Specificity of direct transition from wake to REM sleep in orexin/ataxin-3 transgenic narcoleptic mice.

To create operational criteria for polygraphic assessments of direct transitions from wake to REM sleep (DREM), as a murine analog of human cataplexy, we have analyzed DREM episodes in congenic lines of orexin/ataxin-3 transgenic [TG] mice and wild-type littermates. The sleep stage of each 10-second epoch was visually scored using our standard criteria. Specificity of DREM for narcoleptic TG mice and sensitivity to detect DREM was evaluated using different DREM criteria. We found that DREM transitions by 10-second epoch scoring are not specific for narcoleptic TG mice and also occur in WT mice

Instantaneous acceleration and amplification of hippocampal theta wave coincident with phasic pontine activities during REM sleep.

Rapid eye movement (REM) sleep is characterized by hippocampal theta waves and phasic spike-like waves originating from the pons, termed ponto-geniculo-occipital (PGO) waves in cats and pontine (P) waves in rats. While the theta wave and PGO/P wave have been suggested to participate in higher-order brain functions, their generation mechanisms and roles in brain functions have been studied independently. Therefore, the present study investigated instantaneous aspects of the relationship between theta waves and PGO/P waves in both cats and rats. Theta wave was instantaneously accelerated several

Effect of slow wave and REM sleep on thyropharyngeus and stylopharyngeus activity during induced central apneas.

The pharyngeal constrictors have been hypothesized to play an important role in the regulation of upper airway (UAW) patency in patients with sleep apnea. However, little research has focused on the activation and control of muscles that determine the lateral and posterior wall of the retropalatal airway dimensions. Our aim was to investigate the effects of slow wave sleep (SWS) and rapid eye movement (REM) sleep on the activation of pharyngeal constrictor (thyropharyngeus; TP) and dilator (stylopharyngeus; SP) muscles during eupneic breathing and induced central apneas. In nine goats, we foun

Spectral features of EEG alpha activity in human REM sleep: two variants with different functional roles?

Evidence suggests that an important contribution of spectral power in the alpha range is characteristic of human REM sleep. This contribution is, in part, due to the appearance of well-defined bursts of alpha activity not associated with arousals during both tonic and phasic REM fragments. The present study aims at determining if the REM-alpha bursts constitute a different alpha variant from the REM background alpha activity. Since previous findings showed a selective suppression of background alpha activity over occipital regions during phasic REM fragments and, on the other hand, the density

Decreased activity of striatal monoamine oxidase B after rapid eye movement (REM) sleep deprivation in rats.

The striatum seems to be the main brain region involved in stereotyped behavior induced by dopaminergic agonists. Rapid eye movement (REM) sleep deprivation increases dopaminergic agonist-induced stereotypy and produces biochemical changes in striatal dopaminergic neurotransmission. However, the mechanism underlying the increased dopaminergic sensitivity induced by REM sleep deprivation has not been elucidated. In an attempt to determine some of the biochemical changes in striatal dopaminergic neurotransmission that could contribute to REM sleep deprivation effects, we measured the activity of

Respiratory muscle activity during rapid eye movement (REM) sleep in patients with chronic obstructive pulmonary disease.

BACKGROUND: In patients with chronic obstructive pulmonary disease (COPD) periods of hypopnoea occur during rapid eye movement (REM) sleep, but the mechanisms involved are not clear. METHODS: Ten patients with stable COPD were studied during nocturnal sleep. Detailed measurements were made of surface electromyographic (EMG) activity of several respiratory muscle groups and the accompanying chest wall motion using magnetometers. RESULTS: Hypopnoea occurred in association with eye movements during phasic rapid eye movement (pREM) sleep. During pREM sleep there were reductions in EMG activity o

An Intensified Training Period Reduces REM Sleep but this Decline Can Be Mitigated by Ketone Ester Supplementation.

INTRODUCTION: We previously showed that a strenuous training day disrupts sleep efficiency and REM sleep, and that these disturbances can be counteracted by ketone monoester (R)-3-hydroxybutyl-(R)-3-hydroxybutyrate (KE) supplementation. However, the impact of prolonged intensified training on sleep quality, as well as the potential modulating effect of KE are unknown. METHODS: We performed a double-blind study in which twenty-four trained male athletes completed a 7-week training program with a gradual increase in training load while receiving either 25g of a control drink (CON, n=12) or 25g

REM Sleep is Associated with Cognition and Biomarkers Longitudinally in Older Adults Across the Alzheimer Disease Continuum.

