BACKGROUND: The ability to provide safe and effective pharmacotherapy during obstetric complications, such as preterm labor or postpartum hemorrhage, is hampered by the systemic toxicity of therapeutic agents leading to adverse side effects in the mother and fetus. Development of novel strategies to target tocolytic and uterotonic agents specifically to uterine myocytes would improve therapeutic efficacy while minimizing the risk of side effects. KRN568 is a calcimimetic compound which acts on the calcium sensing receptors (CaR) on the parathyroid gland to suppress secretion of PTH. Idiopathic Parkinson's disease may derive from the action of an environmental 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-like compound. Monoamine oxidase (MAO) B converts MPTP to an actual neurotoxin, 1-methyl-4-phenylpyridinium (MPP+) whilst prior administration of an MAO B inhibitor, (-)deprenyl, prevents the conversion.
