Non-invasive vagus nerve stimulation and the motivation to work for rewards: A replication of Neuser et al. (2020, Nature Communications).

The vagus nerve is thought to be involved in the allostatic regulation of motivation and energy metabolism via gut-brain interactions. A recent study by Neuser and colleagues (2020) provided novel evidence for this process in humans, by reporting a positive effect of transcutaneous auricular vagus nerve stimulation (taVNS) on the invigoration of reward-seeking behaviors, especially for food rewards. We conducted an independent direct replication of Neuser et al. (2020), to assess the robustness of their findings. Following the original study, we used a single-blind, sham-controlled, randomized

The Baltimore Classification of Viruses 50 Years Later: How Does It Stand in the Light of Virus Evolution?

Fifty years ago, David Baltimore published a brief conceptual paper delineating the classification of viruses by the routes of genome expression. The six "Baltimore classes" of viruses, with a subsequently added 7th class, became the conceptual framework for the development of virology during the next five decades. During this time, it became clear that the Baltimore classes, with relatively minor additions, indeed cover the diversity of virus genome expression schemes that also define the replication cycles. Here, we examine the status of the Baltimore classes 50 years after their advent and

Wolbachia strain wAlbB blocks replication of flaviviruses and alphaviruses in mosquito cell culture.

BACKGROUND: Wolbachia pipientis are bacterial endosymbionts of arthropods currently being implemented as biocontrol agents to reduce the global burden of arboviral diseases. Some strains of Wolbachia, when introduced into Aedes aegypti mosquitoes, reduce or block the replication of RNA viruses pathogenic to humans. The wAlbB strain of Wolbachia was originally isolated from Aedes albopictus, and when transinfected into Ae. aegypti, persists in mosquitoes under high temperature conditions longer than other strains. The utility of wAlbB to block a broad spectrum of RNA viruses has received limite

Kaposi's sarcoma-associated herpesvirus (human herpesvirus 8) replication and transcription factor activates the K9 (vIRF) gene through two distinct cis elements by a non-DNA-binding mechanism.

The replication and transcription activator (RTA) of Kaposi's sarcoma-associated herpesvirus (KSHV), or human herpesvirus 8, a homologue of Epstein-Barr virus BRLF1 or Rta, is a strong transactivator and inducer of lytic replication. RTA acting alone can induce lytic replication of KSHV in infected cell lines that originated from primary effusion lymphomas, leading to virus production. During the lytic replication process, RTA activates many kinds of genes, including polyadenylated nuclear RNA, K8, K9 (vIRF), ORF57, and so on. We focused here on the mechanism of how RTA upregulates the K9 (vIR

Enteroviruses identifications and differentiation on the basis of the selective group--specific inhibitory effect of chemical compounds.

Two new enteroviruses (EV) inhibitors with the selective group-specific effect were detected and studied representing the products of the original chemical synthesis. One of them--nifan (arylfuran derivative) inhibits poliomyelitis virus replication, the other one--belvtazide (synchonic acid derivative) blocks non-poliomyelitis EV (ECHO and Coxsackie B) replication. The study of the reference strains of poliomyelitis virus type 1-3, twenty-three ECHO virus types (from the 1st to the 33rd), Coxsackie B virus type 1-6 and 288 primary EV isolates did not reveal type or strain specific variability

Analysis of La Crosse virus S-segment RNA and its positive-sense transcripts in persistently infected mosquito tissues.

La Crosse (LAC) virus is an important cause of pediatric arboviral encephalitis in the United States. LAC virus is biologically transmitted by the mosquito Aedes triseriatus, and, like other arthropod-borne viruses, it establishes a persistent, nonpathogenic infection in its vector following oral infection. To investigate LAC virus persistent infection of mosquitoes, a reverse transcription-PCR assay was developed for the amplification of LAC virus negative-sense small (S) genome RNA segment, its full-length complement, and its mRNA transcript for qualitative analysis of transcription and repl

Development, Reliability, and Initial Validation of the Fear of Institutional Betrayal Questionnaire.