Sleep disturbances represent a potentially modifiable risk factor for Alzheimer disease (AD). The extent to which changes in rapid eye movement (REM) sleep are related to the accumulation of AD pathology and brain tissue loss is not well understood. In the current study, ninety-four individuals 74 clinically unimpaired [CU] and 20 clinically impaired [CI]) underwent polysomnography (PSG), cognitive assessments, and neuroimaging. Cross-sectional and longitudinal models examined the associations between PSG outcomes of interest (percentage of sleep period spent in REM [%REM] and REM latency) and

Cortical and subcortical morphometric changes and their relation to cognitive impairment in isolated REM sleep behavior disorder.

OBJECTIVE: To date, very few studies have focused on structural changes and their association with cognitive performance in isolated REM sleep behaviour disorder (iRBD). Moreover, the results of these studies are inconclusive. This study aims to evaluate differences in the associations between brain morphology and cognitive tests in iRBD and healthy controls. METHODS: Sixty-three patients with iRBD and thirty-six controls underwent MRI with a 3 T scanner. The cognitive performance was assessed by a comprehensive neuropsychological battery. Based on performance, the iRBD group was divided into

The REM-sleep-related characteristics of narcolepsy: a nation-wide multicenter study in Turkey, the REMCON study.

INTRODUCTION: Narcolepsy type 1 (NT1) is caused by hypocretin deficiency, the pathophysiology of narcolepsy type 2 (NT2) has not been delineated. Except for the hypocretin deficiency and cataplexy, all clinical and laboratory features used in the diagnosis of NT2 are identical to those used for NT1. The aim of this study was to assess the rapid eye movement (REM) sleep-related characteristics in the patients with narcolepsy; the characteristics of REM sleep in polysomnography (PSG) and multiple sleep latency test (MSLT) recordings, the quantification of REM sleep without atonia (RSWA) and aton

REM sleep in naps differentially relates to memory consolidation in typical preschoolers and children with Down syndrome.

Sleep is recognized as a physiological state associated with learning, with studies showing that knowledge acquisition improves with naps. Little work has examined sleep-dependent learning in people with developmental disorders, for whom sleep quality is often impaired. We examined the effect of natural, in-home naps on word learning in typical young children and children with Down syndrome (DS). Despite similar immediate memory retention, naps benefitted memory performance in typical children but hindered performance in children with DS, who retained less when tested after a nap, but were mor

Distribution of REM latencies and other sleep phenomena in depression as explained by a single ultradian rhythm disturbance.

The McCarley-Hobson model, describing the alternation of NREM and REM sleep in the cat, was applied to human electroencephalographic data. The influence of initial conditions on oscillatory behavior was especially emphasized. It appears that the distribution of REM latency in depression, the abnormal accumulation of REM sleep, the variability of NREM-REM cycle duration, the frequent stage shifts, and frequent awakenings can be explained in this model by means of a decrease in the initial value of a single variable, which may be regarded as representing the strength of REM inhibition. The obser

The Impact of REM Sleep Deprivation on ER Stress and Alzheimer-Like Pathology: Therapeutic Potential of Melatonin.

REM sleep deprivation (REMSD) contributes to neurodegenerative diseases like Alzheimer’s Disease (AD). This study examined REMSD’s effects on cognition, ER stress, and AD-like pathology in rats, alongside melatonin’s therapeutic potential. REMSD was induced for six days using the modified multiple platform method (MMPM). Cognitive function was tested via the Morris Water Maze (MWM). Rats received melatonin (20 mg/kg), 4-PBA (as a positive control, 100 mg/kg), or vehicle during deprivation/recovery periods. Western blotting and ELISA assessed ER stress markers (BiP, pIRE1, pPERK, peIF2-α, CHOP

Caffeine-augmented exercise as a pretreatment for locomotor and balance impairments induced by REM sleep deprivation in rats.

