In rat self-filling blind loops and in the contiguous regions of the intestinal tract, considerable functional impairment, accompanied by pronounced morphological transformations of the mucosa, has been observed. The histological alterations consist of mucosal hypertrophy and a reduction in the villus height: crypt length ratio, which is indicative of a hyper-regenerative change. Various enzyme activities of the epithelial cells are reduced and the absorption otinal juices is greatly altered in favour of the free acids. Reactive oxygen species (ROS) are small oxygen-derived molecules that are used to control infections by phagocytic cells. In macrophages, the oxidative burst produced by the NOX2 NADPH-oxidase is essential to eradicate engulfed pathogens by both oxidative and non-oxidative killing. Indeed, while the superoxide anion ( O2- ) produced by NOX2, and the other ROS derived from its transformation, can directly target pathogens, ROS also contribute to activation of non-oxidative microbicidal effectors. The response of pathogens to the phagocytic oxidative burst includes the expression of different en

Despite the immense potential against diverse chronic inflammatory and metabolic disorders, the clinical translation of microbial therapies is largely thwarted by the delivery challenges of live biotherapeutics. Probiotics often succumb to harsh gastrointestinal conditions and oxidative stress. We report ARMOG, a microenvironment responsive platform achieving in situ biointerfacial transformation. Composed of hyaluronic acid with an increased dopamine grafting ratio cross-linked by arylboronic acid, the matrix undergoes cleavage triggered by the inflammatory microenvironment, leading to locali

Fusobacterium nucleatum tumor infiltration is a hallmark of colorectal cancer (CRC) progression. However, whether and how other microbes influence F. nucleatum in CRC remains unclear. Here, we discover that the commensal fungus Candida albicans synergizes with F. nucleatum to promote CRC pathogenesis and contributes to poor patient outcomes. Co-inoculation of C. albicans and F. nucleatum accelerates CRC progression compared to either microbe alone in animal models. C. albicans strains isolated from CRC patients exhibiting enhanced hyphal morphogenesis and elevated FLO9 expression promote F. nu

Inspired by naturally occurring biomaterials, autonomously grown engineered living materials (ELMs) feature cell-driven growth and programmable biological functions. However, the "livingness" of cells poses a short life span and low tolerance to harsh conditions, limiting the practical use of such materials. Here, we developed materials with programmable and dormant functionalities, grown from a mixture of Komagataeibacter rhaeticus and Bacillus endospores under engineered medium conditions. K. rhaeticus produces the bacterial cellulose (BC) matrix that integrates Bacillus spores within, where

Volatile organic compounds emitted from landfills posed adverse effect on health. In this study, gaseous benzene was biologically treated using an in-situ biofilter without air pump. Its performance was investigated and the removal efficiency of benzene reached over 90 %. The decrease in the average benzene concentration was consistent with first-order reaction kinetics. Mycolicibacterium dominated the bacterial consortium (41-57 %) throughout the degradation. Annotation of genes by metagenomic analysis helped to deduce the degradation pathways (benzene degradation, catechol ortho-cleavage and

Mercury (Hg) and its organic compounds, especially monomethylmercury (MeHg), cause major damage to the ecosystem and human health. In surface water or sediments, microorganisms play a crucial role in the methylation and demethylation of Hg. Given that Hg transformation processes are intracellular reactions, accurate assessment of the bioavailability of Hg(II)/MeHg in the environment, particularly for microorganisms, is of major importance. Compared with traditional analytical methods, bacterial whole-cell biosensors (BWCBs) provide a more accurate, convenient, and cost-effective strategy to as

Small intestinal bacterial overgrowth (SIBO) is a common, yet underrecognized, problem. Its prevalence is unknown because SIBO requires diagnostic testing. Although abdominal bloating, gas, distension, and diarrhea are common symptoms, they do not predict positive diagnosis. Predisposing factors include proton-pump inhibitors, opioids, gastric bypass, colectomy, and dysmotility. Small bowel aspirate/culture with growth of 10-10 cfu/mL is generally accepted as the "best diagnostic method," but it is invasive. Glucose or lactulose breath testing is noninvasive but an indirect method that require

Chlamydia, a major human bacterial pathogen, assumes effective strategies to protect infected cells against death-inducing stimuli, thereby ensuring completion of its developmental cycle. Paired with its capacity to cause extensive host DNA damage, this poses a potential risk of malignant transformation, consistent with circumstantial epidemiological evidence. Here we reveal a dramatic depletion of p53, a tumor suppressor deregulated in many cancers, during Chlamydia infection. Using biochemical approaches and live imaging of individual cells, we demonstrate that p53 diminution requires phosph

The obligate intracellular bacterial pathogen Chlamydia trachomatis (Ctr) has been associated with cervical and ovarian cancer development. However, establishment of causality and the underlying mechanisms remain outstanding. Our analysis of Ctr-induced alterations to global host histone modifications revealed distinct patterns of histone marks during acute and persistent infections. In particular, pH2AX (Ser139) and H3K9me3, hallmarks of DNA double-strand breaks (DSBs) and senescence-associated heterochromatin foci (SAHF), respectively, showed sustained upregulation during Ctr infection. Ctr-

