MAP2K6 directly phosphorylates BCL2L13 to mediate mitophagy for suppressing tumorigenicity.

The mitogen-activated protein kinase (MAPK) pathway widely regulates development and cancer. However, the subcellular localization and funct … g the interaction between BCL2L13 and LC3B, which, in turn, promoted mitophagy, inhibited oxidative phosphorylation (OXPHOS), and prevented tumor growth. Our studies not only revealed that MKK6-BCL2L13 phosphorylation at the interorganellar site can affect mitochondrial quality in tumorigenicity but also might provide a potential therapeutic strategy for LUAD tre …

Tau Phosphorylation as an Adaptive Physiological Response: Implications for the Therapy of Tauopathies.

Recent evidence demonstrates that tau phosphorylation, traditionally viewed as a hallmark of neurodegeneration, also occurs in completely reversible physiological contexts such as mammalian hibernation and human neonatal development. These findings challenge the classical protein-centric "proteinopathy" model of Alzheimer's disease (AD) and other tauopathies. Instead, we propose that phosphorylated tau (p-tau) functions as an adaptive molecular response to metabolic or neuronal activity shifts, and that tauopathies such as AD represent a failure of broader mechanisms that normally restore tau …

PET Imaging of Phosphorylated RIPK1 Reveals Necroptosis Activation In Vivo.

Necroptosis is a regulated, pro-inflammatory form of programmed cell death implicated in degenerative and inflammatory diseases, with RIPK1 acting as a key upstream regulator. However, the utility of existing RIPK1 PET radiotracers, as well as various RIPK1 inhibitors, remains uncertain, as it is unclear whether they reflect total protein abundance or phosphorylation-dependent activation (pRIPK1), limiting accurate assessment of in vivo signaling. Unlike previously reported RIPK1 PET tracers that primarily reflect total RIPK1 distribution, we investigated whether [18F]RIPA56-3-F ([18F]1) uptak…

Ag-Catalyzed Multicomponent Reaction To Access 9-Phosphorylated Xanthenes.

This study describes an efficient multicomponent reaction of 2-hydroxybenzaldehyde, secondary phosphine oxides, and β-alkynyl ketones, which affords various 9-phosphorylated xanthenes in moderate to good yields under silver catalysis. The method is highly atom- and step-economical, representing a novel synthetic strategy for the construction of multisubstituted 9-phosphorylated xanthenes. Mechanistic studies of key intermediates indicate that the process proceeds via a sequence involving phospha-aldol, alkylation, cyclization, dehydration and cyclization.

PAK4 phosphorylates and stabilizes MYC to promote acute myeloid leukemia.

MYC dysregulation plays a crucial role in acute myeloid leukemia (AML), yet the mechanisms governing its stabilization remain incompletely understood. MYC protein turnover is tightly regulated by post-translational modifications (PTMs), especially phosphorylation-dependent ubiquitination. Our previous study identified phosphorylation at MYC Serine 67 (S67) is critical to sustain its oncogenic activity in T-cell acute lymphoblastic leukemia (T-ALL). Here, we demonstrate that MYC S67 phosphorylation is also present in AML and catalyzed by p21-activated kinase 4 (PAK4). PAK4 directly binds MYC vi…

The oncogenic potential of YAP requires Y357 phosphorylation in cholangiocytes but not in hepatocytes.

PURPOSE: The transcriptional cofactor yes-associated protein (YAP) is activated in primary liver cancers (PLCs). YAP is canonically inhibited through serine 127 (S127) phosphorylation via the Hippo pathway but activated by phosphorylation at tyrosine 357 (Y357) by SRC family kinases. Due to liver plasticity, PLCs can originate from different cell types. However, it is unknown whether YAP activation favors tumorigenesis from specific cells of origin or promotes lineage commitment during tumorigenesis. This study investigates how YAP Y357 phosphorylation influences tumor phenotype and cell of or…

Proton MRI and MRSI as a Superior Methodology to Assess Oxidative Phosphorylation.

RATIONALE AND OBJECTIVES: Accurate determination of energy metabolism is crucial for disease indication and measurement of tissue health in disease conditions. Creatine recovery kinetics following exercise correlates to oxidative phosphorylation (OXPHOS) in tissues and can be used as a marker for understanding energy metabolism. In this study, we demonstrate that creatine chemical exchange saturation transfer (CrCEST), when combined with proton magnetic resonance spectroscopic imaging (1H-MRSI), provides a more accurate assessment of creatine recovery kinetics in a muscle-specific manner when …

GRAMD1B is a regulator of lipid homeostasis, autophagic flux and phosphorylated tau.

