Importance of Inclusion of Pregnant and Breastfeeding Women in COVID-19 Therapeutic Trials.

Investigators are employing unprecedented innovation in the design of clinical trials to rapidly and rigorously assess potentially promising therapies for coronavirus disease 2019 (COVID-19); this is in stark contrast to the continued near-universal regressive practice of exclusion of pregnant and breastfeeding women from these trials. The few trials that allow their inclusion focus on postexposure prophylaxis or outpatient treatment of milder disease, limiting the options available to pregnant women with severe COVID-19 to compassionate use of remdesivir, or off-label drug use of hydroxychlor

Analysis of circulating tumor DNA by targeted ultra-deep sequencing across various non-Hodgkin lymphoma subtypes.

Although targeted deep sequencing of cell-free DNA (cfDNA) was recently used to investigate tumor somatic mutations in particular subtypes of non-Hodgkin lymphomas (NHLs), the immense genetic heterogeneity across subtypes poses a hurdle to design a universal gene panel applicable for diverse subtypes of NHLs. We designed a panel targeting 66 genes associated with NHLs and performed targeted deep sequencing to analyze plasma cfDNA from patients with various subtypes of NHLs. Genetic profiling in plasma cfDNA using the method resulted in 88.0% sensitivity and >99% specificity in detecting mutati

Application of photocatalytic cadmium sulfide nanoparticles to detection of enzymatic activities of glucose oxidase and glutathione reductase using oxidation of 3,3',5,5'-tetramethylbenzidine.

It was found out that semiconductor CdS nanoparticles (NPs) are able to catalyze photooxidation of the well known chromogenic enzymatic substrate 3,3',5,5'-tetramethylbenzidine (TMB) by oxygen. The photocatalytical oxidation of TMB does not require hydrogen peroxide and its rate is directly proportional to the quantity of CdS NPs produced in situ through the interaction of Cd(2+) and S(2-) ions in an aqueous medium. This phenomenon was applied to development of colorimetric sensitive assays for glucose oxidase and glutathione reductase based on enzymatic generation of CdS NPs acting as light-p

Approaches on genetic polymorphism of Cryptococcus species complex.

Neisseria meningitidis is exclusively a human-adapted bacterium, most frequently found in asymptomatic carriage that promotes natural immunity. However, it is also the causative agent of severe invasive infections, such as septicaemia and/or meningitis that may lead to life-threatening septic shock. Vaccination with capsular polysaccharidic antigens (either plain or conjugated) induces serogroup specific protective antibodies. Meningococcal capsular polysaccharide vaccines are only available against serogroups A, C, Y and W135. There is no available capsular vaccine against serogroup B. Future

Improving the selective neonatal BCG program.

OBJECTIVES: The aim of the work was to develop and implement an improved selective neonatal Bacille-Calmette-Guerin (BCG) vaccine program in which neonates are systematically assessed, and those identified as at "high risk of being exposed to tuberculosis" are given neonatal BCG. DESIGN: Service models used in other parts of the country were assessed and their limitations and difficulties were carefully considered. Possible service models for local use were then considered. A selective neonatal BCG program involving universal neonatal screening was implemented. SAMPLE: A program was implemen

[Development and role of comparative sequence analysis in medical virology].

INTRODUCTION: Development of the polymerase chain reaction and deoxyribonucleic acid sequencing techniques has enabled precise identification, classification and taxonomy of viruses. COMPARATIVE SEQUEENCE ANALYSIS: (Comparative sequence analysis methods can be used in medical virology for many practical purposes. They may be classified into three broad categories: I - reconstruction of genealogical relationships between individual viral isolatesfor detection and monitoring of sources, reservoirs and modes of viral transmission; II - virus genotyping, that is determination of relationships betw

Fluorinated Self-Assembled Monolayers for Highly Reversible Anode-Free Zinc Batteries.

