Dietary muramidase degrades bacterial peptidoglycan to NOD-activating muramyl dipeptides and reduces duodenal inflammation in broiler chickens.

Muramidases constitute a superfamily of enzymes that hydrolyse peptidoglycan (PGN) from bacterial cell walls. Recently, a fungal muramidase derived from Acremonium alcalophilum has been shown to increase broiler performance when added as a feed additive. However, the underlying mechanisms of action are not yet identified. Here, we investigated the hypothesis that this muramidase can cleave PGN to muramyl dipeptide (MDP), activating nucleotide-binding oligomerisation domain-containing protein 2 (NOD2) receptors in eukaryotic cells, potentially inducing anti-inflammatory host responses. Production and characterization of chicken blood hydrolysate with antihypertensive properties.

Chicken blood has limited utilization despite its high protein content. Production of a blood hydrolysate exhibiting angiotensin I-converting enzyme (ACE)-inhibitory activity would be means of valorizing chicken blood. The optimized conditions used to produce chicken blood corpuscle hydrolysate (BCH) by Alcalase were 51.1°C, 4% enzyme, and pH 9.6 for 6 h, resulting in a 35.8% degree of hydrolysis and 37.7% ACE inhibition at a peptide concentration of 0.2 mg/mL. The permeate of a 1-kDa membrane, BCH-III, showed a 2.5-fold increase in ACE inhibition compared with that of BCH. BCH-III was resista

Enzymatic generation of the NO/HNO-releasing IPA/NO anion at controlled rates in physiological media using β-galactosidase.

We introduce a strategy for generating mixtures of nitric oxide (NO) and nitroxyl (HNO) at tunable rates in physiological media. The approach involves converting a spontaneously HNO/NO-generating ion to a caged (prodrug) form that is essentially stable in neutral media, but that can be activated for HNO/NO release by adding an enzyme capable of efficiently opening the cage to regenerate the ion. By judiciously choosing the enzyme, substrate, and reaction conditions, unwanted scavenging of the HNO and NO by the protein can be minimised and the catalytic efficiency of the enzyme can be maintaine

Structural characterization and stability studies of hemp peptides and chelates for efficient chelation of ferrous ions.

Synthesis of peptide-iron chelates provides a novel approach to alleviating iron deficiency in humans. The study used hemp protein isolate to make hemp protein hydrolysates (HPHs), which were combined with ferrous chloride to make hemp protein peptide iron chelates (HPH-Fe). The HPH-Fe were structurally characterized and analyzed for stability. The strongest ferrous ion-binding fraction HPH-I (<3 kDa) was isolated by ultrafiltration, and Sephadex G-15 was used to obtain fractions with high ferrous ion-binding capacity (82.47%), identified by amino acid sequences as QASSDGFEWVSFK (1482.31 Da), …

Integrative approach to assessing bioactivity from hempseed protein isolate extracted and dehydrated by different methods: Synergising in silico prediction and in vitro validation.

This study demonstrated a comprehensive workflow combining in silico screening and prediction with in vitro validation to investigate the bioactivity of hempseed protein isolate (HPI) extracted and dehydrated using different methods. By adopting an in silico approach, 13 major proteins of HPI were hydrolysed by 20 selected enzymes, leading to the prediction of 20 potential bioactivities. With papain hydrolysis, dipeptidyl peptidase-IV (DPP4) and angiotensin-converting enzyme (ACE) inhibitory activities emerged as having the highest potential. In vitro experiments confirmed these predictions, w

Characterization of a metallo-proteinase from Bothrops asper (terciopelo) snake venom.

Metalloproteinase from the venom of Bothrops asper (proteinase G) is a glycoprotein with 1% neutral hexose and 3.5 moles of sialic acid per mole of protein. It hydrolyses a number of protein substrates such as casein, hemoglobin, gelatin and fibrinogen, whose alpha chain is degraded preferentially. The pH optimum of hydrolysis of casein is approximately 9.5. The protease is devoid of hemorraghic, esterolytic and amidolytic activities. The proteolytic activity of the enzyme increases by about 20% in the presence of 0.2 mM Ca2+ and Mg2+. Among the other ions tested, only Cd2+ and Fe2+ markedly d

Corn stover variability drives differences in bisabolene production by engineered Rhodotorula toruloides.

UNLABELLED: Microbial conversion of lignocellulosic biomass represents an alternative route for production of biofuels and bioproducts. While researchers have mostly focused on engineering strains such as Rhodotorula toruloides for better bisabolene production as a sustainable aviation fuel, less is known about the impact of the feedstock heterogeneity on bisabolene production. Critical material attributes like feedstock composition, nutritional content, and inhibitory compounds can all influence bioconversion. Further, the given feedstocks can have a marked influence on selection of suitable

Potential Effect of Enzymatic Porcine Placental Hydrolysate (EPPH) to Improve Alcoholic Liver Disease (ALD) by Promoting Lipolysis in the Liver.