A group of repetitive sequences, known as the Family of Repeats (FR), is a critical cis-acting sequence required for EBV latent infection. The FR sequences are heterogeneous among EBV strains, and they are sometimes subject to partial deletion when subcloned in E. coli-based cloning vectors. However, the FR stability in EBV-BAC (bacterial artificial chromosome) system has never been investigated. We found that the full length FR of the Akata strain EBV was not stably maintained in a BAC vector. By contrast, newly obtained BAC clones of the B95-8 strain of EBV stably maintained the full length

Follow up management in a patient already treated with decompressive craniotomy for a space-occupying endocarditic stroke is difficult. While immediate valve replacement eliminates the focus and therefore the high risk of re-embolization, a neurosurgical intervention is considered a contraindication to early cardiosurgery. Herein, the first report is presented of a critically ill patient with bacterial endocarditis and a space-occupying cerebellar infarction with imminent herniation, who successfully underwent mitral valve replacement only 24 h after decompressive craniotomy. To prevent reblee

The genes encoding the beta- and beta'-subunits of RNA polymerase (rpoB and rpoC respectively) are fused as one continuous open reading frame in Helicobacter pylori and in other members of this genus, but are separate in other bacterial taxonomic groups, including the closely related genus Campylobacter. To test whether this beta-beta' tethering is essential, we used polymerase chain reaction-based cloning to separate the rpoB and rpoC moieties of the H. pylori rpoB-rpoC fusion gene with a non-polar chloramphenicol resistance cassette containing a new translational start, and introduced this c

The rapid spread of antibiotic resistances in a wide variety of bacteria is mainly due to the location of antibiotic resistance genes on mobile genetic elements such as plasmids and transposons. Principal ways of transfer of plasmid- and transposon-encoded resistance genes are presented using examples of the predominant genes mediating resistances to protein biosynthesis inhibitors such as tetracyclines, aminoglycosides, macrolide-lincosamide-streptogramin B antibiotics, and chloramphenicol in staphylococci. Transfer between different staphylococcal cells is substantially based on transduction

The gene for a new bacterial aquaporin, AqpX, was cloned from the pathogenic Gram-negative bacterium Brucella abortus. The gene was mapped on the large chromosome of B. abortus. It is flanked by one upstream and two downstream copies of the Brucella repeated sequence Bru-RS. Prediction from the nucleotide sequence indicated that the protein is a member of the MIP family, which comprises channels for water and/or solute transport. Expression in Xenopus oocytes and cryoelectron microscopy of Escherichia coli cells transformed with the aqpX gene confirmed that the protein is an efficient water ch

BACKGROUND: Polysaccharide depolymerases derived from phages play a pivotal role in the degradation of bacterial surface polysaccharides, offering a promising strategy to address antibiotic-resistant bacterial strains. However, the identification of these enzymes along with their associated host capsular serotypes poses significant challenges. This study introduces DposFinder, a transformer-based deep learning framework designed to predict depolymerases alongside their host capsular serotypes, thereby facilitating large-scale discovery and application of these enzymes. METHODS: We developed Dp

BACKGROUND AND AIMS: Diagnosis code classification is a common method for cohort identification in cirrhosis research, but it is often inaccurate and augmented by labor-intensive chart review. Natural language processing using large language models (LLMs) is a potentially more accurate method. To assess LLMs' potential for cirrhosis cohort identification, we compared code-based versus LLM-based classification with chart review as a "gold standard."
APPROACH AND RESULTS: We extracted and conducted a limited chart review of 3788 discharge summaries of cirrhosis admissions. We engineered zero-sh

Engineered microbial therapies provide a promising strategy for in situ production of therapeutic molecules at disease sites; however, their clinical translation remains limited by insufficient retention at target tissues and the lack of external control after administration. Here, we develop a wearable bioelectronic-assisted living hydrogel platform that enables rapid in situ immobilization and remote regulation of engineered bacteria. Optogenetically engineered bacteria capable of green-light-induced anti-interleukin-6 (anti-IL-6) secretion were encapsulated within a photoresponsive Gelatin

Avian pseudotuberculosis infection usually presents as well-demarcated visceral necrotic foci, typically affecting the gastrointestinal tract, liver and spleen. This case series describes an atypical presentation of Yersinia pseudotuberculosis (Yptb) characterized by severe chronic myositis, arthritis and osteomyelitis in five village weavers (Ploceus cucullatus), and acute osteomyelitis and myositis associated with septicaemia in an oriental magpie robin (Copsychus saularis) from a zoological collection. Clinical signs of the weavers included lethargy, poor flying ability and focally extensiv

Soil-transmitted helminths (hookworms, whipworms, and large roundworms) are agents of intestinal roundworm diseases of poverty that infect upwards of 2 billion people worldwide. A great challenge in treating these diseases is the development of anthelmintic therapeutics that are inexpensive, can be produced in great quantity, and are capable of delivery under varied and adverse environmental conditions. A potential solution to this challenge is the use of live bacteria that are acceptable for human consumption, e.g., Bacillus subtilis, and that can be engineered with therapeutic properties. In
