RAS mutation is associated with hyperdiploidy and parental characteristics in pediatric acute lymphoblastic leukemia.

We explored the relationship of RAS gene mutations with epidemiologic and cytogenetic factors in a case series of children with leukemia. Diagnostic bone marrow samples from 191 incident leukemia cases from the Northern California Childhood Leukemia Study were typed for NRAS and KRAS codon 12 and 13 mutations. A total of 38 cases (20%) harbored RAS mutations. Among the 142 B-cell acute lymphoblastic leukemia (ALL) cases, RAS mutations were more common among Hispanic children (P=0.11) or children born to mothers <30 years (P=0.007). Those with hyperdiploidy at diagnosis (>50 chromosomes) had th

Nipple development and pup-induced prolactin release in male rats treated prenatally with the antiandrogen flutamide.

While maternal female rats display increases in circulating prolactin (PRL) concentrations in response to pup exposure, parental male rats fail to show such an increase. One possible explanation for the lack of an acute PRL response in parental male rats is that males do not have nipples and therefore do not receive stimuli from the pups comparable to those experienced by parental female rats. To examine the contribution of nipple presence and possible stimulation, i.e. suckling, in this sexually differentiated endocrine response, male rats were exposed from days 12-15 of gestation to the anti

Follow-up studies on genome damage in children after Chernobyl nuclear power plant accident.

As children are more susceptible to ionizing radiation than adults, each nuclear accident demands special attention and care of this vulnerable population. The Chernobyl nuclear disaster occurred in a region populated with a large number of children, but despite all efforts and expertise of nuclear specialists, it was not possible to avoid casualties. As vast regions of Ukraine, Belarus and Russia were exposed to doses of ionizing radiation, which are known to be related with different diseases, shortly after the accident medical surveillance was launched, which also included analysis of genom

Transgenerational Inheritance Effects of Copper Oxide Nanoparticles (CuONPs) Induced Asthenospermia and Infertility via Gamete H3K9me3 Insufficiency Pathway in Mice.

The widespread use of colloidal copper oxide nanoparticles (CuONPs) poses substantial health risks to humans. CuONPs can penetrate the blood-testis barrier and induce spermatocide, and the understanding of the adverse effects of asthenospermia on spermatogenesis, embryonic development, and transgenerational inheritance is limited. In this study, male mice were orally administered different doses of CuONPs via continuous exposure for one spermatozoon development period (35 days) and then exposed without CuONPs for another 35 days. The CuONPs that accumulated in the testes induced oxidative stre

The effects of prenatal exposure to alcohol and environmental tobacco smoke on risk for ADHD: a large population-based study.

Attention deficit hyperactivity disorder (ADHD) is caused by the interaction of genetic and environmental factors. The objective of this study was to examine the effects of prenatal exposure to alcohol and environmental tobacco smoke (ETS). Among the 30,552 parents who responded to a survey, the answers of 19,940 who replied to questions on prenatal exposure to ETS, alcohol consumption, and completed the DuPaul Rating Scale were analyzed. Results revealed that risk of ADHD significantly increased as a result of exposure to alcohol by 1.55 times (95% CI 1.33-1.82), maternal smoking during pregn

Childhood cancer in the offspring born in 1921-1984 to US radiologic technologists.

We examined the risk of childhood cancer (<20 years) among 105 950 offspring born in 1921-1984 to US radiologic technologist (USRT) cohort members. Parental occupational in utero and preconception ionising radiation (IR) testis or ovary doses were estimated from work history data, badge dose data, and literature doses (the latter doses before 1960). Female and male RTs reported a total of 111 and 34 haematopoietic malignancies and 115 and 34 solid tumours, respectively, in their offspring. Hazard ratios (HRs) and 95% confidence intervals (CIs) were calculated using Cox proportional hazards reg

Parental occupational exposure to extremely low frequency magnetic fields and childhood cancer: a German case-control study.

Extremely low frequency magnetic fields (ELF-MFs) have been classified as possibly carcinogenic to humans by the International Agency for Research on Cancer. The authors investigated, in a population-based case-control study in Germany, if children whose parents were exposed preconceptionally at work to ELF-MFs had an increased risk of developing cancer. Cases aged 0-14 years were ascertained from the German Childhood Cancer Registry. Controls were selected from local resident registration offices. The parental occupational history was recorded in questionnaires and telephone interviews, and p

Adult paclobutrazol exposure impairs zebrafish reproduction: evidence from fertility outcomes and gonadal histology.

Paclobutrazol (PBZ), a triazole-based plant growth retardant, is widely employed in agriculture and horticulture to control plant height and promote root growth. However, its persistence and mobility in the environment raise concerns regarding its bioavailability and toxicity in aquatic ecosystems. In this study, we investigated the reproductive toxicity of PBZ in zebrafish (Danio rerio), a well-established model for aquatic toxicology and vertebrate reproductive research. Adult zebrafish were exposed to PBZ (0, 0.1, 1 ppm) for 2, 4, 6, or 8 weeks. After exposure, breeding was conducted using

Cancer in the offspring of radiation workers: a record linkage study.

