Down-regulation of liver Galbeta1, 4GlcNAc alpha2, 6-sialyltransferase gene by ethanol significantly correlates with alcoholic steatosis in humans.

Hepatic steatosis and steatohepatitis are frequent results of long-term ethanol exposure. We have previously demonstrated that long-term ethanol down-regulates Galbetal, 4GlcNAc alpha2, 6-sialyltransferase (ST6Gal1), leading to defective glycosylation of a number of proteins including apolipoprotein (apo) E and apo J and the appearance of asialoconjugates in the blood of continuously alcohol-fed animals as well as in human alcoholics. In the current study, we have explored the possibility of whether ethanol-induced down-regulation of ST6Gal1 could contribute toward alcoholic steatosis in human

Investigation of the differential transport mechanism of cinnabar and mercury containing compounds.

BACKGROUND: Cinnabar has a long history of uses in Chinese traditional medicines as an ingredient in various remedies. However, the detailed mechanism of cinnabar in medication remains unclear, and the toxicity of cinnabar has been a debate due to its containing mercury sulfide. This study was designed to investigate the differential transport mechanism of cinnabar and other Hg-containing compounds HgCl2, MeHg and HgS, and to determine if organic anion transporters OAT1 and OAT3 were involved in the differential transport mechanism. MATERIALS AND METHODS: The 293T cells were employed to inves

Calcium as a molecular switch that regulates Annexin A11 N- and C-terminal domains interaction and its role in ALS.

Amyotrophic Lateral Sclerosis (ALS) is a fatal neurodegenerative disease marked by progressive motor neuron loss, leading to muscle paralysis and respiratory failure. Genetic mutations, notably in the ANXA11 gene, have been implicated in both familial and sporadic ALS forms. ANXA11 functions as a cellular "tether," orchestrating the transport of RNA-protein complexes and lysosomes through its N-terminal (Nt) and C-terminal (Ct) domains, respectively. This study uncovers a novel calcium-dependent regulatory mechanism governing the intramolecular interaction between these domains. Using biochemi

XIAP promotes metastasis of bladder cancer cells by ubiquitylating YTHDC1.

X-linked inhibitor of apoptosis protein (XIAP), a member of the IAP family, is overexpressed in a variety of tumors and plays an important role in tumor progression. Increasing evidence suggests that XIAP promotes metastasis of bladder cancer but the underlying mechanism is not very clear. The RNA N6-methyladenosine (m6A) reader YTHDC1 regulates RNA splicing, nuclear transport, and mRNA stability and is a potential tumor target; however, its ubiquitin E3 ligase has not been described. In this study, screening of proteins that specifically interact with XIAP identified YTHDC1 as its degradation

A post-transcriptional respiratome regulon in trypanosomes.

Post-transcriptional regulons coordinate the expression of groups of genes in eukaryotic cells, yet relatively few have been characterized. Parasitic trypanosomatids are particularly good models for studies on such mechanisms because they exhibit almost exclusive polycistronic, and unregulated, transcription. Here, we identify the Trypanosoma brucei ZC3H39/40 RNA-binding proteins as regulators of the respiratome; the mitochondrial electron transport chain (complexes I-IV) and the FoF1-ATP synthase (complex V). A high-throughput RNAi screen initially implicated both ZC3H proteins in variant sur

Identification of Pasteurella multocida transcribed genes in porcine lungs through RNAseq.

Pasteurella multocida is one of the most important pathogen that causes pneumonia in swine. Although several virulence factors are known, the pathogenesis of this bacterium is not well-studied. Therefore, to study the pathogenesis of P. multocida infection in porcine lung, next-generation RNA sequencing was used to compare the transcriptomes of P. multocida grown in vivo and in vitro, respectively. After P. multocida infection a total of 704 genes were expressed in vitro, 1422 genes were expressed in vivo, and 237 genes were differentially expressed based on statistical analyses, padj of ≤0.1.

Effects of NELL2 on the regulation of GnRH expression and puberty in female rats.

