With increasing passage in culture, the human fibroblast cell strain GM11 lost the Mer+ phenotype (the ability to support the growth of adenovirus 5 damaged prior to infection by MNNG). All of 46 embryonic strains prepared either from various organs of 20 fetuses from 6-7 to 13-14 weeks of gestational age or from two hydatidiform moles showed normal repair of MNNG-treated virus. We conclude that human fetal strains are not usually deficient in such repair, and that the behavior of GM11 is atypical.

Cell death appears to be a paradoxical process but one that is necessary for embryonic and fetal morphogenesis, being a necessary stage in the morphogenetic creode of the organ considered, as dynamically and qualitatively modelled by René Thom. Subtle environmental processes result in "cell suicide" by liberation of the intracellular necrosis enzyme. This phenomenon exists just as naturally during separation of the fingers, fusion of the facial buds, and craniofacial suture growth. Dysfunction produces the syndrome of "cell non-death" as seen for example in Apert's syndrome in which a stomodea

We have added another case of duplication of the gallbladder to the 207 collected cases reviewed by Harlaftis et al. It is the first case in which separate drainage into the duodenum is clearly established. Development from a separate diverticulum of the embryonic foregut is proposed by the authors. We agree with other authors that when multiple gallbladders found during rountine intra-abdominal exploration present no symptoms attributable to the biliary tract, no treatment is required. We also agree that any multiple organ found to be pathologic should be removed.

Hydrolysis of retinylpalmitate and retinylacetate was studied in rat blood, liver tissue, intestine, testes, placenta and embryonic liver. The retinylpalmitate hydrolase activity was observed only in walls of small intestine and, less distinctly, - in liver tissue of some animals. Retinylpalmitate was hydrolyzed in rat liver very slowly, that was determined apparently by the metabolic requirements of the organism. The retinylpalmitate hydrolase activity occurred in dog blood after absorption of vitamin A oil solutions. This phenomenon was probably related to an increased secretion of pancreati

Cell transplantation is a treatment option for diabetes, metabolic liver disease in children, and leukemia. Except for the latter indication, solid organ transplantation is one of the available therapies but can be replaced by cell transplantation. However, due to the limited amount of cells that can be transplanted and due to rejection, results of cell transplants are still inferior to solid organ transplantation; there is a general shortage of donor organs, and cell isolation is limited to organs which cannot be transplanted as a whole for anatomic reasons. Therefore, alternatives to islets

Applications of regenerative medicine technology may offer new therapies for patients with injuries, end-stage organ failure, or other clinical problems. Currently, patients suffering from diseased and injured organs can be treated with transplanted organs. However, there is a shortage of donor organs that is worsening yearly as the population ages and new cases of organ failure increase. Scientists in the field of regenerative medicine and tissue engineering are now applying the principles of cell transplantation, material science, and bioengineering to construct biological substitutes that w

The Drosophila gene atonal encodes a basic helix-loop-helix protein similar to those encoded by the proneural genes of the achaete-scute complex (AS-C). The AS-C are required in the Drosophila PNS for the selection of neural precursors of external sense organs. We have isolated mutants of atonal, which reveal that this gene encodes the proneural gene for chordotonal organs and photoreceptors. In atonal mutants, all observable adult chordotonal organs, and almost all embryonic chordotonal organs fail to form; all adult photoreceptors are missing. For both types of sense organ, this defect is al

The association between ear and kidney anomalies has long been recognized. However, little is known about the underlying mechanisms. In the last two decades, embryonic development of the inner ear and kidney has been studied extensively. Here, we describe the developmental pathways shared between both organs with particular emphasis on the genes that regulate signalling cross talk and the specification of progenitor cells and specialised cell types. We relate this to the clinical features of oto-renal syndromes and explore links to developmental mechanisms.

Ectodermal dysplasia is a hereditary disorder associated with abnormal development of embryonic ectodermally-derived organs including teeth, nails, hair and sweat glands. Hypodontia of the primary and permanent dentition is the most com-mon oral finding. Therefore, affected patients need dental prosthetic treatments during their developmental years. This re-port presents two cases of children affected by ectodermal dysplasia with anodontia. Oral rehabilitation was accomplished with removable acrylic prostheses. Treatment had major impacts on self-esteem, masticatory function, speech and facial

The adrenal cortex shares a common embryological heritage with the gonad and kidney: all are derived from components of the urogenital ridge. Adrenocortical specification from this embryonic structure and the ultimate organogenesis of the adrenal gland are directed by the sequential expression of transcriptional regulators, with later development being additionally mediated by endocrine hormones that regulate organ maintenance and tissue differentiation. Recent advancements in the field of human and mouse genetics have accumulated evidence for a host of novel factors involved in these processe

A 55-year old woman underwent cholecystectomy for cholelithiasis. The removed gallbladder showed a serosal encapsulated nodule, measuring 14 x 6 mm, that histologically was an ectopic functioning liver. This nodule had no connection with the main liver. Accessory liver tissue is nearly always an incidental finding in the course of operations and postmortem examinations. It arises in the neighbourhood of the gallbladder from residual liver cells in the caudal part of the embryonic liver; it may also be found in some other organs and structures in close proximity to the liver during fetal life.

