Engineering microbial platforms for diversified biosynthesis of antidepressant tryptophan-derived neuroactive compounds.

Tryptophan-drived neuroactive compounds, including serotonin, melatonin, 5-hydroxytryptophan (5-HTP), psilocybin, and related indole derivatives, represent an important class of therapeutics for depression and other neuropsychiatric disorders. However, conventional production through chemical synthesis or natural extraction remains limited by low sustainability, poor selectivity, and high manufacturing costs. Advances in synthetic biology, metabolic engineering, and enzyme engineering have enabled the development of microbial cell factories for the efficient biosynthesis of these high-value co…

Tryptophan-kynurenine metabolic reprogramming along the gut-brain axis alleviates Alzheimer's pathology.

The gut–brain axis influences neuroinflammation and metabolic homeostasis in Alzheimer’s disease (AD). Disruption of gut microbiota and barrier function promotes amyloid and tau pathology via immune and metabolic dysregulation. In this study, Limosilactobacillus fermentum SRK414 (SRK414) was orally administered to ADLPAPT mice, resulting in reduced Aβ and tau pathology and improved cognition. Multi-omics analysis revealed that SRK414 altered gut microbial composition and increased hippocampal kynurenic acid (KYNA), a metabolite linked to neuroimmune regulation. Increased hippocampal KYNA was a

Tryptophan metabolism in tumor immune escape: mechanisms, cellular crosstalk, and therapeutic opportunities.

Tumor immune escape is increasingly recognized as an immunometabolic process shaped not only by immune checkpoints and suppressive cell populations, but also by nutrient competition and metabolic signaling within the tumor microenvironment. This nutrient-competitive environment is not limited to tryptophan depletion, but also involves glucose restriction, glutamine dependence, arginine metabolism, amino acid transporter competition, and impaired mitochondrial fitness of effector T cells. Among amino acid pathways, tryptophan metabolism has emerged as a central regulator of tumor-immune interac…

An enzyme-responsive nanoplatform rewires host-microbe tryptophan metabolism for cancer immunotherapy.

Immunotherapy shows great promise against cancer, yet imbalanced tryptophan (Trp) metabolism drives tumor immune tolerance, severely limiting its efficacy. To address this, we advance a synergistic host-microbe metabolism-regulated strategy that concurrently amplifies microbial indole pathway while suppressing the host kynurenine (Kyn) pathway to correct metabolic and immune dysregulation, thereby enhancing antitumor immunity. To produce an oral co-delivery nanoparticle, mesoporous silica nanoparticles are loaded with Trp via ester bonds and coated with agar containing the metabolism inhibitor…

Tryptophan metabolism in neonicotinoids exposure-induced diabetes: Emerging insights and predictive implications.

BACKGROUND: Exposure to individual neonicotinoids (NNIs) suggestively correlates with the type 2 diabetes mellitus (T2DM) risk in animals. H … NNIs increase the risk of developing both T2DM and PDM. Non-targeted metabolomics revealed that the tryptophan (Trp) metabolic pathway mediates the development of diabetes mellitus, with kynurenine (KYN)-mediated THD association with T2DM showing the largest proportion of mediation at 24.93%. A predictive model for NNI-diabetes was developed by incorporating …

Branching architecture of tryptophan metabolism determines therapeutic vulnerability in acute myeloid leukemia.

Acute myeloid leukemia (AML) exhibits metabolic reprogramming that supports immune evasion and treatment resistance. The kynurenine pathway … rom AML patients before and after induction therapy. Treatment was associated with normalization of tryptophan depletion and kynurenine accumulation in responders, indicating partial restoration of systemic KP homeostasis. Notably, baseline (pre-treatment) samples from patients who were later classified as non-responders exhibited a distinct metabolic phenot …

Tryptophan Metabolism-Related Biomarkers in Acute Myocardial Infarction Identified by Machine Learning.

Emerging evidence suggests that tryptophan metabolism (TrM) is dysregulated in acute myocardial infarction (AMI), but the underlying mechanisms remain unclear. In this study, we integrated weighted gene co-expression network analysis, differential expression analysis, and four machine learning algorithms to identify key TrM-related genes in AMI. The diagnostic model was further validated by quantitative polymerase chain reaction (qPCR), and functional enrichment, immune infiltration, molecular docking, molecular dynamics simulation, and single-cell analyses were performed to investigate the po…

Tryptophan metabolism in colorectal cancer: From mechanistic insights to novel therapeutic strategies.

