Perception of apparent motion of colored stimuli after commissurotomy.

One subject with complete forebrain commissurotomy (L.B.), another with posterior callosotomy (D.K.), and eight normal controls were presented with successive pairs of red and/or green lights, on either side of the retinal midline and within the left and right visual fields. All of the subjects could discriminate the direction of apparent motion in all three locations, although L.B. was poorer on bilateral than unilateral presentations. Moreover, on bilateral presentations, L.B. was significantly poorer at identifying the color of the first light than the color of the second light. In contrast

Postembryonic Fish Brain Proliferation Zones Exhibit Neuroepithelial-Type Gene Expression Profile.

In mammals, neuroepithelial cells play an essential role in embryonic neurogenesis, whereas glial stem cells are the principal source of neurons at postembryonic stages. By contrast, neuroepithelial-like stem/progenitor (NE) cells have been shown to be present throughout life in teleosts. We used three-dimensional (3D) reconstructions of cleared transgenic wdr12:GFP medaka brains to demonstrate that this cell type is widespread in juvenile and to identify new regions containing NE cells. We established the gene expression profile of optic tectum (OT) NE cells by cell sorting followed by RNA-se

Chronic high-frequency deep brain stimulation of the STN/SNr for progressive myoclonic epilepsy.

Chronic high-frequency deep brain stimulation (DBS) may also be effective in patients with refractory epilepsy. A possible benefit has been postulated because of the connections that exist between the subthalamic nucleus (STN) and the superior colliculus. Individual case reports and pilot studies of successful DBS in different types of epilepsy have already been presented. Here, the case of a 39-year-old male with progressive myoclonic epilepsy is reported who remained severely impaired despite VNS and combined antiepileptic drug therapy. Bilateral DBS electrodes were implanted into the STN, f

The L1 cell adhesion molecule is essential for topographic mapping of retinal axons.

The retinocollicular projection is a preferred axon guidance pathway for investigating molecular mechanisms of synaptic targeting in the mammalian CNS. Here we identify a previously unrecognized role of the L1 cell adhesion molecule in topographic mapping of retinal ganglion cell (RGC) axons to their targets in the mouse superior colliculus (SC). L1 was transiently expressed on RGC axons during axon growth and targeting. DiI labeling of retinal axons revealed that temporal axons of L1-minus mice bypassed correct target locations in the anterior SC, forming termination zones at incorrect poster

Optic nerve hypoplasia in an acute exposure model of the fetal alcohol syndrome.

The effects of acute prenatal exposure to ethanol on the postnatal optic nerve have been examined in a C57B1/6J mouse model. Pregnant mice were exposed by intraperitoneal injection to ethanol (25% ethanol, each dose at 0.015 ml/g separated by 4 h), or to saline on the 8th gestational day and the optic nerve examined at P15. There was a significant difference in the cross-sectional areas of optic nerves from ethanol-exposed mice (Mean +/- SD: 32,800 +/- 11,000 microns2) compared to control nerves (Mean +/- SD: 52,100 +/- 8,900 microns2). This difference was mainly the result of a reduction in t

Trying to understand axonal regeneration in the CNS of fish.

In contrast to the situation in mammals and birds, neurons in the central nervous system (CNS) of fish--such as the retinal ganglion cells--are capable of regenerating their axons and restoring vision. Special properties of the glial cells and the neurons of the fish visual pathway appear to contribute to the success of axonal regeneration. The fish oligodendrocytes lack the axon growth inhibiting molecules that interfere with axonal extension in mammals. Instead, fish optic nerve oligodendrocytes support--at least in vitro--axonal elongation of fish as well as that of rat retinal axons. Moreo

An optimized method for retrograde labelling and quantification of rabbit retinal ganglion cells.

Rabbits are a commonly used animal model in glaucoma research, but their application has been limited by the techniques used to assess optic nerve injury (ONI). Our study devised an optimized method for retrograde labelling and analysing rabbit retinal ganglion cells (RGCs). This method involved improvements over the conventional method regarding the stereotaxic device, the positioning of superior colliculi, the target of axonal tracer delivery, and the visualization and analysis of labelled RGCs. To evaluate its efficacy, eight New Zealand White rabbits were divided into naïve and ONI groups.…

Coordination of distinct sources of excitatory inputs enhances motion selectivity in the mouse visual thalamus.

Multiple sources innervate the visual thalamus to influence image-forming vision prior to the cortex, yet coordination between non-retinal and retinal inputs in shaping thalamic visual selectivity remains unclear. Using dual-color two-photon calcium imaging in the thalamus of awake mice, we observed a higher fraction of direction-selective boutons among input from superior colliculus neurons than from retinal ganglion cells, both providing strong converging excitatory input to thalamic neurons. Collicular and retinal axons exhibit retinotopic organization with similar precision. At a fine scal

Interretinal transduction of injury signals after unilateral optic nerve crush.

In this study, we report that partial unilateral optic nerve crush in the rat affects the number of retinal ganglion cells of the contralateral eye still in continuity with the ipsilateral superior colliculus. The reduction in cell number of the uncrossed retinal projection was accompanied by a microglia response and could be prevented by the local intravitreal application of the anti-inflammatory agent dexamethasone. Interestingly, the level of neuronal activity after optic nerve crush as evidenced by thallium autometallography was enhanced in the termination area of the uncrossed projection,

Progressive supranuclear palsy and corticobasal degeneration: lumping versus splitting.

