Six patients with Parkinson's disease (PD) reporting unusually fast or sudden onset of sleep under the addition of dopamine agonists to a previous levodopa-containing therapy were examined using a sleep-wake diary, the Epworth sleepiness scale (ESS), polysomnography, multiple sleep latency tests (MSLT), a standardized vigilance test, and driving simulation. In all patients, ESS scores were increased and polysomnography showed disruption of the sleep pattern, a tendency towards poor sleep efficiency, and reduced proportions of slow- wave and rapid eye movement sleep. Pathological results in the

BACKGROUND: Developmental and Epileptic Encephalopathy with Spike-Wave Activation in Sleep (D/EE-SWAS) is a rare and complex pediatric epilepsy syndrome characterized by cognitive and/or behavioral regression and nearly continuous spike-wave discharges during slow-wave sleep. D/EE-SWAS encompasses a spectrum of encephalopathies with heterogeneous etiologies and outcomes.
OBJECTIVE: To assess etiology, clinical course, and treatment response in 50 children with D/EE-SWAS, with a focus on how underlying causes influence neuropsychological outcomes.
METHODS: We retrospectively analyzed 50 child

Sleep of 11 narcoleptic subjects was recorded on baseline and after 16 and 24 hours of prior wakefulness (16 and 24 hours sleep deprivation). Eleven sex- and age-matched control subjects were recorded for comparisons. All recordings in narcoleptic subjects were characterized by frequent sleep onset rapid eye movement (REM) episodes, increased amounts of wake time after sleep onset and low sleep efficiencies. Mean total sleep time (TST) was significantly decreased in narcoleptic subjects after sleep deprivation (SD). Recovery sleep after 24 hours SD showed reduced nonREM (NREM) sleep stage 2 pe

To determine whether the circadian disruption of the sleep/wake cycle observed in sleeping sickness, human African trypanosomiasis (HAT), can be reversed after trypanosomicide treatment, 10 Congolese patients infected by Trypanosoma brucei gambiense underwent 24-h polysomnographic recordings before treatment with melarsoprol and after each of three weekly treatment sessions. Polysomnography consisted of a continuous recording of the electroencephalogram, electromyogram and electro-oculogram on a Minidix Alvar polygraph. Sleep traces were analysed in 20-sec epochs for wakefulness, REM sleep, an

OBJECTIVE: Sleep disturbances are frequent in Angelman syndrome (AS); however, beside the few studies which have investigated sleep disorders in AS by means of questionnaires, to our knowledge, no systematic polysomnographic recordings have been carried out in AS patients. The present study represents the first attempt to study sleep patterns of AS by polysomnography, to evaluate the influences of sleep on the paroxysmal electroencephalogram (EEG) patterns of AS and to assess the eventual age-related changes of sleep architecture and of sleep EEG abnormalities in children and adolescents with

The regional integrity of brain subcortical structures has been implicated in sleep-wake regulation, however, their associations with sleep parameters remain largely unexplored. Here, we assessed association between quantitative Magnetic Resonance Imaging (qMRI)-derived marker of the myelin content of the brainstem and the variability in the sleep electrophysiology in a large sample of 18-to-31 years healthy young men (N = 321; ~ 22 years). Separate Generalized Additive Model for Location, Scale and Shape (GAMLSS) revealed that sleep onset latency and slow wave energy were significantly associ

Spinocerebellar ataxias (SCA) are autosomal dominant neurodegenerative disorders that affect the cerebellum and its connections, and have a marked clinical and genetic variability. Machado-Joseph disease (MJD) or spinocerebellar ataxia type 3 (SCA3)--MJD/SCA3--is the most common SCA worldwide. MJD/SCA3 is characterized classically by progressive ataxia and variable other motor and non-motor symptoms. Sleep disorders are common, and include rapid eye movement (REM) sleep behaviour disorder (RBD), restless legs syndrome (RLS), insomnia, excessive daytime sleepiness, excessive fragmentary myoclon

OBJECTIVE: To evaluate whether eliciting repetitive cortical and autonomic arousals during sleep is able to induce the occurrence of periodic leg movements during sleep (PLMS).
METHODS: Fifteen normal subjects underwent one night of uninterrupted and two sequential nights of experimental sleep fragmentation achieved by auditory and mechanical stimuli eliciting frequent EEG arousals. Sleep was polygraphically recorded and subsequently used to determine the frequency of arousals and occurrence of leg movement (LM) activity during the first (baseline) and the second fragmentation night. Also, he

Thirty-six narcoleptic patients with overwhelming sleep episodes, cataplexy, and sleep onset REM (SOREM) episodes were recorded for 34 continuous hours in the laboratory starting at 2200 h and ending at 0800 h a day and a half later. There were 94 SOREM and 60 sleep onset NREM (SONREM) episodes. While SONREM episodes were evenly distributed across daytime, SOREM episodes peaked between 0800 and 1000, 1200 and 1400, and 1600 and 1800 h. The ratio of SOREM to SONREM episodes was at its highest level between 1200 and 1400 h. Correlation coefficients between night 1 and night 2 for total sleep tim

