokine receptor CXCR4 is overexpressed in many cancers and contributes to pathogenesis, disease progression, and resistance to therapies. CXCR4 is known to form oligomers, but the potential functional relevance in malignancies remains elusive. Using a nanobody-based BRET method, we demonstrate that oligomerization of endogenous CXCR4 on lymphoid cancer cell lines correlates with enhanced expression levels. Specific disruption of CXCR4 oligomers reduced basal cell migration and prosurvival signaling via changes in the phosphoproteome, indicating the existence of constitutive CXCR4 oligom

Mapping the initial effects of carcinogen-induced oncogenic transformation in the mouse bladder.

Characterizing the initial stages of oncogenic transformation allows the identification of tumor-promoting processes before the inherent clonal selection of the aggressive clones. Here, we used global proteomics, genetic cell tracing, and immunofluorescence to dynamically map the very early stages of cancer initiation in a mouse model of bladder cancer. We observed a very rapid and incremental proteome dysregulation, with changes in the energy metabolism, proliferation and immune signatures dominating the landscape. The changes in the lipid metabolism were immediate and defined by an increase

The critical role of the proto-oncogene c-Kit in TSC renal cystogenesis.

The epithelium of kidney cysts in mouse Tuberous Sclerosis complex (TSC) models and TSC patients is composed of proliferating A-intercalated cells. The ablation of the Foxi1 gene abolished renal cystogenesis in principal cell-specific Tsc1 knockout (Tsc1-KO) mice. RNAseq studies comparing kidneys of Tsc1-KO vs. wild-type (WT) and Tsc1/Foxi1-double-knockout identified c-Kit, a tyrosine kinase receptor (RTK), as a transcript whose expression significantly increased in Tsc1-KO mice. Overexpression of FOXI1 in kidney M-1 cells significantly increased c-Kit expression levels. Kidney cystogenesis wa

The IL-17 axis in gastric carcinogenesis: From inflammation to oncogenic transformation.

Gastric cancer (GC) remains a leading cause of global cancer mortality, with Helicobacter pylori (Hp)-induced chronic inflammation and immune dysregulation playing a central pathogenic role. This review consolidates current understanding of interleukin-17 (IL-17) in gastric oncogenesis, delineating its molecular mechanisms, Hp-driven inflammatory pathways, and emerging therapeutic implications. Moving beyond prior analyses of isolated IL-17 signaling components, we systematically evaluate its impact on tumor initiation, progression, metastasis, and therapy resistance. Produced primarily by Th1…

Oncogenic Osteomalacia as the Initial Presentation of Pleural Epithelioid Hemangioendothelioma: A Case Report.

Epithelioid hemangioendothelioma (EHE) is a rare form of vascular neoplasm that can manifest with various symptoms or be discovered incidentally in asymptomatic patients. In this report, we describe a case of a 56-year-old male who presented with progressive lower limb weakness over four years. The evaluation revealed severe hypophosphatemia, an inappropriately normal fibroblast growth factor 23 C-terminal (cFGF-23) level, and a 30 x 20 mm hypermetabolic right pleural mass, which was subsequently proven to be EHE. Following tumor resection, the patient experienced complete recovery, with norma

Uncovering the role of FXYD3 as a potential oncogene and early biomarker in pancreatic cancer.

Pancreatic cancer is an aggressive cancer with silent symptoms and high mortality with less than 11% of the 5-year survival rate. Until now, the significance of genes as clinical biomarkers in the early stages of pancreatic cancer has not been fully understood. Hence, this study aims to reveal the significant genes in the early stages of pancreatic cancer using bioinformatic analysis and in vitro experiments, and to serve as clinical biomarkers for early detection. We used Cancer RNA-Seq Nexus database and identified one tumor suppressor gene (NAGK), and five oncogenes (FXYD3, ACTR1A, B3GNT3, …

Immunotherapies targeting the oncogenic fusion gene CLDN18-ARHGAP in gastric cancer.

The CLDN18-ARHGAP fusion gene is an oncogenic driver newly discovered in gastric cancer. It was detected in 9% (8/87) of gastric cancer patients in our center. An immunogenic peptide specifically targeting CLDN18-ARHGAP fusion gene was generated to induce neoantigen-reactive T cells, which was proved to have specific and robust anti-tumor capacity both in in vitro coculture models and in vivo xenograft gastric cancer models. Apart from the immunogenic potential, CLDN18-ARHGAP fusion gene was also found to contribute to immune suppression by inducing a regulatory T (Treg) cell-enriched microenv

The impact of the BCR-ABL oncogene in the pathology and treatment of chronic myeloid leukemia.

Chronic Myeloid Leukemia (CML) is characterized by chromosomal aberrations involving the fusion of the BCR and ABL genes on chromosome 22, resulting from a reciprocal translocation between chromosomes 9 and 22. This fusion gives rise to the oncogenic BCR-ABL, an aberrant tyrosine kinase identified as Abl protein. The Abl protein intricately regulates the cell cycle by phosphorylating protein tyrosine residues through diverse signaling pathways. In CML, the BCR-ABL fusion protein disrupts the first exon of Abl, leading to sustained activation of tyrosine kinase and resistance to deactivation me

The FABD domain is critical for the oncogenicity of BCR/ABL in chronic myeloid leukaemia.

BACKGROUND: Abnormally expressed BCR/ABL protein serves as the basis for the development of chronic myeloid leukaemia (CML). The F-actin binding domain (FABD), which is a crucial region of the BCR/ABL fusion protein, is also located at the carboxyl end of the c-ABL protein and regulates the kinase activity of c-ABL. However, the precise function of this domain in BCR/ABL remains uncertain. METHODS: The FABD-deficient adenovirus vectors Ad-BCR/ABL△FABD, wild-type Ad-BCR/ABL and the control vector Adtrack were constructed, and 32D cells were infected with these adenoviruses separately. Accumulation of Oncogenic Mutations During Progression from Healthy Tissue to Cancer.

Cancers are typically fueled by sequential accumulation of driver mutations in a previously healthy cell. Some of these mutations, such as inactivation of the first copy of a tumor suppressor gene, can be neutral, and some, like those resulting in activation of oncogenes, may provide cells with a selective growth advantage. We study a multi-type branching process that starts with healthy tissue in homeostasis and models accumulation of neutral and advantageous mutations on the way to cancer. We provide results regarding the sizes of premalignant populations and the waiting times to the first c…
