Arthroscopic treatment of meniscal cysts in the horse.

REASONS FOR PERFORMING STUDY: To describe the clinical symptoms, treatment, and outcome of meniscal cysts in horses. These structures have not been previously described in the literature as a potential cause of lameness in the horse. HYPOTHESIS: Meniscal cysts are an uncommon condition of the femorotibial joint but can be a significant cause of lameness. Symptoms can be resolved by arthroscopic excision. METHODS: Records of horses diagnosed with meniscal cysts and treated by cyst excision and meniscal debridement at 2 surgical practices were reviewed. Clinical outcome was determined by repea

Arthroscopic reattachment of osteochondritis dissecans cartilage flaps of the femoropatellar joint: long-term results.

REASONS FOR PERFORMING STUDY: Long-term efficacy of arthroscopic cartilage reattachment for the treatment of osteochondritis dissecans (OCD) lesions in the equine femoropatellar joint is unknown. OBJECTIVE: To evaluate radiographic outcome and long-term performance of horses undergoing OCD reattachment. HYPOTHESIS: Separated OCD cartilage flaps may be reincorporated into the joint surface by reattachment rather than flap removal. METHODS: Polydioxanone pins were utilised arthroscopically to reattach OCD lesions in 40 of 44 joints from 27 horses. Cartilage was reattached when it had persisti

Treatment of osteoarthritis with infrapatellar fat pad derived mesenchymal stem cells in Rabbit.

Osteoarthritis (OA) is a progressively debilitating disease that affects mostly cartilage, with associated changes in the bone. Increasing incidence of OA and the aging population coupled with insufficient therapeutic choices has led to focus on the potential of stem cells as a novel strategy for cartilage repair. In this study, we used scaffold free mesenchymal stem cells obtained from infrapatellar fat pad in an experimental animal model of OA by direct intraarticular injection. Mesenchymal stem cells isolated from a 2.8kg White New Zealand rabbit. The cells were expanded and grown in vitro.

Factors influencing the oxygen concentration gradient from the synovial surface of articular cartilage to the cartilage-bone interface: a modeling study.

OBJECTIVE: There is very little information on the gradients of oxygen concentration from the synovial surface to the subchondral bone in articular cartilage. Cartilage is usually regarded as hypoxic, even though cellular metabolism is inhibited at low oxygen concentrations. We therefore measured rates of cellular consumption of oxygen and used these rates to calculate oxygen tension profiles across articular cartilage. METHODS: The rate of oxygen consumption by bovine articular chondrocytes was measured in vitro, either in intact cartilage slices or in isolated chondrocytes. The oxygen tensi

Three-dimensional T1rho-weighted MRI at 1.5 Tesla.

PURPOSE: To design and implement a magnetic resonance imaging (MRI) pulse sequence capable of performing three-dimensional T(1rho)-weighted MRI on a 1.5-T clinical scanner, and determine the optimal sequence parameters, both theoretically and experimentally, so that the energy deposition by the radiofrequency pulses in the sequence, measured as the specific absorption rate (SAR), does not exceed safety guidelines for imaging human subjects. MATERIALS AND METHODS: A three-pulse cluster was pre-encoded to a three-dimensional gradient-echo imaging sequence to create a three-dimensional, T(1rho)-

Equine upper limb kinematics: Comparison between biplanar high-speed fluoroscopic kinematography and optoelectronic motion capture in one Mini Shetland Pony.

BACKGROUND: There are no studies describing the kinematics of the proximal limb skeleton in living equids. In horses like in all other theri … ived from FluoKin data always showed higher values. This was most pronounced in the elbow joint and stifle with a deviation between both systems of up to 33.0 and 28.1°, respectively. CONCLUSION: First insights into the detailed skeletal movements of a pony's proximal limb during walking and trotting have been obtained.

A single intra-articular injection with IL-4 plus IL-10 ameliorates blood-induced cartilage degeneration in haemophilic mice.

The combination of interleukin (IL)-4 and IL-10 protects against blood-induced cartilage damage in vitro. It has been hypothesized that the combination of these cytokines is effective if applied early in the process of cartilage damage. The present study investigated whether a single intra-articular injection of IL-4 plus IL-10 immediately after a joint bleed limits cartilage damage in an in vivo haemophilia mouse model of blood-induced joint damage. Factor VIII knockout mice with severe haemophilia A were punctured once with a needle below the patella to induce a joint haemorrhage. Subsequent

A worsening crisis: community reflections on violence against women during COVID-19 lockdowns.

The COVID-19 lockdown restrictions, while necessary to curb the spread of the virus, also led to the increase in gender-based violence (GBV) in many countries, including South Africa. Despite numerous studies on GBV, the voices of local community members, as key informants with situated knowledge, have largely been excluded. This study therefore explored community members' accounts of GBV during the COVID-19 lockdowns. Participants included men and women from an informal settlement in Johannesburg South. Three focus group discussions, each consisting of 12-13 participants, were undertaken to c

Anti-tumor activity of CDYL2b in prostate cancer.

Prostate cancer is a leading cause of death among men, yet the molecular underpinnings of this malignancy are still not fully understood. We discovered that two histone demethylases driving prostate tumorigenesis, the JMJD2A and JMJD2B enzymes, suppressed transcription of the CDYL2 epigenetic reader gene. Bioinformatic analyses showed that low CDYL2 expression in prostate tumors was associated with more metastasis and disease recurrence as well as reduced survival. Out of the four predicted CDYL2 isoforms, all of which were capable of forming homo- and heteromers, only CDYL2b was appreciably e

The need to incorporate polysubstance use in neuropsychopharmacology research: Biological lessons and new opportunities.

Critical evaluation and refinement of animal models is essential for neuroscientists to understand complex physiological and pathological processes related to psychiatric diseases. In general, preclinical studies modeling drug dependence and problematic substance use have been limited to the administration of a single substance; however, there is a growing appreciation that this approach has failed to capture the complexities of humans and has stifled translational efforts. Polysubstance use represents the overwhelmingly common patterns of alcohol and drug use in humans. For example, epidemiol…

Evaluating the outcome after center of rotation of angulation (CORA)-based leveling osteotomy (CBLO) technique to repair unilateral cranial cruciate ligament deficiency using a pressure-sensitive walkway system.

The objective of this study was to evaluate the mid-term outcome of center of rotation of angulation (CORA)-based leveling osteotomy (CBLO) in dogs with cranial cruciate ligament (CrCL) deficiency using a pressure-sensitive walkway system. Fifteen dogs with unilateral CrCL deficiency were treated with CBLO for an average of 50 wk (median 46 wk, range: 38 to 91) prior to the evaluation. The contralateral hind limb was confirmed free of any signs of pathology by clinical examination and lateral stifle radiography. A control group of 20 healthy dogs was included to establish reference values for …

A transition point for the blood flow wall shear stress environment in the human fetal left ventricle during early gestation.

Development of the fetal heart is a fascinating process that involves a tremendous amount of growth. Here, we performed image-based flow simulations of 3 human fetal left ventricles (LV), and investigated the hypothetical scenario where the sizes of the hearts are scaled down, leading to reduced Reynolds number, to emulate earlier fetal stages. The shape and motion of the LV were retained over the scaling to isolate and understand the effects of length scaling on its fluid dynamics. We observed an interesting cut-off point in Reynolds number (Re), across which the dependency of LV wall shear s
