Monitoring chemical exposure using breast milk: a methodological review.

Retrospective exposure assessments are often a major weakness in environmental epidemiologic studies. Many environmentally important chemicals are persistent lipophilic compounds which partition into fat and breast milk. The analysis of these chemicals in breast milk provides a noninvasive means of collecting large volumes of a biologic fluid that correlates well with body burden. This may provide an integrated measure of cumulative absorption over several years which can be used to assess exposure in several different epidemiologic study designs. However, consideration of toxicokinetic princi

Biological valuation of extra-corporeal techniques in acute poisoning.

The efficiency of dialysis methods a/o hemoperfusion in acute poisoning cannot be clinically estimated, because: a) Concomitant intestinal absorption, hepatic metabolism and urinary excretion must be taken into account. b) With supportive treatment alone, spontaneous recovery usually occurs in 98% of the intoxications in Intensive Care Units. The efficiency of these methods can only be estimated biologically. Measuring the blood level at the beginning and the end of the treatment as well as measuring the clearances of the drug is misleading. A better method is to measure the amount of extracte

Placental transfer of thiamphenicol in the rat.

Measurement of thiamphenicol transfer to the rat embryo during organogenesis was performed as one step of the determination of possible drug embryotoxicity in man. Extrapolation of data on animal embryotoxicity to man will only become possible when data on the toxicokinetic properties of the substance under investigation are available for both animal and man. 1) Thiamphenicol, given between day 11.5 and 14 of rat gestation, rapidly reached the embryo; 4--6 h after single i.v. or s.c. injection, embryonic and maternal thiamphenicol levels became equal and decreased from that time on at the same

Food additives, contaminants and other minor components: effects on human gut microbiota-a review.

Gut bacteria play an important role in several metabolic processes and human diseases, such as obesity and accompanying co-morbidities, such as fatty liver disease, insulin resistance/diabetes, and cardiovascular events. Among other factors, dietary patterns, probiotics, prebiotics, synbiotics, antibiotics, and non-dietary factors, such as stress, age, exercise, and climatic conditions, can dramatically impact the human gut microbiota equilibrium and diversity. However, the effect of minor food constituents, including food additives and trace contaminants, on human gut microbiota has received

Effect of various exposure scenarios on the biological monitoring of organic solvents in alveolar air. II. 1,1,1-Trichloroethane and trichloroethylene.

The purpose of the present study was to investigate the influence of different exposure scenarios on the elimination of trichloroethylene (TRI) and 1,1,1-trichloroethane (1,1,1-TRI) in alveolar air and other biological fluids in human volunteers. In addition, it was sought to establish an interactive process between experimental data gathering and simulation modeling in an attempt to predict the influence of the different scenarios of exposure to TRI and 1,1,1-TRI on their respective biological monitoring indices and thus to establish the flexibility and validity of simulation models. Multistate models of developmental toxicity: Application to valproic acid-induced malformations in the zebrafish embryo.

For the determination of acute toxicity of chemicals in zebrafish (Danio rerio) embryos, the OECD test guideline 236, relative to the Fish Embryo Toxicity Test (FET), stipulates a dose-response analysis of four lethal core endpoints and a quantitative characterization of abnormalities including their time-dependency. Routinely, the data are analyzed at the different observation times separately. However, observations at a given time strongly depend on the previous effects and should be analyzed jointly with them. To solve this problem, we developed multistate models for occurrence of developme

Curcumin-based nanotherapeutics for colibacillosis in chickens: advances in veterinary microbiology.

One of the most important bacterial agents of colibacillosis is Avian Pathogenic Escherichia coli (APEC), which affects the poultry industry by increasing mortality, reducing growth performance, and ultimately decreasing production efficiency. According the surveillance studies carried out in the European Union during the period of 2019-21, resistance to antibiotics has increased in E. coli due to transfer of genes between bacteria and humans. The rapid emergence of high antimicrobial resistance has accelerated research towards effective, natural, and antibiotic-free substitutes. Curcumin is a

Aluminum and Alzheimer's disease in the 21st century: An umbrella review of evidence and controversies.

