The seasonal characteristic features of the efficacy of antioxidants in tetracycline affections of the liver were revealed in the experiments with 182 noninbred albino male rats. It was shown that sodium selenite had the most pronounced hepatoprotective effect in autumn and winter, while vitamin E was most effective in spring and summer. A rapid assay for tetracyclines in premixes and mixed feeds is described, which uses the extraction and dilution systems of AOAC methods, and a strain of Bacillus stearothermophilus ATCC 12980 selected to grow at 40 degrees C. The study comprised 49 patients suffering from acne who were treated over a period averaging 4.7 +/- 1.2 months with tetracyclines administered either orally (1000 mg daily for 5 days, then 250 mg daily) or locally (2% alcoholic solution). On the basis of a comparison of different methods used to determine the concentration of active lymecycline in Fallopian tube tissue, a method was adopted which allowed the determination of lymecycline in serosa and mucosa tissue of the Fallopian tube in 16 women undergoing hysterectomy. A difference in the concentration of active drug was found in serosa (greater than 1.7 mg/kg tissue) and mucosa (greater than 0.8 mg/kg tissue) in 16 patients on lymecycline therapy for 24 hours. A 60-year-old woman with recurrent papular and vesiculobullous lesions of erythema elevatum diutinum responded to treatment with 100 mg of oral niacinamide three times a day and 250 mg of tetracycline hydrochloride four times a day. Prior therapy with 2.5 mg of dexamethasone daily and 1.0 g of erythromycin daily had been unsuccessful. After four weeks, tetracycline therapy was discontinued, and niacinamide alone sufficed to suppress the disorder. Recurrent lesions developed whenever niacinamide therapy was stopped.

The influence of certain compounds on intestinal absorption of tetracycline was tested, using two different in vivo techniques: a) Disappearence of the drug from perfused intestinal segments of the rabbit; b) Measuring urinary excretion in the rat after oral dosage. Ca2+ and Fe2+ ions both reduced the permeation rate of tetracycline by about 30%. In presence of Ca2+ and salicylate ions, leading to ion-pair formation with the antibiotic, intestinal uptake was significantly increased in comparison to experiments where only calcium was present. Forty-two adults who had Shigella isolated from stool (26 symptomatic, 16 asymptomatic) received a single oral dose of tetracycline hydrochloride (2.5 g). Minimal inhibitory concentrations of the 42 isolates demonstrated that 41% were sensitive to tetracycline. Sixteen of 18 patients with diarrhea who had tetracycline-resistant Shigella and all eight patients with diarrhea who had tetracycline-sensitive Shigella were clinical well and had Shigella-negative stools 48 hours after therapy. Fifteen of 16 asymptomatic patients demonstrated clearing of Shigella from stool within 48 hours of therapy.