Alcoholic liver disease is associated with the production of highly reactive free radicals by ethanol and its metabolites. Free radicals not only induce liver oxidation and damage tissues, but also stimulate an inflammatory response in hepatocytes, leading to severe liver disease. In order to improve alcoholic liver disease, enzymatic porcine placenta hydrolysate was studied by exploring various materials. Enzymatic porcine placenta hydrolysate (EPPH) contains various amino acids, peptides, and proteins, and is used as a useful substance in the body. In this study, changes were confirmed in in

Evaluation of infant feeding in acute gastroenteritis.

Sixty-eight bottle-fed babies under 9 months of age with mild acute gastroenteritis were observed to evaluate current feeding regimens following acute gastroenteritis in infancy. All babies were fed for 24 h with a glucose-electrolyte mixture (GEM) and then randomly assigned to either a gradual reintroduction to their normal milk, i.e., slow regrade; immediate return to full-strength formula; or a rapid regrade to a hypoallergenic whey hydrolysate formula. All groups were well matched for age, sex, ethnic origin, nutritional state, and degree of hydration. There was no significance difference

Sequential Ultrasound-Assisted Enzymatic Hydrolysis and Microwave Treatment of Gluten-Starch Complexes: Functionality, Volatile Profiles, In Vitro Immunoreactivity and Interactions.

Sequential non-thermal and thermal processing offers a route to jointly adjust the functionality, color/volatile profile and immunoreactivity of gluten-containing systems. This study aimed to evaluate the effects of sequential ultrasound-assisted enzymatic hydrolysis, wheat/rice starch incorporation and microwave heating on soluble gluten hydrolysate-starch composites, and to probe the underlying starch-gluten interactions. Gluten hydrolysates were blended with wheat or rice starch and microwave-heated to induce the Maillard reaction. Sequential processing enriched the <1 kDa peptide fraction

Discovery and validation of pumpkin seed protein-derived bioactive peptides targeting α-glucosidase: Insights into the molecular mechanisms.

Pumpkin seed protein (PSP) is a promising precursor of bioactive peptides, but the mechanisms underlying the α-glucosidase inhibitory activity of its hydrolysates are not yet fully understood. In the present work, a previously unreported tetrapeptide with α-glucosidase inhibitory activity, YYPW, was obtained from PSP through simulated gastrointestinal digestion and in silico screening. Kinetic assays demonstrated that YYPW is a potent mixed-type inhibitor (IC50 = 279.61 μmol/L). Molecular docking revealed that YYPW engages both the catalytic dyad (Asp443/Asp542) and peripheral aromatic residue

Molecular identification of salty and saltiness-enhancing peptides from Stropharia Rugosoannulata and their salinity perception with secondary structure and sodium chloride.

Given the significant health risks associated with high-salt foods, reducing salt intake in daily diets is very crucial. Saltiness enhancement peptides were identified and evaluated from Stropharia rugosoannulata protein hydrolysates using filtration methods, LC-MS/MS, and electronic tongue. Among those peptides, PQGDGW and WGFGDD with the lowest binding energies, recognized by two different receptors, transmembrane channel-like 4 (TMC4) and transient receptor potential vanilloid 1 (TRPV1), were synthesized. The sodium chloride perception concentration was 608.29 mmol/L for 0.1 % (w/v) PQGDGW …

Quantitative sequence-activity modeling of ACE peptide originated from milk using ACC-QTMS amino acid indices.

Up to now, numerous peptides/hydrolysates derived from casein and whey protein have shown angiotensin-I-converting enzyme (ACE) inhibitory. In this research, quantum topological molecular similarity (QTMS) indices of amino acids were utilized in quantitative sequence-activity modeling (QSAM) to predict the activity of a set of milk-driven peptides with ACE inhibition. Since the derived peptides have not the same number of residues, we overcame this issue by auto cross covariance (ACC) methodology. Then, some QSAMs were built to predict the pIC50 value of ACE peptides derived from Bovine Casein

Therapy of Crohn's disease in childhood.

The aim of therapy in Crohn's disease in childhood is to induce and to maintain a remission of disease activity so that normal growth and development of the child may occur. Enteral nutrition may now be recommended as the first-line treatment for most children with Crohn's disease. However, the evidence for remission is better for children with Crohn's disease of the small intestine rather than of the large intestine. There is evidence that amino acid feeds (elemental), whole protein (polymeric) and protein hydrolysate feeds (semi-elemental) may all be successful. Such a therapeutic approach c
