Paraventricular nucleus neuropeptide expression in Huntington's disease patients.

Neuroendocrine, metabolic and autonomic nervous system dysfunctions are prevalent among patients with Huntington's disease (HD) and may underlie symptoms such as depression, weight loss and autonomic failure. Using post-mortem paraffin-embedded tissue, we assessed the integrity of the major neuropeptide populations in the paraventricular nucleus (PVN)-the hypothalamic neuroendocrine and autonomic integration center-in HD patients. The number corticotropin-releasing hormone, cocaine- and amphetamine-regulated transcript, arginine vasopressin and oxytocin immunoreactive (ir) neurons did not diff

A Hypothalamic Oxytocinergic-Sympathetic Axis Couples Anxiety Dysregulation With Impaired Tendon-Bone Repair.

Rotator cuff injury (RCI) is frequently accompanied by psychological comorbidities, particularly anxiety-related symptoms, which predict poor functional recovery, yet the central mechanisms linking affective disturbance to impaired tendon-bone healing remain unclear. Here, by integrating clinical neuroimaging, imaging transcriptomics, and mechanistic animal experiments, we identify hypothalamic paraventricular nucleus (PVN) oxytocinergic (OXT) neurons as a critical central node coordinating emotional regulation and bone-tendon interface (BTI) repair. In patients with RCI, resting-state fMRI (r

Excitotoxicity and compensatory upregulation of GAD67 in fetal rat hippocampus caused by prenatal nicotine exposure are associated with inhibition of the BDNF pathway.

Prenatal nicotine exposure (PNE) can cause hypersensitivity of hypothalamic-pituitary-adrenal (HPA) axis in offspring with intrauterine grow … es the imbalance of signal output in the fetal hippocampus. The negative feedback regulation of the paraventricular hypothalamic nucleus by the hippocampus is unbalanced, eventually causing hypersensitivity of the HPA axis of the offspring.

Central regulation of hypothalamic-pituitary-thyroid axis under physiological and pathophysiological conditions.

TRH is a tripeptide amide that functions as a neurotransmitter but also serves as a neurohormone that has a critical role in the central regulation of the hypothalamic-pituitary-thyroid axis. Hypophysiotropic TRH neurons involved in this neuroendocrine process are located in the hypothalamic paraventricular nucleus and secrete TRH into the pericapillary space of the external zone of the median eminence for conveyance to anterior pituitary thyrotrophs. Under basal conditions, the activity of hypophysiotropic TRH neurons is regulated by the negative feedback effects of thyroid hormone to ensure

Neuropeptide Y localization in telencephalic and diencephalic structures of the ground squirrel brain.

The distribution of neuropeptide Y-immunoreactive (NPY-IR) perikarya, fibers, and terminals was investigated in the brain of two species of hibernatory ground squirrels, Spermophilus tridecemlineatus and S. richardsonii, by means of immunohistochemistry. In the telencephalic and diencephalic structures studied, distinct patterns of NPY-IR were observed which were essentially identical in male and female animals of both species. No differences in amount or distribution of NPY-IR structures were observed between animals which had been in induced hibernation for several months before sacrifice in

Tert-Butylhydroquinone Reversed Age-Dependent Hypothalamic Mitochondrial Dysfunction Linking Maternal Fructose Exposure to Synaptic Vulnerability.

Maternal nutrition shapes offspring brain metabolism and later-life vulnerability. Excessive fructose intake during pregnancy and lactation has been linked to hypothalamic metabolic dysregulation, yet the mitochondrial basis of this programming remains unclear. We examined female Sprague-Dawley offspring from dams fed a normal diet (ND) or high-fructose diet (HFD) during gestation and lactation at 1 and 3 months of age. Maternal HFD produced age-dependent suppression of hypothalamic electron transport chain activity, accompanied by increased oxidative protein modification, together with a shif

Distribution and pharmacological characterization of somatostatin receptor binding sites in the sheep brain.

Somatostatin binding sites have been localized and quantified in the sheep brain using 125I-Tyr0-DTrp8-somatostatin, by quantitative high resolution light microscopic autoradiography. Sections were analyzed by densitometry on radioautographic film, and subsequently on slides coated with photoemulsion. Specific somatostatin binding sites were concentrated in the medial habenula, superior colliculus, dorsal motor nucleus of the vagus nerve, inferior olive, spinal trigeminal nucleus, and cerebellum. In competition experiments, octreotide, a sst2/sst3/sst5 selective agonist only partially displace

The cerebellar-hypothalamic axis: basic circuits and clinical observations.

Experimental studies on a variety of mammals, including primates, have revealed direct and reciprocal connections between the hypothalamus and the cerebellum. Although widespread areas of the hypothalamus project to cerebellum, axons arise primarily from cells in the lateral, posterior, and dorsal hypothalamic areas; the supramammillary, tuberomammillary, and lateral mammillary nuclei; the dorsomedial and ventromedial nuclei; and the periventricular zone. Available evidence suggests that hypothalamocerebellar cortical fibers may terminate in relation to neurons in all layers of the cerebellar

Oxytocin significantly enhances GABAergic transmission in the central amygdala of alcohol-naïve and alcohol-dependent female rats.

Oxytocin is a promising treatment for alcohol use disorder (AUD) and has been shown to decrease alcohol consumption and withdrawal symptoms associated with alcohol dependence in animal models. Using a rat model of alcohol dependence to investigate oxytocin's effects on dependence-induced alcohol drinking and GABAergic transmission in the central amygdala (CeA), we previously reported that oxytocin blocked escalated alcohol drinking and the acute ethanol-induced enhanced GABA transmission in alcohol-dependent rats. However, these studies were performed only in male rats. We also previously repo

The gut vagal sensory pathway drives postprandial sleep via activation of PVH-projecting GABAergic neurons in the NTS.