Sleep deprivation (SD) is a common problem that can lead to various neurological disorders. This study was carried out to examine how SD impacts locomotor performance and coordination in rats. Moreover, we aimed to investigate the potential synergistic benefits of caffeine supplementation coupled with treadmill exercise in mitigating any locomotor disorders induced by SD. Male rats were assigned to five groups: control, SD, SD + caffeine, SD + exercise, SD + caffeine + exercise. After 5 weeks of receiving caffeine supplementation (30 mg/kg) and/or treadmill exercise, the rats underwent 72 h of

The inter-night variability of REM sleep without atonia in adult patients with central disorders of hypersomnolence.

STUDY OBJECTIVES: Rapid eye movement (REM) sleep without atonia (RWA), the neurophysiological hallmark of REM sleep behavior disorder, is a typical feature of nocturnal sleep in patients with narcolepsy type 1 (NT1). Its reproducibility across different nocturnal polysomnographies (PSG) has never been investigated in patients with CDH. We assessed the inter-night variability of RWA. METHODS: Two-hundred-fifty adults (45 % males) hospitalized for suspected CDH, including 92 NT1, 29 narcolepsy type 2 (NT2), 36 idiopathic hypersomnia (IH), and 93 subjective hypersomnolence (sHS), undergoing two

Opportunities and Pitfalls of REM Sleep Behavior Disorder and Olfactory Dysfunction as Early Markers in Parkinson's Disease.

During its pre-motor stage, Parkinson's disease (PD) presents itself with a multitude of non-motor symptoms with different degrees of specificity and sensitivity. The most important among them are REM sleep behavior disorder (RBD) and olfactory dysfunction. RBD is a parasomnia characterized by the loss of REM sleep muscle atonia and dream-enacting behaviors. Olfactory dysfunction in individuals with prodromal PD is usually described as hyposmia (reduced sense of smell) or anosmia (complete loss of olfactory function). These symptoms can precede the full expression of motor symptoms by decades.

Occipital hypometabolism is a risk factor for conversion to Parkinson's disease in isolated REM sleep behaviour disorder.

PURPOSE: Isolated REM sleep behaviour disorder (iRBD) patients are at high risk of developing clinical syndromes of the α-synuclein spectrum. Progression markers are needed to determine the neurodegenerative changes and to predict their conversion. Brain imaging with 18F-FDG PET in iRBD is promising, but longitudinal studies are scarce. We investigated the regional brain changes in iRBD over time, related to phenoconversion. METHODS: Twenty iRBD patients underwent two consecutive 18F-FDG PET brain scans and clinical assessments (3.7 ± 0.6 years apart). Seventeen patients also underwent 123I-M

Presence but not the timing of onset of REM sleep behavior disorder distinguishes evolution patterns in Parkinson's disease.

BACKGROUND: Rapid eye movement (REM) sleep behavior disorder (RBD) could develop preceding or come after motor symptoms during Parkinson's disease (PD). It remains unknown that whether PD with different timing of RBD onset relative to motor symptoms suggests different spatiotemporal sequence of neurodegeneration. This study aimed to explore the sequence of disease progression in crucially involved brain regions in PD with different timing of RBD onset. METHOD: We recruited 157 PD, 16 isolated RBD (iRBD), and 78 healthy controls. PD patients were identified as (1) PD with RBD preceding motor s

REM sleep without atonia is associated with increased rigidity in patients with mild to moderate Parkinson's disease.

OBJECTIVE: Increased muscle activity during rapid eye movement (REM) sleep (i.e. REM sleep without atonia) is common in people with Parkinson's disease (PD). This study tested the hypotheses that people with PD and REM sleep without atonia (RSWA) would present with more severe and symmetric rigidity compared to individuals with PD without RSWA and age-matched controls. METHODS: Sixty-one individuals participated in this study (41 PD, 20 controls). An overnight sleep study was used to classify participants with PD as having either elevated (PD-RSWA+) or normal muscle activity (PD-RSWA-) during

The combined effect of REM sleep behavior disorder and hyposmia on cognition and motor phenotype in Parkinson's disease.

BACKGROUND: Olfactory dysfunction and REM sleep behavior disorder (RBD) are recognized as pre-motor symptoms of Parkinson's disease (PD). Cognitive dysfunction is observed at a high rate even in the early stages of PD as an important non-motor symptom. PD has been classified in different subtypes and it is unknown if olfactory dysfunction and RBD occur more often in one particular subtype. We investigated the relationship between olfactory impairment, RBD, initial cognitive performance and motor phenotype in PD.
