Time-dependent changes in CT of radiation-induced liver injury: a preliminary study in gastric cancer patients.

In this study, the time-dependent changes on dynamic computed tomograph (CT) of radiation-induced liver injury in gastric cancer patients was examined. The CT images of 52 gastric cancer patients who had received chemoradiotherapies were reviewed on the PACS system. Dynamic CT scan was performed in all the subjects. Our results showed that 18 patients were found to have radiation-induced liver injury. The CT findings of radiation-induced liver injury in gastric cancer patients tend to show up one month after radiation treatment. The damaged area was of low density on all three phases, and then

Qualitative effect on mRNAs of injury-associated proteins by cell phone like radiation in rat facial nerves.

Rats were exposed to cell phone radiation for 6 hours per day for 18 weeks. The buccal and mandibular branches of the facial nerve were evaluated for this study. The mRNA levels of four proteins that are usually up regulated when an injury has occurred were investigated; included were Calcium ATP-ase, Endothelin, Neural Cell Adhesion Molecule, and Neural Growth Factor. These isolated mRNAs were subjected to RT-PCR and all four were up regulated. The mandibular nerve showed a higher and broader level of up regulation than the buccal nerve. All four mRNA up regulations for the mandibular nerve a

Mobilization of circulating vascular progenitors in cancer patients receiving external beam radiation in response to tissue injury.

PURPOSE: Endothelial-like vascular progenitor cells (VPCs) are associated with the repair of ischemic tissue injury in several clinical settings. Because the endothelium is a principal target of radiation injury, VPCs may be important in limiting toxicity associated with radiotherapy (RT) in patients with cancer. METHODS AND MATERIALS: We studied 30 patients undergoing RT for skin cancer (n = 5), head-and-neck cancer (n = 15), and prostate cancer (n = 10) prospectively, representing a wide range of irradiated mucosal volumes. Vascular progenitor cell levels were enumerated from peripheral blo

Orazipone, a locally acting immunomodulator, ameliorates intestinal radiation injury: a preclinical study in a novel rat model.

PURPOSE: Intestinal radiation injury (radiation enteropathy) is relevant to cancer treatment, as well as to radiation accidents and radiation terrorism scenarios. This study assessed the protective efficacy of orazipone, a locally-acting small molecule immunomodulator. METHODS AND MATERIALS: Male rats were orchiectomized, a 4-cm segment of small bowel was sutured to the inside of the scrotum, a proximal anteperistaltic ileostomy was created for intraluminal drug administration, and intestinal continuity was re-established by end-to-side anastomosis. After three weeks postoperative recovery, t

Potential of T2 relaxation time measurements for early detection of radiation injury to the brain: experimental study in pigs.

PURPOSE: To investigate the MR T2 relaxation time and histologic changes after a single-fraction 25-Gy dose of radiation to the brain of pigs. METHODS: The right hemisphere of 10 Meishan pigs was irradiated with a single dose of 25 GY at the 90% isodose, using a 12-MeV electron beam. T2 relaxation time was measured within three regions of interest in the brain: those that had received 90%, 70%, and 40% of the total dose, respectively. T2 kinetics over time was compared with histologic studies. RESULTS: Brain T2 values were noted to increase within the irradiated areas. T2 kinetics were analy

[Injury of the large vessels of the neck caused by radiation of neoplasms of the otorhinolaryngologic region: a study using Doppler echography].

Twelve patients undergoing radiotherapy for pharyngo-laryngeal neoplasms were compared to a control group by means of duplex-Doppler US in order to evaluate early and late radiation-induced damage to the main arterial vessels in the neck region. On first examination the case distribution regarding the presence/absence of preexisting vascular lesions was: no lesions (8.3%), mild (33.3%), moderate (16.6%), and severe (41.6%) atherosclerotic lesions. The subjects included in the control group presented with similar characteristics, so that they had similar features to the patients undergoing radi

Duplex imaging and incidence of carotid radiation injury after high-dose radiotherapy for tumors of the head and neck.

Radiation-induced carotid artery disease following high-dose (greater than 50-Gy) radiotherapy for head and neck cancer may become more common as improved treatment results in longer survival. Duplex ultrasound scans were obtained in 91 consecutive patients to determine whether increased incidence and severity of extracranial carotid disease correlate with prior radiotherapy. Fifty-three patients who underwent radiotherapy an average of 28 months previously and 38 patients who received no radiotherapy were studied. Thirty percent of the irradiated group had lesions of the carotid arteries that

Delayed radiation injury to the retrobulbar optic nerves and chiasm. Clinical syndrome and treatment with hyperbaric oxygen and corticosteroids.

Thirteen patients with delayed radiation injury to the optic nerves and chiasm were treated with hyperbaric oxygen (HBO) and corticosteroids. These patients experienced painless, abrupt loss of vision in one (6 patients) or both (7 patients) eyes between 4 and 35 months after receiving radiation doses of at least 4500 cGy to the region of the chiasm. Diagnostic evaluation including neuro-imaging and lumbar puncture showed no recurrent tumor and no other cause for visual loss. No patient's vision improved during treatment or follow-up lasting between 1 and 4 years. There were no serious complic

Serum tissue polypeptide antigen (TPA): a marker of acute injury of salivary glands during radiation therapy.