Feeding status bidirectionally modulates sleep; however, the neural circuitry that integrates the sensing of gastrointestinal (GI) state and sleep remains poorly understood. The afferent fibers of the vagus nerve extensively innervate the GI tract, transmitting postprandial satiety signals to the brain. This study investigates the key role of the upper gut-innervating vagal sensory neurons in modulating sleep-wake states and promoting postprandial sleep, uncovering the underlying circuit mechanisms. Both feeding and activation of stomach/duodenum-innervating vagal sensory neurons reduce wakefu

Patterns of TRPM7 expression in hypothalamic and hippocampal neurons in modeling of nutritional magnesium deficiency.

Patterns of expression of TRPM7, the major cellular magnesium transporters in neurons of the hypothalamic region and hippocampus, were studied immunohistochemically. Multidirectional nature and different levels of the expression of the above antigen were revealed during modeled magnesium deficiency with regard to structural and functional features of neuron organization in the hypothalamic paraventricular and supraoptic nuclei as well as hippocampal field CA1 and CA3. Changes in the structural characteristics of neurons in the studied areas (absolute and relative indicators) and TRPM7 expressi

Contrasting effects of systemic and central sibutramine administration on the intake of a palatable diet in the rat.

Sibutramine hydrochloride monohydrate is the only centrally active weight-modifying agent currently approved by the FDA for long-term use in the treatment of obesity. Systemic sibutramine treatment has been shown to reduce food intake in humans and rodent models in a manner that is consistent with the enhancement of satiety mechanisms. Although it is generally assumed that the hypophagic effects of the drug are mediated by actions within the brain, the locus or loci of these effects remains unclear. These experiments compared the effects of systemic and intracranial injections of sibutramine o

Characterization of the multisynaptic neuronal control of the rat pineal gland using viral transneuronal tracing.

Knowledge of the polysynaptic pathway conveying photic information to the pineal gland is based upon studies employing lesions, knife cuts and classical tracers. In the present investigation we used viral transneuronal tracing to re-examine the organization of this circuitry. This was accomplished by injecting a neurotropic alpha herpesvirus (pseudorabies virus) into the gland and localizing viral antigen in infected neurones at various postinoculation intervals. This approach is based upon the demonstrated ability of this virus to invade axon terminals, replicate in neurones and pass retrogra

Reactivity pattern of vasopressin-containing neurons and its relation to the antipyretic reaction in the pregnant guinea pig.

Changes in the content of vasopressin-immunoreactive material in neurons and their projections were examined in pregnant and nonpregnant guinea pigs as well as in mother and newborn animals. Before sacrifice all animals used in the present study were submitted to a pyrogen test, during which the pregnant animals displayed a reduced fever response to exogenous pyrogen. The unlabeled enzyme-immunoperoxidase method was used in the present study. Light microscopic examination showed that, in comparison to all other groups examined, the pregnant animals exhibited a reduced content of the vasopressi

Oxytocin Receptor Expression and Activation in Parasympathetic Brainstem Cardiac Vagal Neurons.

Autonomic imbalance, particularly reduced activity from brainstem parasympathetic cardiac vagal neurons (CVNs) is a major characteristic of many cardiorespiratory diseases. Therapeutic approaches to selectively increase CVN activity have been limited by lack of identified selective translational targets. Recent work has shown that there is an important excitatory synaptic pathway from oxytocin (OXT) neurons in the paraventricular nucleus of the hypothalamus (PVN) to brainstem CVNs, and that OXT could provide a key selective excitation of CVNs. In clinical studies, intranasal OXT increases para

Chronic high-fat diet induces overeating and impairs synaptic transmission in feeding-related brain regions.

Obesity is linked to overeating, which can exacerbate unhealthy weight gain. However, the mechanisms for mediating such linkages are elusive. In the current study, we hypothesized that synaptic remodeling occurs in feeding-related brain regions of obese mice. To investigate this, we established a high-fat diet (HFD)-induced obese mouse model and observed that these mice consumed excessive calories. The effect of chronic HFD feeding on lipid droplet accumulation in different brain structures was also investigated. We found that lipid droplets accumulated on the ependyma of the third ventricle (

Localization of fibroblast growth factor 23 protein in the rat hypothalamus.

Fibroblast growth factor 23 (FGF23) is an endocrine growth factor and known to play a pivotal role in phosphate homeostasis. Interestingly, several studies point towards a function of FGF23 in the hypothalamus. FGF23 classically activates the FGF receptor 1 in the presence of the co-receptor αKlotho, of both gene expression in the brain was previously established. However, studies on gene and protein expression of FGF23 in the brain are scarce and have been inconsistent. Therefore, our aim was to localise FGF23 gene and protein expression in the rat brain with focus on the hypothalamus. Also,

Functional connections between the suprachiasmatic nucleus and the thyroid gland as revealed by lesioning and viral tracing techniques in the rat.

Frequent blood sampling via intraatrial cannula revealed daily rhythms of TSH and thyroid hormones in both male and female Wistar rats. Thermic ablation of the biological clock, i.e. the suprachiasmatic nucleus (SCN), eliminated the diurnal peak in circulating TSH and thyroid hormones. In addition, SCN lesions produced a clear decrease of 24-h mean T4 concentrations. A more pronounced effect of SCN-lesions on thyroid hormones, as opposed to TSH, suggested routes of SCN control additional to the neuroendocrine hypothalamopituitary-thyroid axis. Retrograde, transneuronal virus tracing was used t