Twenth-four-hour patterns of plasma GH, PRL, and cortisol concentrations concomitant with sleep stages were studied in four male patients with typical narcolepsy, aged 30--34 yr, and four normal subjects. All medications were withdrawn 2 weeks before the study. Blood was drawn every 20 min during a 24-h fasting period, except for the first sleep cycle of nocturnal sleep when samples were drawn at 5-min intevals. In all of the narcoleptics, a plasma GH peak associated with slow wave sleep at the nocturnal sleep onset was absent (n = 2) or markedly decreased (n = 2). The normal rise of PRL durin

In contrast to the effects of GHRH in young normal human subjects, in which repetitive i.v. administration of GHRH prompts an increase in the amount of slow wave sleep (SWS) and in GH secretion and blunting of cortisol release, both in young and in old patients with depression there is no effect on SWS and cortisol release after GHRH, while GH secretion is stimulated. We assume that HPA activity and SWS are inert to the influence of GHRH during acute depression because of a slight CRH overactivity, whereas GHRH exerts effects on GH secretion.

OBJECTIVE: Sleep restriction in humans increases risk for obesity, but previous rodent studies show weight loss following sleep deprivation, possibly due to stressful methods used to prevent sleep. Obesity-resistant (OR) rats exhibit consolidated-sleep and resistance to weight gain. It was hypothesized that sleep disruption by a less-stressful method would increase body weight, and the effect of partial sleep deprivation (PSD) on body weight in OR and Sprague-Dawley (SD) rats was examined.
DESIGN AND METHODS: OR and SD rats (n = 12/group) were implanted with transmitters to record sleep/wake.

AIM: Toxoplasma gondii (Tg) is an intracellular parasite infecting more than a third of the human population. Yet, the impact of Tg infection on sleep, a highly sensitive index of brain functions, remains unknown. We designed an experimental mouse model of chronic Tg infection to assess the effects on sleep-wake states.
METHODS: Mice were infected using cysts of the type II Prugniaud strain. We performed chronic sleep-wake recordings and monitoring as well as EEG power spectral density analysis in order to assess the quantitative and qualitative changes of sleep-wake states. Pharmacological a

Both partial and total sleep deprivation frequently result in a rebound of paradoxical sleep (PS), as well as of slow-wave or delta sleep. Acute administration of ethanol inhibits PS in normal volunteers. This effect is dose-dependent and consists of increased latency to and reduced duration of paradoxical sleep. It has also been shown that PS rebound may occur on the same night, as blood alcohol concentration (BAC) declines. The present study examined the effects of sleep deprivation prior to ethanol administration on nocturnal-sleep parameters in healthy male volunteers. Polysomnograms were

AIM: Disrupted sleep may be a prodromal symptom or a predictor of depressive disorders. In this study we investigated the relationship between depression symptoms and disrupted sleep using a novel model of stress-mimicked sleep disorders in rats.
METHODS: SD rats were injected with corticosterone (10, 20 or 40 mg/kg, sc) or vehicle for 7 d. Their sleep-wake behavior was monitored through implanted EEG and EMG electrodes. Their depressive behaviors were assessed using forced swim test, open field test and sucrose preference test.
RESULTS: The corticosterone-treated rats showed significantly r

Sleep is both a state of disconnection from the environment and a critical period for restoration. But how does sleep balance responsiveness with the protection of key functions? The infraslow fluctuation of sigma power (ISFS)-the clustering of sleep spindles over 10-100 s-is thought to regulate this trade-off in rodents by alternating between fragile periods of high arousability and protected periods of spindle-rich stability. However, the organization of arousal and memory reactivation markers within the human ISFS and its conservation in younger ages remain unclear. This study characterizes

We studied the arousal and ventilatory responses to hypoxia during sleep in three trained dogs, before and 1-4 wk after carotid body denervation (CBD). During the studies the dogs breathed through a cuffed endotracheal tube inserted via a chronic tracheostomy. Eucapnic progressive hypoxia was induced by a rebreathing technique, and arterial O2 saturation (Sao2) was measured with an ear oximeter. Sleep stage was determined by electroencephalographic and behavioral criteria. Following CBD, all dogs exhibited hypoventilation under resting conditions; hypoxic ventilatory responses during wakefulne

Obstructive sleep apnea (OSA) patients show highly fragmented sleep with frequent stage transitions, challenging existing automatic sleep staging in feature extraction, multimodal correlation modeling, and interpretability. Current methods rely on fixed-threshold denoising and standard positional encoding, suffering from obvious module stacking and a lack of design tailored to OSA's non-stationary signals. To address these issues, this paper proposes WaveCrossNet, an enhanced Transformer-based model for OSA sleep staging. Instead of simple module stacking, WaveCrossNet embeds adaptive wavelet

The author studied the effect of diazepam in doses of 1 and 3 mg/kg on rats with a chronic cortical cobalt-gelatin focus and implanted cortical and subcortical electrodes. Focal spike activity localized at the site of the focus and hypersynchronous generalized episodes of spikes (and waves) of 8--9/sec frequency were studied in the electroencephalogram and the main phases of vigilance (waking, telencephalic slow waves/SWS/and REM sleep) after diazepam were evaluated. The effect of diazepam on rats temporarily immobilized with tubocurarine was also evaluated. 1. Focal spike activity during slee
