Parvovirus B19 infection induces apoptosis of erythroid cells in vitro and in vivo.

OBJECTIVE: intrauterine parvovirus B19 infection is related to non-immune hydrops fetalis, the pathogenesis of which is based on the strict tropism of B19 for erythroid precursor cells and the massive destruction of the infected erythroid cells, although the mechanism of beta19-induced cytotoxicity has not been studied in detail. The purpose of this study is to provide empirical evidence that beta19 induces apoptosis of erythroid cells both in vitro and ill vivo. METHODS: we analysed culture cells infected in vitro by B19 and tissues of nine cases of hydrops fetalis caused by B19 intrauterine…

Hydranencephaly and cerebellar hypoplasia in two kittens attributed to intrauterine parvovirus infection.

Six weeks after vaccination with modified live feline parvovirus vaccine, a cat gave birth to five kittens, three of which died soon afterwards. The remaining two kittens (A and B) survived, but at 8 weeks of age were unable to walk and showed abnormal behaviour, with lack of menace and oculovestibular responses, and severe dysmetria. These signs suggested multifocal disease associated with the cerebrum and cerebellum. Magnetic resonance imaging demonstrated severe bilateral (kitten A) or unilateral (kitten B) hydrocephalus or hydranencephaly, combined with cerebellar agenesis (kitten A) or se…

[The study on detection of human parvovirus B19 DNA in spontaneous abortion tissues].

116 spontaneous abortion tissues were detected for human parvovirus B19 (HPV B19) DNA by nested polymerase chain reaction. Of the abortion tissues detected, 66 were paraffin embedded and 50 were fresh abortion tissues, At the same time, 25 abortion tissues from healthy artificial abortion women were collected as control. The results showed that 27.3% (34/116) of 116 spontaneous abortion tissues were positive for HPV B19 DNA, and only 4% (1/25) were positive in the control group, the discrepancy was significant chi 2 = 5.769(P < 0.05). It suggests that there is a relationship between spontaneou…

Immediate and long term outcome of human parvovirus B19 infection in pregnancy.

OBJECTIVE: To estimate more precisely the risk of fetal loss and congenital abnormalities after maternal parvovirus B19 infection, and to assess the long term outcome for surviving infants. DESIGN: Prospective cohort study of pregnant women with confirmed B19 infection with follow up of the surviving infants. The rate of fetal loss in the study cohort was compared with that in pregnant women with varicella. SETTING: Cases reported by laboratories in England and Wales between 1985-1988 and 1992-1995. SAMPLE: Four hundred and twenty-seven pregnant women with B19 infection and 367 surviving in…

Detection of parvovirus B19 infection in first and second trimester fetal loss.

Fetal and placental tissues and maternal sera from a series of 273 cases of first and second trimester fetal loss were collected to detect the frequency of parvovirus B19 infection. In addition, fetal tissues were studied for the presence of congenital anomalies. Serology of maternal sera, histology of fetal tissues and placenta, polymerase chain reaction (PCR), in situ hybridization (ISH), and immunohistochemistry (IHC) were used for the detection of parvovirus B19 infection. Sera were tested for B19-specific immunoglobulin M (IgM) and/or IgG using an enzyme-linked immunosorbent assay techniq…

Detection of parvovirus B19 in macerated fetal tissue using in situ hybridisation.

AIMS: To compare the application of a non-radioactive in situ hybridisation (ISH) technique with an immunocytochemical technique for the detection of human parvovirus B19 in formalin fixed, paraffin wax embedded sections of macerated fetal tissue. METHODS: Archived samples of liver, lung or kidney from 19 human fetuses were investigated for parvovirus B19 using a full length digoxigenin labelled DNA probe of 5.5 kb; bound probe was detected using an anti-digoxigenin (alkaline phosphatase) conjugate and visualised using NBT/BCIP. Immunocytochemical detection of parvovirus B19 was performed usi…

Dot immunoperoxidase assay for detection of parvovirus B19 antigens in serum samples.

We describe a simple and rapid dot immunoperoxidase assay for the direct detection of parvovirus B19 capsid antigens in human sera. The assay was performed with serum specimens dotted onto nylon membranes. VP1 and VP2 B19 antigens, which represent 4 and 96% of the capsid, respectively, were detected with a pool of monoclonal antibodies directed against the two proteins, and the complex was visualized by immunoperoxidase staining. The assay could be performed in about 4 h, and positive results were revealed at the end of the reaction as dark blue spots on the nylon membrane at the site of posit…

Human parvovirus B19 infection and unbalanced translocation in a case of hydrops fetalis.

In a case of hydrops fetalis, serological examination showed a recent maternal human parvovirus B19 infection. Amniocentesis revealed a unique unbalanced translocation between chromosomes 3 and 11 of the fetus. The mother proved to have a balanced reciprocal translocation between chromosomes 3 and 11. A grossly macerated hydropic male fetus was delivered with a flat nose and low implanted deformed ears. Histopathological examination revealed nuclear inclusion bodies in fetal erythroid cells, confirming human parvovirus B19 infection. Parvovirus B19 DNA was demonstrated by in situ hybridization…

Parvovirus B19 as a cause of acquired chronic pure red cell aplasia.