Neural tissue-specific epidermal growth factor-like repeat domain-containing protein (NELL2) was previously found to play an important role in nerve growth, neural differentiation, neural elasticity, synaptic transport, and vesicle release. In this study, we examined the effect of NELL2 on gonadotropin-releasing hormone (GnRH) neurons and the initiation of puberty in female rats. We studied changes in NELL2 mRNA and protein expression at different stages of sexual development (postnatal days 30, 35, and 45) in female rats to determine the impact of NELL2 on GnRH mRNA expression. We also invest

Lipid-siRNA conjugate accesses perivascular transport and achieves durable knockdown throughout the central nervous system.

Short-interfering RNA (siRNA) has gained significant interest for treatment of neurological diseases by providing the capacity to achieve sustained inhibition of nearly any gene target. Yet, efficacious drug delivery throughout deep brain structures of the CNS remains a considerable hurdle for intrathecally administered therapeutics. We herein describe an albumin-binding lipid-siRNA conjugate that transports along meningeal and perivascular CSF pathways, leading to broad dispersion throughout the CNS parenchyma. We provide a detailed examination of the temporal kinetics of gene silencing, high

Molecular and Functional characterization of a novel C-type lectin during WSSV infection in Eriocheir sinensis.

C-type lectins (CTLs) are key pattern recognition receptors in innate immunity; however, their functions during white spot syndrome virus (WSSV) infection remain to be elucidated. Here, we identified a novel CTL gene from Chinese mitten crab (Eriocheir sinensis), designated as EsLec2. EsLec2 encodes a protein with a conserved carbohydrate-recognition domain (CRD) and shows phylogenetic clustering with other crustacean CTLs. RNA interference (RNAi) and recombinant protein (rEsLec2) injection assays revealed that EsLec2 downregulates ie1 and vp28 expression, indicating that it suppresses WSSV re

The pathogenic mechanism of TAR DNA-binding protein 43 (TDP-43) in amyotrophic lateral sclerosis.

The onset of amyotrophic lateral sclerosis is usually characterized by focal death of both upper and/or lower motor neurons occurring in the motor cortex, basal ganglia, brainstem, and spinal cord, and commonly involves the muscles of the upper and/or lower extremities, and the muscles of the bulbar and/or respiratory regions. However, as the disease progresses, it affects the adjacent body regions, leading to generalized muscle weakness, occasionally along with memory, cognitive, behavioral, and language impairments; respiratory dysfunction occurs at the final stage of the disease. The diseas

Cryptochrome 1 activation inhibits melanogenesis and melanosome transport through negative regulation of cAMP/PKA/CREB signaling pathway.

Cutaneous pigmentation was recently shown to be an event regulated by clock proteins. Cryptochrome (CRY) is a key protein composing the feedback loop of circadian clock, however, the function of CRY in melanocytes remains unclear. Here, we found that KL001, a synthetic small molecule modulator of CRY1, inhibited melanin synthesis, as well as reduced melanocyte dendrite elongation and melanosome transport. In addition, the dominant role of CRY1 in KL001-induced anti-melanogenesis was revealed by small interfering RNA transfection. Cellular tyrosinase activity and expression level of melanogenic

Functional Landscape of hnRNPA3 in Disease Pathogenesis.

The heterogeneous nuclear ribonucleic acid protein family participates in various intracellular reactions, such as RNA splicing, transport, DNA repair, cellular signal transduction, and gene expression regulation, and is involved in various disease processes. As a late-discovered member, heterogeneous nuclear ribonucleoprotein A3 has received increasing attention, but its main physiological functions and exact mechanisms involved in disease processes have not yet reached a consensus. In this review, we summarize the function of heterogeneous nuclear ribonucleoprotein A3 and the literature on i

Novel-and Not So Novel-Inhibitors of the Multifunctional CRM1 Protein.