The modifications of serum concentrations of TPA were monitored in patients undergoing curative radiation therapy. Patients with tumors localized in the head and neck were treated with one of four different schedules based on conventional fractionation or multiple daily fractionation where the dose per fraction and total daily dose varied. Serum TPA increased immediately on the first day of irradiation: the higher the dose, the greater the increase. These increases disappeared rapidly after the first few days of treatment. A more limited rise was observed in some cases when treatment was renew

Post-Irradiation Treatment with a Superoxide Dismutase Mimic, MnTnHex-2-PyP5+, Mitigates Radiation Injury in the Lungs of Non-Human Primates after Whole-Thorax Exposure to Ionizing Radiation.

Radiation injury to the lung is the result of acute and chronic free radical formation, and there are currently few effective means of mitigating such injury. Studies in rodents indicate that superoxide dismutase mimetics may be effective in this regard; however, studies in humans or large animals are lacking. We hypothesized that post-exposure treatment with the lipophilic mitochondrial superoxide dismutase mimetic, MnTnHex-2-PyP5+ (hexyl), would reduce radiation-induced pneumonitis and fibrosis in the lungs of nonhuman primates. Rhesus monkeys (Macaca mulatta) received 10 Gy whole thorax irr

Potentiating effect of fetal thymus on fetal liver cells for the promotion of recovery from the radiation injury in murine allogenic radiation chimeras or a little bit of thymus goes a long, long way.

Lethally irradiated CBA (H-2k) mice were treated with small numbers of liver cells from strain-A (H-2a) fetal donors. Recovery from the radiation injury was dependent upon the dose of liver cells administered. Giving very small numbers of fetal thymus cells, in addition to liver cells, had a highly significant and salutary effect on survival. The results suggest that clinical transplantation of liver cells from a single fetus might succeed if thymus cells are added.

Berberine alleviates radiation-induced lung injury by promoting microbiota-derived inosine via the gut-lung axis.

Radiation-induced lung injury (RILI) is a common complication of radiotherapy. Although berberine (BBR) has been suggested to be associated with reduced RILI incidence, the underlying mechanisms remain unknown. Here, we investigated whether the gut microbiota mediates the radioprotective effects of BBR using a C57BL/6 RILI mouse model with 20 Gy thoracic irradiation (n = 6 per group). BBR (100 mg/kg) and inosine (INO, 300 mg/kg) were administered orally in vivo. Antibiotic depletion and fecal microbiota transplantation were performed to assess microbiota dependence. Lung injury was assessed by

Upregulation of S100A8/A9 Potentiates Radiation-Induced Intestinal Injury via MAPK and NF-κB Signaling Pathways.

PURPOSE: Radiation-induced intestinal injury (RIII) is a serious complication of radiation therapy without effective therapies. The alarmin, S100A8/A9, a biomarker of gastrointestinal inflammation, is highly elevated following abdominal irradiation (ABI). However, it remains unclear whether it functions as an active driver of RIII pathogenesis. The study aims to assess the effects of S100A8/A9 on the pathogenesis of RIII. Our goal is to identify its dynamic alteration during RIII and determine whether it can be used as a therapeutic target in the injury and repair phases of acute RIII. METHOD

Impact of COVID-19 vaccination on radiation-induced lung injury in breast cancer patients: a retrospective cohort study.

BACKGROUND: The safety of COVID-19 vaccination during radiotherapy remains a concern among oncologists. This study investigates whether vaccination increases the risk of radiation-induced lung injury (RILI) in breast cancer patients. METHODS: We retrospectively analyzed 157 breast cancer patients treated with radiotherapy between April 2021 and October 2022. Lung injury was assessed through post-treatment imaging and correlated with COVID-19 vaccination status. RESULTS: Of 157 patients, 70 (44%) developed radiologic signs of radiation induced lung injury. Most patients (85%) were vaccinated.

Integrative multi-omics profiling implicates purine metabolism and the PI3K-Akt pathway in radiation-induced lung injury.

BACKGROUND: Thoracic radiotherapy commonly causes radiation-induced lung injury (RILI); however, its mechanisms remain poorly defined, and no FDA-approved therapies are available. METHODS: To identify key pathways and regulators, we analyzed metabolic and transcriptomic alterations in a murine model of RILI. Following a 15 Gy thoracic irradiation, lung tissues were assessed by histopathology (H&E and Masson staining), LC-MS-based metabolomics, and transcriptomics. RESULTS: Irradiated lungs showed erythrocyte exudation, inflammatory infiltration, fibrosis, and epithelial damage, whereas contr

Unraveling the temporal dynamics of radiation-induced lung injury: a multi-omics integration of transcriptomic, proteomic, and metabolic reprogramming.

Radiation-induced lung injury (RILI) arises from the unavoidable exposure of normal lung tissue during radiotherapy for thoracic malignancies. It remains a dose-limiting complication in thoracic radiotherapy, critically impacting treatment efficacy and long-term patient survival. Despite its clinical significance, the dynamic molecular reprogramming underlying RILI progression remains poorly characterized. Utilizing a C57BL/6J thoracic irradiation mouse model validated by histopathological analysis, we implemented an integrative tri-omics approach (RNA-seq, LC-MS/MS proteomics, and UHPLC-QTOF

Effects of probiotics BB-12 and Bifico on radiation-induced brain injury, cognitive dysfunction and gut microbiota composition.

BACKGROUND: Whole brain irradiation (WBI) is an essential treatment for brain tumors and metastases, but often leads to radiation-induced brain injury (RIBI), and cognitive dysfunction, significantly impairing patients' quality of life. Emerging evidence highlights the role of gut microbiota dysbiosis in neurological disorders via the microbiota-gut-brain axis (MGBA). This study investigated the neuroprotective effects of probiotics Bifidobacterium animalis subsp. lactis BB-12 (BB-12) and Bifico on WBI-induced cognitive impairment in mice.
