CASE: The authors report a case of an 6-year-old pediatric patient with a history of acute onset of proptosis of his right eye. He was admitted at hospital 6 months ago for proptosis concomitant with orbital trauma. Computed tomography scan demonstrated a mass involving the right orbit, right maxillary sinus, and zygoma with endocranial extension.
RESULTS: Incisional biopsy of the mass revealed after of histopathologic and immuno-histochemical evaluation a T-cell lymphoblastic lymphoma. Systemic examination and bone marrow aspirate show a acute lymphoblastic leukemia. The patient was treated

We describe 10 cases of B-cell non-Hodgkin lymphoma (NHL) that did not express immunoglobulin kappa or lambda light chains by dual-color flow cytometry. Cases were identified from 298 consecutive cases of B-cell NHL and included follicular center cell lymphoma, diffuse large B-cell lymphoma, small noncleaved cell lymphoma, and small lymphocytic lymphoma. One case did not express any immunoglobulin heavy chain (IgH) as well; however, isolated expression of IgG heavy chain was seen in another case. Immunoglobulin heavy chains were not part of the lymphoma panel in other cases. All 3 cases in whi

Acute Lymphoblastic Leukemia (ALL) is one of the most aggressive hematological malignancies, and its early diagnosis remains challenging due to non-specific clinical symptoms and reliance on invasive procedures such as bone marrow biopsies. To address these limitations, we propose Meta-Conformer-XAI, a novel meta-learned hybrid deep learning framework for non-invasive ALL detection using microscopic peripheral blood smear images. Unlike conventional CNN-Transformer pipelines, our approach integrates three key innovations: (1) a Dual Attention Feature Fusion (DAFF) block that adaptively combine

Outcomes in B-cell acute lymphoblastic leukemia (ALL) have improved over time due to therapeutic advances, and improved disease monitoring and prognostication. The BCR::ABL1 tyrosine kinase inhibitors (TKIs) were the first targeted therapies to significantly impact the therapeutic trajectory and outcomes of Philadelphia chromosome-positive (Ph-positive) ALL. Similarly, antibodies targeting CD19/20/22 and engineered chimeric antigen receptor (CAR) T-cell immunotherapies have drastically changed the therapeutic landscape and outcomes of B-cell ALL. Incorporation of the bispecific T-cell engager

Blinatumomab is the first globally approved bispecific T cell engager targeting the B-cell surface antigen CD19, and numerous clinical trials have demonstrated its significant efficacy in B-acute lymphoblastic leukemia (B-ALL) patients. However, with its widespread application, the problem of drug resistance is gradually becoming apparent. The specific mechanisms of drug resistance mainly include antigen escape, lineage switch, immune checkpoint pathway dysregulation and T cell exhaustion. In this review, the mechanisms of resistance to blinatumomab in B-ALL are systematically described, and r

Acute lymphoblastic leukaemia (ALL) is characterized by uncontrolled proliferation of lymphoid progenitor cells. Advances in genomic and epigenomic profiling have enabled the identification of over 40 molecular subtypes defined by distinct genetic drivers, transcriptional programmes and regulatory alterations. These insights have refined the classification, particularly of B cell precursor ALL (B-ALL) and are increasingly informing risk stratification, therapeutic decision-making and disease monitoring. By contrast, the classification of T cell ALL (T-ALL) has historically relied on immunophen

Recently, the identification of minimal residual disease (MRD), also known as measurable residual disease, has seen significant advances driven by the evolving technological landscape in this field. Initially based on raw morphology, the field has since evolved to incorporate karyotyping, cytogenetics, flow cytometry, and other sensitive methods. This updated review discusses key technical considerations for successful MRD detection. We explore the correlation between the results obtained by flow cytometry and molecular genetic techniques.

BACKGROUND: Voxel-based morphometry (VBM) reveals diverse alterations in gray matter volume in acute lymphoblastic leukemia (ALL) patients after chemotherapy. However, the reported results are inconsistent. Therefore, our objective was to conduct a meta-analysis to synthesize findings from existing VBM studies and identify consistent patterns of altered gray matter volume in ALL patients post-chemotherapy.
MATERIALS AND METHODS: A systematic search was conducted across PubMed, Web of Knowledge, Embase, Google Scholar, and CNKI for VBM studies that compared ALL patients post-chemotherapy with

The DUX4 gene, located within repetitive subtelomeric arrays on chromosomes 4 and 10, plays a critical role in early embryogenesis and has been implicated in several human diseases, including facioscapulohumeral muscular dystrophy (FSHD) and cancer. In B-cell acute lymphoblastic leukemia (B-ALL), DUX4 rearrangements (DUX4-r) define a distinct genomic subtype affecting 5-10% of cases, which is more frequent among older children and teenagers. These rearrangements produce truncated DUX4 proteins with neomorphic transcriptional activity, resulting in aberrant gene expression programs and alternat

Haemophagocytic lymphohistiocytosis (HLH) that occurs concomitantly with leukaemia can be initially missed due to overlapping clinical features. In a series of three cases, Tanabe and colleagues illustrate the need for prompt recognition of HLH and institution of HLH-directed therapy to prevent hyperinflammation-mediated multi-organ damage and death. Commentary on: Tanabe et al. Paediatric acute lymphoblastic leukaemia-associated haemophagocytic lymphohistiocytosis develops during prednisolone prephase. Br J Haematol 2024; 205:2031-2035.

BACKGROUND: Children with acute lymphoblastic leukaemia (ALL) experience multiple symptoms that occur in complicated patterns and negatively affect patient outcomes. To date, no systematic review has been performed on the prevalence of symptoms in children with ALL.
OBJECTIVE: The study aimed to report and analyse the prevalence of symptoms in children with ALL during treatment.
METHODS: A systematic search was conducted in eight databases (PubMed, Ovid Embase, Web of Science, CINAHL, PsycINFO, China WanFang Database, China Science and Technology Journal Database, and China National Knowledg

Francisella philomiragia is a Gram-negative coccobacillus, which is a very rare human opportunistic pathogen causing pneumonia and systemic infection. It is difficult to identify this bacterium through conventional Gram-staining and biochemical methods due to an amorphous Gram stain appearance after 24 h culture and its relatively fastidious and slow growth giving weak and/or delayed reactions in biochemical tests. It is often misidentified as other bacteria including Haemophilus spp., Pseudomonas aeruginosa, or Sphingomonas paucimobilis. False identification may delay the therapy of the patie

The association between individual-level poverty and relapse in children receiving maintenance treatment for acute lymphoblastic leukemia (ALL) remains unclear. In a secondary analysis of COG-AALL03N1, we used data from US Census Bureau to categorize patients living below year-specific federal poverty thresholds, calculated using self-reported annual household income and size of household. Participants with federal poverty thresholds above 120% of their yearly household income were categorized as living in extreme poverty. Hazard of relapse was estimated using multivariable proportional subdis

RNA-sequencing (RNA-seq) efforts in acute lymphoblastic leukaemia (ALL) have identified numerous prognostically significant genomic alterations which can guide diagnostic risk stratification and treatment choices when detected early. However, integrating RNA-seq in a clinical setting requires rapid detection and accurate reporting of clinically relevant alterations. Here we present RaScALL, an implementation of the k-mer based variant detection tool km, capable of identifying more than 100 prognostically significant lesions observed in ALL, including gene fusions, single nucleotide variants an