Chromosome Region Maintenance 1 (CRM1), also known as Exportin 1 (XPO1), is a protein that is critical for transport of proteins and RNA to the cytoplasm through the nuclear pore complex. CRM1 inhibition with small molecule inhibitors is currently being studied in many cancers, including leukemias, solid organ malignancies and brain tumors. We review the structure of CRM1, its role in nuclear export, the current availability of CRM1 inhibitors, and the role of CRM1 in a number of distinct cellular processes. A deeper understanding of how CRM1 functions in nuclear export as well as other cellul

Analysis of the Babesia microti proteome in infected red blood cells by a combination of nanotechnology and mass spectrometry.

Proteomics of Babesia microti has lagged behind other apicomplexans despite recent genome and transcriptome studies. Here, we used a combination of nanotechnology and mass spectrometry to provide a proteomic profile of B. microti acute infection. We identified ∼500 parasite proteins in blood with functions such as transport, carbohydrate and energy metabolism, proteolysis, DNA and RNA metabolism, signaling, translation, lipid biosynthesis, and motility and invasion. We also identified surface antigens with roles in the immune response to the parasite. This first evaluation of the B. microti pr

TRPV6.

The ion channel TRPV6 is likely to function as an epithelial calcium channel in organs with high calcium transport requirements such as the intestine, kidney, and placenta. Transcriptional regulation of TRPV6 messenger RNA (mRNA) is controlled by 1,25-dihydroxyvitamin D, which is the active hormonal form of vitamin D3, and by additional calcium-dependent and vitamin D3-independent mechanisms. Under physiological conditions, the conductance of the channel itself is highly calcium-selective and underlies complex inactivation mechanisms triggered by intracellular calcium and magnesium ions. l-amino acid oxidases Lao1 and Il4i1 are major determinants of early intestinal transcriptional responses to DSS-induced stress.

The upstream regulators that initiate early transcriptional responses to intestinal stress remain poorly defined. Here, we used germ-free wild-type, Lao1-knockout (LAO1KO), and Lao1/Il4i1 double-knockout (DKO) mice to determine how two l-amino acid oxidases shape acute transcriptional responses to dextran sulfate sodium (DSS). RNA-sequencing performed 24 h after DSS exposure revealed that Lao1 accounted for 80.1% of DSS-induced transcriptional changes, while Il4i1 contributed an additional 14.7%, collectively accounting for 94.8% of the total response. Pathway analysis revealed that Lao1 was r

Comprehensive landscape and future perspectives of exosomal circular RNAs in cardiovascular disease.

Cardiovascular diseases continue to be a predominant cause of global mortality, driven by pathological molecular transport networks. Exosomes, which encapsulate diverse molecular cargo reflective of their cellular origins and associated pathological states, serve as valuable resources for identifying diagnostic and prognostic biomarkers. An increasing number of studies conducted in recent years have shown that exosomal circular RNA (circRNA) is important for a variety of cardiovascular diseases (CVDs), such as coronary heart disease, atherosclerosis, senescence, myocardial infarction, and myoc

GPNMB promotes double-membrane vesicle accumulation and facilitates structural protein transport during PEDV infection.

Porcine epidemic diarrhea virus (PEDV) causes substantial economic losses in the swine industry globally. Host factors regulating the intracellular replication of PEDV, particularly early RNA synthesis and structural protein production, are not well understood, limiting antiviral strategies. We report that glycoprotein non‑metastatic melanoma protein B (GPNMB) is a key host factor promoting PEDV infection. Initially identified as a PEDV S1-binding partner, GPNMB was confirmed to enhance infection via loss-and gain-of-function experiments in Vero and IPEC-J2 cells. Genetic knockout of GPNMB inh

Granuloma dual RNA-seq reveals composite transcriptional programs driven by neutrophils and necrosis within tuberculous granulomas.

Mycobacterial granulomas lie at the center of tuberculosis (TB) pathogenesis and represent a unique niche where infecting bacteria survive under nutrient-restricted conditions and in the face of a host immune response. The granuloma's necrotic core, where bacteria reside extracellularly in humans, is difficult to assess in many experimentally tractable models. Here, using necrotic mycobacterial granulomas in adult zebrafish, we develop dual RNA sequencing (RNA-seq) across different host genotypes to identify the transcriptional alterations that enable bacteria to survive within this key microe