Fear of institutional betrayal is a new conceptual extension of the original institutional betrayal framework. The original framework is a way of making sense of, and giving credibility to, the experiences of harm perpetrated by institutions. The original authors developed a checklist of institutional actions or inactions that harm survivors of sexual violence. Building on this work, fear of institutional betrayal is an attitudinal construct that represents the belief that post-secondary institutions will inevitably fail to prevent or acceptably respond to campus sexual violence, and that enga

Validation of the Multiracial Youth Socialization Brief (MY-Soc-B) Scale with adolescents.

The present study validates a brief version of the Multiracial Youth Socialization Scale, the MY-Soc-B, which assesses Multiracial youths' experiences of receiving messages from their caregivers about race and culture. Using a sample of 318 diverse Multiracial American adolescents (Mage = 16.01; SD = 1.30), the original 62 items were reduced to 24 items via an examination of confirmatory factor analyses and conceptual clarity and fit of item content. The model fit strongly supported retaining the eight-factor structure from the original full version. Validity and reliability of the eight subsc

Field porcine reproductive and respiratory syndrome viruses (PRRSV) attenuated by codon pair deoptimization (CPD) in NSP1 protected pigs from heterologous challenge.

Two type 2 field porcine reproductive and respiratory syndrome viruses (PRRSV) isolated from PRRS-affected swine farms were attenuated by de-optimization of codon pair bias in NSP1. In 3-week-old pigs infection, the attenuated viruses showed significantly lower replication ability than the original viruses without distinct clinical sign and pathological lesions, which were observed in pig infected with the original viruses. Regarding induction of PRRSV specific immunity, the level of the neutralizing antibodies as well as secretion of IFN-γ-SCs in PBMCs was not different between the attenuated

In Ovo Administration of Silver Nanoparticles Enhances Post-Hatch Growth Performance, Metabolic and Physiological Responses, and Immune Function in Broiler Chickens.

This study examined the effects of in ovo silver nanoparticle (AgNP) administration on broiler chicken hatchability, growth performance, physiological and metabolic status, antioxidant capacity, and immune responses. A total of 300 fertilized Cobb500 eggs were assigned to five treatments (5 replicates/treatment, 12 eggs/replicate): non-injected control; vehicle control (0.2 mL sterile 0.9% NaCl/egg); and AgNPs at 10, 15, or 20 ppm injected into their amniotic sac on day (d) 18 of incubation. After hatching, 48 chicks/treatment were assigned to 6 replicate cages (8 birds/replicate) for 35 d. In

Super-resolution atlas of SARS-CoV-2 infection reveals protease-dependent organelle maturation, dsRNA landscapes, and intracellular structural proteins.

Coronaviral replication depends on double-membrane vesicles (DMVs), yet where polyprotein processing occurs and how replication organelles mature remain unresolved. We built a multi-color super-resolution atlas of SARS-CoV-2 RNA, non-structural and structural proteins in infected human cells, combining 3D single-molecule localization with radial and angular pair-correlation analysis. The atlas uncovers fundamental spatial principles. First, nsp5 (3CLpro) localizes within the DMV lumen near pores, with nsp7-nsp16 positioned interior to nsp4, supporting a protease-dependent maturation model in w

Amoebae-Based Screening Reveals a Novel Family of Compounds Restricting Intracellular Legionella pneumophila.

The causative agent of Legionnaires' disease, Legionella pneumophila, grows in environmental amoebae and mammalian macrophages within a distinct compartment, the 'Legionella-containing vacuole' (LCV). Intracellular bacteria are protected from many antibiotics, and thus are notoriously difficult to eradicate. To identify novel compounds that restrict intracellular bacterial replication, we previously developed an assay based on a coculture of amoebae and GFP-producing L. pneumophila. This assay was used to screen a pathway-based, highly diverse chemical library, referred to as the Sinergia libr

Transcranial direct current stimulation (tDCS) and the 9-dot problem.

Our principal objective was to replicate the findings of Chi and Snyder (2012) who reported that subjects (45% of sample) receiving active transcranial direct current stimulation (tDCS) involving cathodal stimulation of the left anterior temporal lobe (ATL) and anodal stimulation of the right ATL were able to successfully complete a task considered exceedingly challenging to solve (i.e., the 9-dot problem) relative to participants receiving sham stimulation (solved problem = 0 %). There are, to our knowledge, no published replication studies of Chi and Snyder's original work targeting the ATL.