BACKGROUND: Tryptophan metabolism drives colorectal cancer (CRC) pathogenesis through three interconnected cascades: the kynurenine pathway, the serotonin pathway, and the gut-microbiota-dominated indole pathway. These pathways exert context-dependent dual activities determined by disease stage, metabolite concentration, cell type and tumor microenvironment. Substrate competition and metabolic flux redistribution occur across the three branches, while extra-intestinal tryptophan metabolites mediate multi-organ crosstalk. Despite substantial preclinical findings, clinical translation is hindere…

Microbiota-Derived Tryptophan Metabolite Indole-3-Propionic Acid-Emerging Role in Neuroprotection.

In recent years, gut-brain axis signaling has been recognized as an essential factor modifying behavior, mood, cognition, and cellular viability under physiological and pathological conditions. Consequently, the intestinal microbiome has become a potential therapeutic target in neurological and psychiatric disorders. The microbiota-derived metabolite of tryptophan (Trp), indole-3-propionic acid (IPA), was discovered to target a number of molecular processes and to impact brain function. In this review, we outline the key mechanisms by which IPA may affect neuronal activity and survival and pro…

Does smoking influence tryptophan metabolism in periodontal inflammation? A cross-sectional study.

OBJECTIVES: The aim of this study was to identify the effects of smoking and periodontal inflammation on tryptophan-kynurenine metabolism as well as the correlation between these findings and clinical periodontal parameters. BACKGROUND: It has been shown that the tryptophan amino acid's primary catabolic pathway, the kynurenine pathway (KP), may serve as a key biomarker for periodontal disease. Although there are studies investigating the effect of smoking on KYN-TRP metabolism, the effect of smoking on periodontal disease through KP has not been revealed so far. METHODS: The salivary and se…

Mixture Effects of Tryptophan Intestinal Microbial Metabolites on Aryl Hydrocarbon Receptor Activity.

Aryl hydrocarbon receptor (AHR) plays pivotal roles in intestinal physiology and pathophysiology. Intestinal AHR is activated by numerous dietary, endogenous, and microbial ligands. Whereas the effects of individual compounds on AHR are mostly known, the effects of real physiological mixtures occurring in the intestine have not been studied. Using reporter gene assays and RT-PCR, we evaluated the combinatorial effects (3520 combinations) of 11 microbial catabolites of tryptophan (MICTs) on AHR. We robustly (n = 30) determined the potencies and relative efficacies of single MICTs. Synergistic e…

Myocardial oxygen consumption during histidine-tryptophan-ketoglutarate cardioplegia in young human hearts.

OBJECTIVES: Energy demand and supply need to be balanced to preserve myocardial function during paediatric cardiac surgery. After a latent aerobic period, cardiac cells try to maintain energy production by anaerobic metabolism and by extracting oxygen from the given cardioplegic solution. Myocardial oxygen consumption (MVO2) changes gradually during the administration of cardioplegia. METHODS: MVO2 was measured during cardioplegic perfusion in patients younger than 6 months of age (group N: neonates; group I: infants), with a body weight less than 10 kg. Histidine-tryptophan-ketoglutarate cry…

Modular control of l-tryptophan isotopic substitution via an efficient biosynthetic cascade.

Isotopologs are powerful tools for investigating biological systems. We report a biosynthetic-cascade synthesis of Trp isotopologs starting from indole, glycine, and formaldehyde using the enzymes l-threonine aldolase and an engineered β-subunit of tryptophan synthase. This modular route to Trp isotopologs is simple and inexpensive, enabling facile access to these compounds.

Antidepressant-Like Effect of β-Lactolin, a Glycine-Threonine-Tryptophan-Tyrosine Peptide.

The number of patients with mental illnesses, including depression, is rapidly increasing, and daily lifestyle is closely associated with the development of symptoms. Consequently, corrective measures, such as diet-based treatment for diseases, are receiving great attention. We previously showed that β-lactolin, a β-lactopeptide of glycine-threonine-tryptophan-tyrosine peptide, inhibits monoamine oxidase and improves memory impairment in mice, but the effects on depression have not been investigated. Here we showed that β-lactolin improved depression-like behavior via dopamine-D1-like receptor…

Palladium-Catalyzed Imidoylative Cyclization of Tryptophan-Derived Isocyanides: Access to β-Carbolines.

A palladium-catalyzed imidoylative cyclization of ethyl-3-(1H-indol-3-yl)-2-isocyanopropanoates, derived from readily available tryptophan, to afford β-carboline derivatives has been developed. The reaction proceeds smoothly under mild conditions through sequential isocyanide insertion, intramolecular C-H imidoylation, and aerobic dehydrogenative aromatization with a decent substrate scope. This method provides a general approach for the synthesis of molecules containing the β-carboline fragment.