The potential role of aluminum (Al) in the onset and progression of Alzheimer disease (AD) remains among the most enduring and divisive controversies in environmental neurotoxicology. Although numerous narrative reviews, systematic reviews, meta-analyses, mechanistic appraisals, and toxicological assessments have been published, their conclusions remain heterogeneous and at times contradictory. The present review of reviews critically integrates the PubMed-indexed review literature published in this century (January 1, 2000, and May 2, 2026), focusing on epidemiology, exposure sources, toxicok

Assessing the accumulation of microplastics in earthworms (Eisenia fetida) using traditional bioaccumulation modeling and synchrotron-based microcomputed tomography.

Microplastics (MPs) are recognized as a ubiquitous contaminant in terrestrial ecosystems. Earthworms, which play key roles in soil structure, nutrient cycling, and ecosystem function, are important bioindicators for assessing MP exposure and fate. However, uncertainties remain regarding MP toxicokinetics in earthworms, including ingestion dynamics, retention, and the potential for translocation across the gut epithelium. This study investigated the movement and fate of MPs in Eisenia fetida by quantifying uptake and elimination across five common MP morphologies (films, fibers, foams, fragment

Combination of Multiple-Dose Activated Charcoal and Infusion Therapy That Led to Favorable Outcomes in Colchicine Poisoning: A Case Report.

BACKGROUND The therapeutic margin of colchicine is narrow, and toxicity occurs easily. No standard treatment exists because its toxicokinetics are poorly understood. Symptoms are usually gastrointestinal, and dehydration occurs easily. Few reports have monitored serum and urinary colchicine levels. CASE REPORT A man in his 20s presented with nausea, vomiting, diarrhea, drowsiness, and dyspnea. He ingested 2 dried Colchicum autumnale bulbs (estimated colchicine: 6.24-15.6 mg) with an energy drink. Activated charcoal with laxative was administered at 27 hours post ingestion (h-PI), followed by m

Ecotoxicological implications of environmental neurotoxin β-N-methylamino-L-alanine (BMAA) in fishes: An emerging concern.

Harmful algal blooms (HABs), intensified by climate change, eutrophication, and altered hydrological regimes, are expanding globally, releasing cyanotoxins that threaten aquatic ecosystems and human health. β-N-methylamino-L-alanine (BMAA), a non-protein amino acid with neurotoxic potential, has been recognized as a global emerging concern. Following exposure, BMAA is present in both free and protein-bound forms, forming an endogenous toxin reservoir that exacerbates potential neurotoxicity in aquatic organisms and humans. Its presence in aquatic food webs not only elevates ecological risks fo

Comparative analysis of Caco-2 cells and human jejunal and duodenal enteroid-derived cells in gel- and membrane-based barrier models of intestinal permeability.

Intestinal absorption is a key toxicokinetics parameter. Although the colon carcinoma cell line Caco-2 is the most used in vitro model to estimate human drug absorption, models representing other intestinal segments are available. We characterized the morphology, tissue-specific markers, and functionality of 3 human intestinal cell types: Caco-2, primary human enteroid-derived cells from jejunum (J2), and duodenum (D109) when cultured in the OrganoPlate 3-lane 40 microphysiological system (MPS) or static 24-well Transwells. In both conditions, J2 and D109 formed dome-like structures; Caco-2 fo

Clinical spectrum of wild (mad) honey poisoning: a case series from a tertiary care center in Nepal.

INTRODUCTION AND IMPORTANCE: Wild (mad) honey poisoning is caused by grayanotoxins, naturally occurring neurotoxins found in the nectar of certain Rhododendron species native to mountainous regions of Nepal, India, Bhutan, and Turkey. While typically associated with hypotension and bradycardia, wild honey ingestion may also lead to anaphylactic reactions. This case series presents ten patients who developed varying degrees of toxicity following the ingestion of wild honey. CASE PRESENTATION: We describe ten cases that presented to the emergency department with symptoms following wild (mad) ho

Human exposure and toxicity of per- and polyfluoroalkyl substances: Narrative review and perspectives.