Parvovirus B19 infection causes chronic anaemia in immunodeficient individuals by selective suppression of erythropoiesis. The bone marrow morphology is characteristic of pure red cell aplasia (PRCA). To determine the frequency of B19-induced PRCA we retrospectively analysed a series of 57 PRCA patients. B19 DNA was present in serum of eight patients (14%) and could be extracted from bone marrow aspirate slides from five of these patients. Recent exposure to the virus was confirmed by the presence of anti-B19 IgM in sera from four and by the finding of giant pronormoblasts in marrow aspirates …

Recurrent granulocytic aplasia as clinical presentation of a persistent parvovirus B19 infection.

We report a case of persistent infection with human parvovirus B19 (PVB19), manifesting clinically as recurrent agranulocytosis and in bone marrow biopsy as recurrent pure granulocytic aplasia. Persistence of parvovirus infection was documented by the presence of PVB19 DNA and anti-PVB19-IgM antibodies in the serum for a period of 19 months. Granulocytic aplasia occurred only when anti-PVB19-IgG antibodies were not detectable in the serum and granulopoiesis showed immediate recovery with high dose intravenous immunoglobulin treatment. This case report suggests that the erythroid precursor cell…

Prevalence of antibodies to parvovirus B19 in selected groups of patients and healthy individuals.

The prevalence of antibodies to parvovirus B19 in sera (n = 745) of various groups of patients and healthy individuals was determined by the enzyme immunoassay, using viral particles as antigen. Among healthy individuals, anti-B19 IgG prevalence was highest in nurses (65.4% (17/26)); in medical students it was 34.1% (47/138) and in pregnant females, 24.4% (48/197). 37.0% (44/119) of HIV-negative haemophiliac patients and 91.7% (33/36) of haemophilic patients with HIV infection were anti-B19 IgG-positive. 45.8% (55/120) of dialysis patients and 27.5% (30/109) of patients with asymptomatic HIV i…

[Infection with parvovirus B19: typical course of erythema infectiosum and its complications].

Severe chronic anemia, transitory aplastic crisis (TAC), and lethal hydrops fetalis may be due to an infection with parvovirus B19 i.e. the causative agent of erythema infectiosum. 80% of the patients infected with B19 develop clinical symptoms and typically show the picture of erythema infectiosum--as one of our case reports (daughter) is going to illustrate. Part of the cases, however, take a course without the characteristic slapped-face rash; here we frequently find acute arthritis and arthralgia, as well as fever and rubella-like or purpura-like rashes, predominantly on the smaller joints…

Human parvovirus B19 infections in women of childbearing age and within families.

Human parvovirus B19 (B19) crosses the placenta causing fetal death. We used an indirect capture enzyme immunoassay to measure IgG to B19 in sera of 845 subjects from 2 groups. The first group included 405 women (mean age, 30 years) composed of 85 pediatric nurses and 130 other female hospital employees, 122 women employed caring for preschool children and 68 mothers of preschool children enrolled in day care. Twenty-eight percent of all these women were seropositive. Seropositivity was unrelated to occupational group. Four of 235 women observed between 1983 and 1987 for a mean of 435 days/wom…

Human parvovirus B19-induced transient pancytopenia in a child with hereditary spherocytosis.

We report a child with hereditary spherocytosis who developed human parvovirus B19 (HPV B19)-induced transient pancytopenia. Symptoms of acute-phase illness subsided promptly after infusion of gammaglobulin containing HPV B19 antibody. The HPV B19-containing serum on admission inhibited not only erythroid colony growth but also myeloid and megakaryocyte colony growths. The inhibitory effect was abrogated by convalescent-phase serum or gammaglobulin solution containing HPV B19 antibody. These findings suggest that HPV B19-induced inhibition of haematopoiesis may be pathogenetic for transient bo…

Intrauterine infection with human parvovirus B19: a light and electron microscopy study.

Recent reports have documented human parvovirus B19 infection in pregnancy, which may lead to spontaneous abortion, stillbirth or hydrops fetalis. Transplacental infection in a case of hydrops fetalis is demonstrated by light and electron microscopy observation of virus inclusions in all tissues examined. Parvovirus particles measuring approximately 20 nm were observed within the nuclei of erythropoietic cells. The parvovirus virions were observed most commonly as randomly distributed particles and rarely in crystalline assays. Thus definitive evidence of fetal infection confined to red cell p…

[The effectiveness of vaccinating gilts against parvoviruses in a herd with endemic parvovirus infection].

An inactivated vaccine against swine parvovirosis with a lipoid adjuvant was tested in a herd infested with parvoviruses. The titre of the haemagglutination-inhibition antibodies was studied at the time of the first pregnancy in two groups of gilts included in the herd at the age of 7.5 months - one group vaccinated, the other left untreated. In the vaccinated group the geometrical means of the titres were significantly higher than in the non-vaccinated group throughout the time of study. The difference in the average number of piglets per litter between the two groups was evaluated after part…