Dissociation in focus: replicating and extending network analytic insights based on the dissociative experiences scale.

Background: Factor-analytic studies of the Dissociative Experiences Scale (DES) have reported between one- and seven-factor solutions, leaving its dimensional architecture unresolved. This inconsistency compromises the interpretability of scores and limits the scale's utility for screening and treatment planning. To ensure diagnostic relevance and capture dissociation across mental disorders, we focused on a clinical cohort. Emerging evidence from network-analytic approaches suggests a stable three-factor structure, though replication is limited. Objective: We employed Exploratory Graph Analysi

Reduction of antisense transcription affects bovine leukemia virus replication and oncogenesis.

In sheep infected with bovine leukemia virus (BLV), transcription of structural, enzymatic, and accessory genes is silenced. However, the BLV provirus transcribes a series of non-coding RNAs that remain undetected by the host immune response. Specifically, three RNAs (AS1-L, AS1-S, and AS2) are consistently expressed from the antisense strand, originating from transcriptional initiation at the 3'-Long Terminal Repeat (LTR). To investigate the role of these non-coding RNAs in viral replication and pathogenesis, a reverse genetics approach was devised, capitalizing on a mechanistic disparity in

Alphavirus-based replicons demonstrate different interactions with host cells and can be optimized to increase protein expression.

Alphavirus replicons are being developed as self-amplifying RNAs aimed at improving the efficacy of mRNA vaccines. These replicons are convenient for genetic manipulations and can express heterologous genetic information more efficiently and for a longer time than standard mRNAs. However, replicons mimic many aspects of viral replication in terms of induction of innate immune response, modification of cellular transcription and translation, and expression of nonstructural viral genes. Moreover, all replicons used in this study demonstrated expression of heterologous genes in cell- and replicon

Expression of the bacterial hemoglobin gene from Vitreoscilla stercoraria increases rifamycin B production in Amycolatopsis mediterranei.

It is well known that the culture for rifamycin B production by Amycolatopsis mediterranei requires high levels of dissolved oxygen, particularly in industrial processes. In this study, we report the construction of a vector for the expression of the bacterial hemoglobin gene (vhb) from Vitreoscilla stercoraria in a rifamycin B-overproducing strain of A. mediterranei. The effect was evaluated in the presence and absence of barbital. The vhb gene was cloned under the control of the PermE promoter, the Amycolatopsis lactamdurans plasmid pULVK2 origin of replication, the kanamycin-resistant gene

Herpes simplex virus infection generates large tandemly reiterated simian virus 40 DNA molecules in a transformed hamster cell line.

When the simian virus 40 (SV40)-transformed Syrian hamster cell line Elona is infected with herpes simplex virus type 1, an excessive amplification of SV40-specific DNA sequences occurs. Analysis of total DNA from herpes simplex virus-infected cells revealed that amplified DNA sequences were present predominantly in a high-molecular-weight form, consisting of a tandem array of many unit-length SV40 DNA molecules. Repeat units of amplified DNA were found to be very similar to standard SV40 DNA as was shown by restriction analyses, except for a small deletion close to the origin of replication,

Immunosuppression in viral infections.

Viruses may cause immunosuppression by a variety of mechanisms. This review delineates four categories. First, immunosuppression can result from the direct effects of viral replication on lymphocyte functions. Either all classes of lymphocytes can be affected, as occurs in measles, or the effect can be restricted to a cell subtype, as is the case with human T cell-lymphotropic virus type III. Second, the activity of soluble factors of viral or host origin released from infected cells can affect immunosuppression. A third mechanism results from viral infection of macrophages and affects the fun

Novel virally targeted therapies of EBV-associated tumors.

EBV is associated to the development of several malignancies of lymphoid and epithelial origin, including Burkitt's Lymphoma, post-transplant lymphoproliferative disorders, Hodgkin's disease, AIDS-associated lymphomas, NK/T cell lymphoma and Nasopharyngeal carcinoma. EBV genes play an essential role in the development of the malignant phenotype and therefore molecules interfering with the function of these genes may represent an essential tool to treat EBV-associated malignancies. Several strategies to inhibit virus-induced tumorigenesis have been developed including antiviral and antitumor mo