OBJECTIVES: To test the "Gardner hypothesis" that childhood leukaemia and non-Hodgkin lymphoma can be caused by fathers' exposure to ionising radiation before the conception of the child, and, more generally, to investigate whether such radiation exposure of either parent is a cause of childhood cancer. DESIGN: Case-control study. SETTING: Great Britain. SUBJECTS: 35,949 children diagnosed as having cancer, together with matched controls. MAIN OUTCOME MEASURES: Parental employment as radiation worker as defined by inclusion in the National Registry for Radiation Workers and being monitored

When health care sickens the health care worker: Commentary on Park et al. (2023).

In their survey, Park and colleagues (2023) reported that many health care and hospital workers perceived that their safety was not a priority during the COVID-19 pandemic, with 33.9% of responders feeling betrayed by institutional leaders. Employees of the health care system reported being viewed more as servants than specialists during the pandemic, and they noted that critical assessment and the expression of one's own opinion were unwelcome (Lederer, 2023). A lack of epidemiologic knowledge was compensated with the inflexible strategy of a hierarchical system. Opting out of testing and vac

Questionnaire assessment of lifetime and recent exposure to environmental tobacco smoke.

In a sample of 149 adult nonsmokers recruited in New Mexico in 1986, the authors assessed the reliability of questionnaire responses on lifetime exposure to tobacco smoke in the home. They also compared urinary cotinine levels with questionnaire reports of environmental tobacco smoke exposure during the previous 24 hours. The agreement of responses obtained on two occasions within six months was high for parental smoking during childhood: 94% for the mother and 93% for the father. For the amounts smoked by the mother and the father during the subject's childhood, the agreement between the two

Congenital heart diseases and parental occupational exposure in a Hungarian case-control study in 1997 to 2002.

The etiology of congenital heart diseases is not fully understood yet, however, endocrine disrupting chemicals may have a causative role in their development. The purpose of our study was to examine the association between congenital heart diseases and periconceptional parental occupational exposure to endocrine disrupting chemicals. In our Hungarian population-based case-control study, we examined 2263 live born cases with any congenital heart disease and 6789 matched controls selected between years 1997 to 2002. Occupational exposure was assessed with a job-exposure matrix developed for endo

Exposure to air pollution is associated with DNA methylation changes in sperm.

Exposure to air pollutants has been associated with adverse health outcomes in adults and children who were prenatally exposed. In addition … elated to neurodevelopment. Together, these data provide compelling support for the hypothesis that paternal exposure to air pollution impacts DNAm in sperm, particularly in regions implicated in neurodevelopment.

Prolonged Exposure to Childhood Adversity and Birth Outcomes in a Bi-Generational Longitudinal Cohort Study.

OBJECTIVES: A higher number of exposures to different adverse childhood experiences (ACEs) has been associated with worse birth outcomes; however, the duration of ACE exposure could also negatively impact pregnancy outcomes. We sought to use a bi-generational longitudinal cohort survey to test if duration of exposure to specific types of childhood adversity exhibited a graded association with adverse birth outcomes. METHODS: The Bureau of Labor Statistics' National Longitudinal Surveys of Youth 1979 cohort was used to create bi-generational cohorts capturing the number and duration of ACE eve

Father knows stress: Interactions among stress, glucocorticoids, and paternal care in mammals.

In female mammals, motherhood and maternal care have bidirectional interactions with stress and glucocorticoid hormones: exposure to stressors and elevated glucocorticoid levels can impair parenting by mothers, while, conversely, pregnancy and lactation blunt mothers' neural, hormonal, and behavioral responses to stressors. In contrast, little is known about the effects of stress and glucocorticoids on paternal care in mammalian fathers or how fatherhood affects the activity of the hypothalamic-pituitary-adrenal (HPA) axis and responses to stressors. We review the available findings on interac

In-office survey of children's hazard exposure in the Chicago area: age-specific exposure information and methodological lessons. Pediatric Practice Research Group.

Anticipatory guidance on injury prevention should reflect the risks children face, yet hazard exposure information is generally unavailable. The objectives of this study were (1) to obtain information on age-specific exposure of Chicago-area children to amusement park rides, sleds, snow discs, bunkbeds, skateboards, fireworks, toboggans, and air guns and (2) to assess methodological issues in gathering exposure information by parental survey in pediatric practices. Questionnaires were received from 679 families, including 1469 children. The proportion of families with at least one exposed chil

Decreased birth weight after prenatal exposure to wildfires on the eastern coast of Korea in 2000.

OBJECTIVES: In April 2000, a series of wildfires occurred simultaneously in five adjacent small cities located on the eastern coast of Korea. These wildfires burned approximately 23,794 hectares of forestland over several days. We investigated the effects of prenatal exposure to the by-products generated by wildfire disasters on birth weight. METHODS: Birth weight data were obtained for 1999-2001 from the birth registration database of the Korean National Statistical Office and matched with the zip code and exposed/unexposed pregnancy week for days of the wildfires. Generalized linear models