In this paper, the status of per- and polyfluoroalkyl substances (PFAS) is narratively reviewed from the perspective of toxicity mechanisms, biodistribution, species differences, human exposure sources, human biomonitoring, and epidemiology. Although toxicological investigations have been conducted, interspecies differences in toxicological mechanisms and toxicokinetics are not fully understood. For example, peroxisome proliferator-activated receptors (PPARα) are a target receptor of PFAS. Although there are distinct differences in response between rodents and humans, human PPARα can be activa

Integrated transcriptomics and metabolomics reveal the mechanism of intestinal damage upon acute patulin exposure in mice.

Mycotoxin contamination has become a major food safety issue and greatly threatens human and animal health. Patulin (PAT), a common mycotoxin in the environment, is exposed through the food chain and damages the gastrointestinal tract. However, its mechanism of enterotoxicity at the genetic and metabolic levels remains to be elucidated. Herein, the intestinal histopathological and biochemical indices, transcriptome, and metabolome of C57BL/6 J mice exposed to different doses of PAT were successively assessed, as well as the toxicokinetics of PAT in vivo. The results showed that acute PAT expos

Exploring the dynamics of the ABCB1 membrane transporter P-glycoprotein in the presence of ATP and active/non-active compounds through molecular dynamics simulations.

P-glycoprotein (Pgp) is a member of the ATP-binding cassette family of transporters that confers multidrug resistance to cancer cells and is actively involved in the pharmacokinetics and toxicokinetics of a big variety of drugs. Extensive studies have provided insights into the binding of many compounds, but the precise mechanism of translocation across the membrane remains unknown; in this context, the major challenge has been to understand the basis for its polyspecificity. In this study, molecular dynamics (MD) simulations of human P-gp (hP-gp) in an explicit membrane-and-water environment

First evidence that emerging pinnatoxin-G, a contaminant of shellfish, reaches the brain and crosses the placental barrier.

Massive proliferation of some toxic marine dinoflagellates is responsible for the occurrence of harmful algal blooms and the contamination of fish and shellfish worldwide. Pinnatoxins (PnTx) (A-H) comprise an emerging phycotoxin family belonging to the cyclic imine toxin group. Interest has been focused on these lipophilic, fast-acting and highly potent toxins because they are widely found in contaminated shellfish, and can represent a risk for seafood consumers. PnTx display a potent antagonist effect on nicotinic acetylcholine receptors (nAChR), and in this study we assessed in vivo the abil

Maternal-fetal disposition of domoic acid following repeated oral dosing during pregnancy in nonhuman primate.

Domoic acid (DA) is a marine algal toxin that causes acute and chronic neurotoxicity in animals and humans. Prenatal exposure to DA has been associated with neuronal damage and cognitive and behavioral deficits in juvenile California sea lions, cynomolgus monkeys and rodents. Yet, the toxicokinetics (TK) of DA during pregnancy and the maternal-fetal disposition of DA have not been fully elucidated. In this study, we investigated the TK before, during, and after pregnancy and the maternal-fetal disposition of DA in 22 cynomolgus monkeys following daily oral doses of 0.075 or 0.15 mg/kg/day of D

Chimeric Mice With Humanized Livers Demonstrate Human-Specific Hepatotoxicity Caused by a Therapeutic Antibody Against TRAIL-Receptor 2/Death Receptor 5.

The activation of tumor necrosis factor (TNF)-related apoptosis-inducing ligand receptor 2 (TRAIL-R2)/death receptor 5 (DR5) induces apoptosis in various tumor cells but not in normal human cells. Because some therapeutic antibodies targeting TRAIL-R2 have demonstrated severe hepatotoxicity in clinical applications, novel in vivo models reflecting clinical hepatotoxicity are now required. In this study, we investigated the hepatotoxicity caused by KMTR2, an anti-human TRAIL-R2 monoclonal antibody, in chimeric mice with humanized livers (PXB-mice). PXB-mice were exposed to KMTR2 by single or re
