Understanding and Reporting PFAS Exposure: Wide-Scope Retrospective Suspect Screening of High-Resolution Mass Spectrometry Data from Exposomics Studies.

Per- and polyfluoroalkyl substances (PFAS) comprise thousands of persistent and structurally diverse chemicals that contribute to chronic human exposure. Yet, routine biomonitoring targets only a limited set of regulated legacy compounds, underestimating overall exposure. We present a retrospective, wide-scope suspect screening analysis framework to extract PFAS information from archived LC-HRMS exposomics datasets. A curated list of 1,666 PFAS, filtered for reversed-phase LC compatibility and enriched with structural descriptors, predicted retention indices, and MS2 fragments, was used to rep

Pericardiocentesis vs. surgical pericardial window for first occurrence of malignancy-related pericardial effusion: a meta-analysis of retrospective studies.

AIMS: Malignancy-related pericardial effusion (MRPE) is a serious complication of advanced cancer, potentially leading to life-threatening cardiac tamponade. While both pericardiocentesis and surgical pericardial window are used for management, data on comparative efficacy and safety remain heterogeneous. This study aims to compare the efficacy and safety of pericardiocentesis vs. surgical pericardial window in the management of the first occurrence of MRPE. METHODS AND RESULTS: We systematically searched PubMed, Cochrane, and Google Scholar up until May 2025. Articles directly comparing peri

Herpes simplex virus type 2 encephalitis in critically ill adults: A retrospective analysis of EURECA and Herpetics cohort studies.

BACKGROUND: Herpes simplex virus type 1 (HSV-1) is the leading cause of sporadic infectious encephalitis. Although typically associated with benign meningitis, HSV-2 can also cause encephalitis in adults, a condition that remains poorly characterized in intensive care unit (ICU). We aimed to compare the characteristics, management, and outcomes of critically ill adult with HSV-2 and HSV-1 encephalitis. METHODS: We performed a retrospective analysis of adults with herpes simplex encephalitis (HSE) admitted to the ICU in two large multicenter cohorts. Patients were classified according to cerebr

Non-invasive cardiac stress studies may not offer significant benefit in pre-kidney transplant evaluation: A retrospective cohort study.

BACKGROUND: Screening with cardiac non-invasive stress studies (NISS) prior to listing for kidney transplantation can help in identifying treatable coronary disease and is considered an integral part of pre-kidney transplant evaluation. However, few studies assessed their effectiveness in all patients evaluated for transplantation in clinical practice. To evaluate the role of NISS in pre-kidney transplant evaluation we analyzed their impact prior to waitlisting in 1053 adult CKD-5 patients consecutively evaluated in Greater Manchester, UK during a 6-year period. METHODS: 918 waitlisted patien

The impact of exposure to antidepressant medications during pregnancy on neonatal outcomes: a review of retrospective database cohort studies.

INTRODUCTION: Concerns with prescription antidepressant use in pregnant women have instigated the examination of potential associations between fetal exposure to antidepressant medication and outcomes including preterm delivery, congenital malformations, perinatal and post-natal adverse events, persistent pulmonary hypertension, and mortality. The retrospective cohort model is an often utilized study design. The objective of this review is to evaluate the literature on antidepressant use in pregnancy conducted as retrospective cohorts in national/regional medical, or claims databases that asse

A Retrospective Study on the Impact of Primary Aldosteronism in Pregnancy (H.A.P.P.I. Study Group).

OBJECTIVE: Primary aldosteronism (PA) is a common cause of hypertension in young women. However, there is a paucity of data regarding its impact during pregnancy. The primary objective of this study was to describe the occurrence of hypertensive disorders of pregnancy (HDP) in PA. DESIGN: This retrospective cohort study compared outcomes in pregnancies according to PA status. PATIENTS: Pregnancies occurred between 2011 and 2022 at the Centre Hospitalier Universitaire de Sherbrooke. All selected pregnancies were screened for PA with an aldosterone-to-renin ratio (ARR) within a 5-year period.

Long-term prospective clinical follow-up after BRCA1/2 presymptomatic testing: BRCA2 risks higher than in adjusted retrospective studies.

BACKGROUND: The risks of breast cancer associated with BRCA1 and BRCA2 mutations vary considerably across studies but few have assessed prospective risks, which are likely to provide more reliable risk estimates for women undergoing presymptomatic testing. METHODS: Prospective breast cancer risks were assessed in 254 unaffected women with BRCA1 mutations and 238 with BRCA2 mutations. Rates of breast cancer were calculated allowing for lead time bias and censored at time of risk reducing mastectomy. Degree of family history was assessed using the Manchester score and genotyping was undertaken