Lipid dyshomeostasis and tau pathology are present in frontotemporal lobar degeneration (FTLD) and Alzheimer's disease (AD). However, the relationship between lipid dyshomeostasis and tau pathology remains unclear. We report that GRAM Domain Containing 1B (GRAMD1B), a nonvesicular cholesterol transporter, is increased in excitatory neurons of human neural organoids (HNOs) with the MAPT R406W mutation. Human FTLD, AD cases, and PS19 tau mice also have increased GRAMD1B expression. We show that overexpression of GRAMD1B increases levels of free cholesterol, lipid droplets, and impairs autophagy

Molecular basis for the regulation of human phosphorylase kinase by phosphorylation and Ca2.

Phosphorylase kinase (PhK) regulates the degradation of glycogen by integrating diverse signals, providing energy to the organism. Dysfunctional mutations may directly lead to Glycogen Storage Disease type IX (GSD IX), whereas the abnormal expression of PhK is also associated with tumors. Here, we use cryo-electron microscopy (cryo-EM) to resolve its near-atomic structures in the inactive and active states. These structures reveal the interactions and relative locations of the four subunits (αβγδ) within the PhK complex. Phosphorylated α and β subunits induce PhK to present a more compact stat

Phosphorylated BLM peptide acts as an agonist for DNA damage response.

Upon exposure to ionizing irradiation, the MRE11-RAD50-NBS1 complex potentiates the recruitment of ATM (ataxia-telangiectasia mutated) kinase to the double-strand breaks. We show that the lack of BLM causes a decrease in the autophosphorylation of ATM in mice mammary glands, which have lost one or both copies of BLM. In isogenic human cells, the DNA damage response (DDR) pathway was dampened in the absence of BLM, which negatively affected the recruitment of DDR factors onto the chromatin, thereby indicating a direct role of BLM in augmenting DDR. Mechanistically, this was due to the BLM-depen…

Frk positively regulates innate antiviral immunity by phosphorylating TBK1.

Type I interferons (IFN-I) are crucial for the initial defense against viral infections. TBK1 serves as a key regulator in the production of IFN-I, with its phosphorylation being essential for the regulation of its activity. However, the regulatory mechanisms governing its activation remain incompletely elucidated. In this study, we validated the function of Fyn-related kinase (Frk) in the antiviral innate immune response and identified the direct target molecule of Frk in the IFN-β signaling pathway. Furthermore, we elucidated the mechanism by which Frk phosphorylates TBK1 during infection an…

Basal MET phosphorylation is an indicator of hepatocyte dysregulation in liver disease.

Chronic liver diseases are worldwide on the rise. Due to the rapidly increasing incidence, in particular in Western countries, metabolic dys … in primary hepatocytes from a preclinical model fed with a Western diet (WD) an increased basal MET phosphorylation and a strong downregulation of the PI3K-AKT pathway. Dynamic pathway modeling of hepatocyte growth factor (HGF) signal transduction combined with global proteomics identifies that an elevated basal MET phosphorylation rate is the main driver of alte …

WIPI2b recruitment to phagophores and ATG16L1 binding are regulated by ULK1 phosphorylation.

One of the key events in autophagy is the formation of a double-membrane phagophore, and many regulatory mechanisms underpinning this remain under investigation. WIPI2b is among the first proteins to be recruited to the phagophore and is essential for stimulating autophagy flux by recruiting the ATG12-ATG5-ATG16L1 complex, driving LC3 and GABARAP lipidation. Here, we set out to investigate how WIPI2b function is regulated by phosphorylation. We studied two phosphorylation sites on WIPI2b, S68 and S284. Phosphorylation at these sites plays distinct roles, regulating WIPI2b's association with AT…

Reductive sequestration of Cr (VI) by phosphorylated nanoscale zerovalent iron.

The cracked phosphorylated nanoscale zerovalent iron (p-nZVI) has a strong electron selectivity towards the reductive removal of many heavy metal ions in waters. However, the unintended environmental risk after interactions with impurities or wastewater are not involved. Therefore, in this study, the phosphate group was successfully adsorbed into p-nZVI, and the cracked p-nZVI was successfully prepared with an optimal P/Fe ratio of 0.5%. The dosages of p-nZVI and temperatures were positively correlated with the removal rates. The removal process of Cr(VI) was more suitable by the Langmuir isot