Progressive supranuclear palsy (PSP) and corticobasal degeneration (CBD) are both sporadic disorders with tau pathology. Criteria have been defined that in most instances allow for adequate diagnosis of the two disorders both clinically and neuropathologically; however, overlap is not uncommon. For example, patients with PSP may present with severe unilateral apraxia and supranuclear gaze palsy can occur in CBD. Pathological overlap also occurs and pathologically "mixed" cases are encountered. Common to both these two tauopathies is that isoforms of four-repeat tau due to splicing of exon 10 d

Retrograde axonal transport of BDNF in retinal ganglion cells is blocked by acute IOP elevation in rats.

PURPOSE: To determine whether acute experimental glaucoma in rats obstructs retrograde transport of brain-derived neurotrophic factor (BDNF) to retinal ganglion cells (RGCs). METHODS: Forty rats had unilateral injection of either (125)I-BDNF (20 animals) or a mixture of (125)I-BDNF and 100-fold excess nonradiolabeled BDNF (20 animals). In each group of 20 animals, eyes contralateral to injection had either normal intraocular pressure (IOP; 10 animals) or IOP elevated to 25 mm Hg below the systolic blood pressure of the eye (10 animals). In each group of 20 rats, ipsilateral eyes had IOP set a

The levels of retinal mRNA for gefiltin, a neuronal intermediate filament protein, are regulated by the tectum during optic fiber regeneration in the goldfish.

Reorganization of the intermediate filament (IF) network during axonal regeneration is accompanied by changes in the expression of various IF proteins. An increase in expression of the neuronal IF subunit gefiltin in goldfish retinal ganglion cells (RGCs) has been linked to the unique ability of the goldfish optic nerve to regenerate following injury. Evidence suggests that the optic tectum, the target of optic fibers, may regulate the expression of gefiltin during regeneration. To address this issue we examined gefiltin mRNA levels during optic fiber regeneration in the presence or absence of

Evidence for the involvement of serotonin in the antinociception induced by electrical or chemical stimulation of the mesencephalic tectum.

A great deal of evidence has shown that electrical stimulation or microinjections of GABAA blockers, such as bicuculline, into the midbrain tectum (MT) produce escape behavior, which has been associated to fear. This study was aimed to examine the characteristics of the analgesia that follows the escape behavior induced by electrical (freezing and escape thresholds) and chemical (bicuculline microinjections) stimulation of the midbrain tectum. Immediately after the expression of the aversive responses the rats were submitted to the tail-flick test. The obtained results show that analgesia alwa

Autoradiographic studies with 3H dihydrotestosterone in the brain of sex reversed mice, heterozygous for androgen insensitive testicular feminization (Tfm). A comparison with normal female mice and sex reversed male mice.

Specific binding sites for 3H dihydrotestosterone are demonstrated by autoradiography in brain nuclei of sex reversed mice heterozygous for testicular feminization (Tfm) which are phenotypically intersexes with testes and accessory sex glands that consist of a mosaic of androgen insensitive Tfm cells which lack specific dihydrotestosterone binding and androgen sensitive normal cells. The nuclear group evaluated include: nucleus (n.) septi lateralis, n. interstitialis striae terminalis, n. medialis amygdalae, the hypothalamic n. arcuatus, n. ventromedialis lateralis, n. pre-mammillaris ventrali

Depression and enhancement of baroreceptor pressor response in cats after intracerebroventricular injection of noradrenergic blocking agents: dependence on supracollicular areas of the brain.

The alpha-adrenergic blocking drugs, phentolamine and Hydergine, both act centrally at different sites to depress and enhance the pressor and sympathetic nerve response to decreased baroreceptor afferent input in anesthetized cats. Depression of the rise in blood pressure and sympathetic nerve discharge during bilateral carotid occlusion (BCO) followed injection of the agents into the 4th cerebral ventricle when the brain was intact but not when connections were interrupted at the midcollicular level by transection or lesion. Enhancement of responses occurred when drug distribution was confine

Intrinsically motivated action-outcome learning and goal-based action recall: a system-level bio-constrained computational model.

Reinforcement (trial-and-error) learning in animals is driven by a multitude of processes. Most animals have evolved several sophisticated systems of 'extrinsic motivations' (EMs) that guide them to acquire behaviours allowing them to maintain their bodies, defend against threat, and reproduce. Animals have also evolved various systems of 'intrinsic motivations' (IMs) that allow them to acquire actions in the absence of extrinsic rewards. These actions are used later to pursue such rewards when they become available. Intrinsic motivations have been studied in Psychology for many decades and th

Visual FMRI responses in human superior colliculus show a temporal-nasal asymmetry that is absent in lateral geniculate and visual cortex.

Eye patching has revealed enhanced saccadic latencies or attention effects when orienting toward visual stimuli presented in the temporal versus nasal hemifields of humans. Such behavioral advantages have been tentatively proposed to reflect possible temporal-nasal differences in the retinotectal pathway to the superior colliculus, rather than in the retinogeniculate pathway or visual cortex. However, this has not been directly tested with physiological measures in humans. Here, we examined responses of the human superior colliculus (SC) to contralateral visual field stimulation, using high sp

A GFP-based genetic screen reveals mutations that disrupt the architecture of the zebrafish retinotectal projection.

The retinotectal projection is a premier model system for the investigation of molecular mechanisms that underlie axon pathfinding and map formation. Other important features, such as the laminar targeting of retinal axons, the control of axon fasciculation and the intrinsic organization of the tectal neuropil, have been less accessible to investigation. In order to visualize these processes in vivo, we generated a transgenic zebrafish line expressing membrane-targeted GFP under control of the brn3c promoter/enhancer. The GFP reporter labels a distinct subset of retinal ganglion cells (RGCs),