A 25 year old female patient with a painful palpable tumour in the right flank was admitted to our hospital presenting signs and symptoms of an acute abdomen. During explorative laparotomy unexpected haemorrhagic infarction of a splenula caused by a volvulus of the vascular pedicle was found. In spite of the documentation of multiple positional abnormalities of visceral organs preoperative diagnostic imaging techniques were unable to point to the diagnosis because of an uncommon rightsideness of the polysplenism. In the case of an indefinite rightsided abdominal mass defects of embryonic morph

Because of the uncertainty concerning the origin of the malformations in the acardiac twin, its structure was studied to see if the malformations were random or with a pattern related to the twinning process. Included were 18 cases of dysmorphic twins in which an autopsy demonstrated a rudimentary or absent heart, and in which some polarity was evident. The organs and long bones with alterations were tabulated according to the embryonal-fetal circulation of blood from and to the placenta. Malformations were more often encountered in the superior limbs and organs; the number of altered organs d

The pineal organ is the only differentiated photoreceptor organ present in embryos and early larvae of the Atlantic halibut (Hippoglossus hippoglossus). We investigated the molecular identity of opsins in the pineal organ, and their expression during different life stages. Using RT-PCR we identified two 681-bp gene sequences, named HPO1 and HPO4, in cDNA from adult pineal and whole embryos. The predicted amino acid sequences showed highest identity to the transmembrane regions of teleostean RH2 green cone opsins (HPO1, 72-91%) and SWS-1 UV cone opsins (HPO4, 71-83%). In situ hybridization reve

PURPOSE OF REVIEW: Currently, patients suffering from diseased and injured organs are treated with transplanted organs or cells. There is, however, a severe shortage of donor tissues and organs that is worsening yearly given the aging population. This paper reviews recent advances that have occurred in regenerative medicine and describes applications of new technologies to treat diseased or damaged organs and tissues.
RECENT FINDINGS: Most current strategies for tissue engineering depend upon a sample of autologous cells from the diseased organ of the patient. Biopsies from patients with exte

Neural rosettes develop from the self-organization of differentiating human pluripotent stem cells. This process mimics the emergence of the embryonic central nervous system primordium, i.e., the neural tube, whose formation is under close investigation as errors during such process result in severe diseases like spina bifida and anencephaly. While neural tube formation is recognized as an example of self-organization, we still do not understand the fundamental mechanisms guiding the process. Here, we discuss the different theoretical frameworks that have been proposed to explain self-organiza

Liver diseases are associated with a marked reduction in the viable mass of hepatocytes. The most severe cases of liver disease (liver failure) are treated by orthotopic liver transplantation. One alternative to whole organ transplantation for patients with hepatic failure (and hereditary liver disease) is hepatocyte transplantation. However, there is a serious limitation to the treatment of liver diseases either by whole organ or hepatocyte transplantation, and that is the shortage of organ donors. Therefore, to overcome the problem of organ shortage, additional sources of hepatocytes must be

BACKGROUND: Hydrocephalus, the pathological expansion of the cerebrospinal fluid (CSF)-filled cerebral ventricles, is a common, deadly disease. In the adult, cardiac and respiratory forces are the main drivers of CSF flow within the brain ventricular system to remove waste and deliver nutrients. In contrast, the mechanics and functions of CSF circulation in the embryonic brain are poorly understood. This is primarily due to the lack of model systems and imaging technology to study these early time points. Here, we studied embryos of the vertebrate Xenopus with optical coherence tomography (OCT

BACKGROUND: Differences in cranial morphology arise due to changes in fundamental cell processes like migration, proliferation, differentiation and cell death driven by genetic programs. Signaling between fibroblast growth factors (FGFs) and their receptors (FGFRs) affect these processes during head development and mutations in FGFRs result in congenital diseases including FGFR-related craniosynostosis syndromes. Current research in model organisms focuses primarily on how these mutations change cell function local to sutures under the hypothesis that prematurely closing cranial sutures contri

Developing populations of connected neurons often share spatial and/or temporal features that anticipate their assembly. A unifying spatiotemporal motif might link sensory, central and motor populations that comprise an entire circuit. In the sensorimotor reflex circuit that stabilizes vertebrate gaze, central and motor partners are paired in time (birthdate) and space (dorsoventral). To determine if birthdate and/or dorsoventral organization could align the entire circuit, we measured the spatial and temporal development of the sensory circuit node: the vestibular ganglion neurons. We discove
