Structural analysis of PpSP15 and PsSP9 sand fly salivary proteins designed with a self-cleavable linker as a live vaccine candidate against cutaneous leishmaniasis.

BACKGROUND: Leishmania parasites are deposited in the host through sand fly bites along with sand fly saliva. Therefore, salivary proteins are promising vaccine candidates for controlling leishmaniasis. Herein, two immunogenic salivary proteins, PpSP15 from Phlebotomus papatasi and PsSP9 from Phlebotomus sergenti, were selected as vaccine candidates to be delivered by live Leishmania tarentolae as vector. The stepwise in silico protocol advantaged in this study for multi-protein design in L. tarentolae is then described in detail. METHODS: All possible combinations of two salivary proteins, P…

[Cutaneous leishmaniasis misdiagnosed as epithelial skin cancer].

Cutaneous leishmaniasis, transmitted by sand flies of the genus Phlebotomus, is one of the most important traveler's dermatoses in Germany. In recent years, an increased incidence of the disease has been observed in travelers returning from endemic areas, including the Mediterranean region of Spain and Italy. In these regions, Leishmania infantum belonging to the Leishmania donovani complex is characteristically the most frequently detected pathogen. We herein report two cases of cutaneous leishmaniasis caused by Leishmania infantum with solitary or multilocular lesions, initially misdiagnosed…

Double Trouble: Leishmaniasis and Cytomegalovirus Colitis in an Immunocompromised Host.

Leishmaniasis is endemic in several areas of the world. It is a chronic protozoan disease transmitted through the bite of the Phlebotomus fly. Unusual localizations of visceral leishmaniasis are extremely rare and are described in immunocompromised patients. We present a case of a 73-year-old man, postliving nonrelated kidney transplant who complained of melena, fatigue, and weight loss. Gastroscopy showed a large ulcer at the lesser curvature of the stomach, and biopsies demonstrated severe ulceration with superimposed fungal organisms. Colonoscopy showed a polyp in the transverse colon, and …

Non-compaction cardiomyopathy and visceral leishmaniasis: uncommon combination with therapeutic challenges in a resource limited-setting.

Non-compaction cardiomyopathy (NCCM) or spongy myocardium is a rare type of congenital cardiomyopathy. Visceral leishmaniasis is a protozoal disease caused by Leishmania donovani and transmitted by the bite of female sand-fly species of Phlebotomus argentipes, which is common in tropical areas like Sudan. We report a 6-year-old female, presented with a fever of unknown origin, weight loss, anemia that necessitated multiple blood transfusions and had hepatosplenomegaly. Developed heart failure later on admission the current case narrates an unusual combination of diseases with therapeutic chall…

Isoenzyme characterization of Leishmania from man, dog and sandflies in the Maltese islands.

As a part of a general survey on leishmaniases and sandflies of the Maltese islands, 22 Leishmania stocks were isolated from human visceral (1) and cutaneous (1) cases, dogs (16) and sandflies (4). They were characterized by the analysis of 15 enzymes. The commonest Mediterranean L. infantum zymodeme, MON 1, was found to cause human and canine visceral leishmaniasis; L. infantum MON 78, which has so far been isolated only in Malta, was the agent of human cutaneous leishmaniasis. Both zymodemes were isolated from the same sandfly species, Phlebotomus perniciosus.

Phlebotomine sand fly vector distribution, seasonality, blood meal sources, and infection rates in cutaneous leishmaniasis endemic areas of Northeast Ethiopia.

Cutaneous leishmaniasis (CL) is a significant health problem in Northeast Ethiopia. Leishmania aethiopica was identified in Phlebotomus longipes sand flies 50 years ago. However, the current sand fly fauna, distribution, seasonality, host preference, and Leishmania infection rates remain poorly understood. This study investigated the sand fly vectors distribution, seasonality, blood meal sources, and Leishmania infection rates in the area. Sand fly vectors were collected via CDC light traps in four villages from February to July 2024, and Leishmania DNA and blood meal sources were identified u…

Leishmania sp. detection and blood-feeding behaviour of Sergentomyia minuta collected in the human leishmaniasis focus of southwestern Madrid, Spain (2012-2017).

Phlebotomine sand flies are the only known vectors of Leishmania spp. protozoan which causes leishmaniasis in 98 countries. In Spain, 11 sand fly species are described, but only Phlebotomus perniciosus and Phlebotomus ariasi are proven vectors of the disease. On the other hand, Sergentomyia minuta is one of the most abundant and ubiquitous sand flies in this territory, although scarce information is available about this species. Sand flies from this genus are known for their preference to feed on cold-blooded animals and are traditionally involved in the transmission of reptile Leishmania. How…

First record of autochthonous canine leishmaniasis in Hungary.

Hungary is traditionally regarded as a leishmaniasis-free country, and human or canine cases diagnosed locally have been recorded as imported. However, recent entomological surveys have verified the presence in Hungary of Phlebotomus neglectus and Phlebotomus perfiliewi perfiliewi, which have been incriminated as competent vectors of Leishmania infantum elsewhere in Europe. Following the occurrence in October 2007 of an undisputable clinical case of L. infantum canine leishmaniasis (CanL) in a 4-year-old female pug in a kennel of 20 dogs in Tolna province, an investigation was performed to ass…

Using vehicle-mounted nets for studying activity of Arabian sand flies (Diptera: Psychodidae).

Vehicle-mounted nets were used on 15 nights in northern Oman to collect 2,216 sand flies. The composition of the catch was compared with that of 100 sticky traps, which were used twice a month over a 15-mo period to collect 3,099 sand flies. Sticky trap catches showed no obvious seasonal change in abundance. The vehicle nets collected up to 260 flies per night, with a mean of 148. This compared with the catch of the 100 sticky traps, which had an overall mean of 115 flies per night. The vehicle nets collected seven species of sand flies (four Phlebotomus and three Sergentomyia). Phlebotomus al…

[Detection of Leishmania infantum MON-24 in the dog].

Two forms of cutaneous leishmaniasis are endemic in Algeria. Zoonotic cutaneous leishmaniasis caused by Leishmania major is widespread in the steppe regions of the northern Sahara. Sporadic cutaneous leishmaniasis caused by Leishmania infantum occurs in the north along the coastline. The main causative agent for the sporadic form is Leishmania infantum zymodeme MON-24. It has been isolated from the sandfly vector Phlebotomus perfiliewi. However all attempts to isolate this variant from dogs, i.e., the main reservoir of Leishmania infantum, have failed. The authors report the first successful d…