Anode-free zinc batteries offer a promising route to maximize energy density, yet their viability is compromised by uncontrolled Zn nucleation and severe parasitic reactions at the anode side. Herein, we address this issue by designing a dual-functional fluorinated interphase on a Cu current collector, which couples high zincophilicity with robust hydrophobicity. This design leverages the electronegative fluorine atoms to enhance the zincophility of the sulfonyl tail group and guide uniform nucleation, while the fluorinated chains establish a compact hydrophobic barrier thermodynamically suppr

Reframing Antimicrobial Peptides beyond Direct Antimicrobial Activity toward Host-Directed Functions and Disease-Specific Therapeutic Applications.

Antimicrobial peptides (AMPs) have been extensively investigated as alternatives to conventional antibiotics to combat multidrug-resistant bacteria. This narrow antibiotic-replacement framework underestimates the biological and translational potential of AMPs. The present review reframes them as disease-context-dependent therapeutic platforms rather than universal antibiotic substitutes. In particular, this review critically examines their microbiological basis, mechanisms of action, emerging applications, engineering strategies, formulation approaches, translational limitations, and criteria

Versatile phosphonium-catechol dispersant for inorganic nanoparticle stability across diverse media and manufacturing processes.

Inorganic nanoparticles (NPs) have significant potential for various applications; however, achieving stable dispersion in organic media and polymers remains a key challenge for their industrial use. We developed a novel dispersant, tributyl(3-((3,4-dihydroxyphenylamino)-3-oxopropyl)phosphonium chloride) (TPC), which contains a catechol group for strong NPs binding and a phosphonium chloride moiety for effective dispersion. The versatility of TPC was demonstrated by stabilizing TiO₂, Fe₂O₃, and ZnO NPs in methanol using bead milling, as confirmed by TGA, DLS, zeta potential, and TEM analyses.

Rubella immunization policy in Japan: Immunity gaps arising from program differences and inconsistent cutoff standards.

Rubella elimination remains a global health priority due to the risk of congenital rubella syndrome (CRS) following maternal infection during early pregnancy. Japan's immunization policy began in 1977 with female-only vaccination, reflecting a narrow focus on CRS prevention, and gradually expanded to universal childhood immunization. However, men born between 1962 and 1978-the so-called "lost generation" - were left with few vaccination opportunities, leading to major outbreaks in 2012-2013 and 2018-2019. In response, the government introduced the Phase 5 catch-up program in 2019, while women

Rational design of PD-1-CD28 immunostimulatory fusion proteins for CAR T cell therapy.

BACKGROUND: In many situations, the therapeutic efficacy of CAR T cells is limited due to immune suppression and poor persistence. Immunostimulatory fusion protein (IFP) constructs have been advanced as a tool to convert suppressive signals into stimulation and thus promote the persistence of T cells, but no universal IFP design has been established so far. We now took advantage of a PD-1-CD28 IFP as a clinically relevant structure to define key determinants of IFP activity. METHODS: We compared different PD-1-CD28 IFP variants in a human leukemia model to assess the impact of distinctive des

Glycan Masking of Epitopes in the NTD and RBD of the Spike Protein Elicits Broadly Neutralizing Antibodies Against SARS-CoV-2 Variants.

Glycan-masking the vaccine antigen by mutating the undesired antigenic sites with an additional N-linked glycosylation motif can refocus B-cell responses to desired epitopes, without affecting the antigen's overall-folded structure. This study examined the impact of glycan-masking mutants of the N-terminal domain (NTD) and receptor-binding domain (RBD) of SARS-CoV-2, and found that the antigenic design of the S protein increases the neutralizing antibody titers against the Wuhan-Hu-1 ancestral strain and the recently emerged SARS-CoV-2 variants Alpha (B.1.1.7), Beta (B.1.351), and Delta (B.1.6

Bacterial contamination of dental chair units in a modern dental hospital caused by leakage from suction system hoses containing extensive biofilm.

Within six months of opening of the new Dublin Dental Hospital in September 1998, areas of corrosion were observed on many of the baseplates of the hospital's 103 dental chair units (DCUs) at the site of attachment of the suction hoses. The corroded areas were heavily contaminated with Pseudomonas spp. and related genera posing a risk of cross-infection, particularly for immunocompromised patients. These species were used as marker organisms to investigate the source of the contamination. P. aeruginosa was the predominant species recovered from 41 selected DCU baseplates (61% prevalence), wher
