Bronchodilator, vasodilator and spasmolytic activities of methanolic extract of Myrtus communis L.

The present study was undertaken to validate some of the folkloric claims about the effectiveness of the use of a Myrtus communis L. crude methanol extract (Mc.Cr) in gastrointestinal, respiratory and vascular diseases. Mc.Cr caused complete relaxation of spontaneous and K⁺ (80 mM)-induced contractions in isolated rabbit jejunum. It caused right ward parallel shift of calcium concentration response curves. Mc.Cr exhibited relaxant effect on CCh- and K⁺ (80 mM)-induced contractions in isolated rabbit tracheal preparations. Furthermore, Mc.Cr caused relaxation of phenylephrine (1 μM)- and K⁺ (80

ADP is a vasodilator component from Lasiodora sp. mygalomorph spider venom.

Members of the spider genus Lasiodora are widely distributed in Brazil, where they are commonly known as caranguejeiras. Lasiodora spider ve … ds from the venom. The results showed that Lasiodora spider venom induced a concentration-dependent vasodilation in rat aortic rings, which was dependent on the presence of a functional endothelium and abolished by the nitric oxide synthase (NOS) inhibitor L-NAME. Western blot experiments revealed that the venom also increased endothelial NOS function by incre …

NO- activates soluble guanylate cyclase and Kv channels to vasodilate resistance arteries.

Nitric oxide (NO) plays an important role in the control of vascular tone. Traditionally, its vasorelaxant activity has been attributed to the free radical form of NO (NO*), yet the reduced form of NO (NO-) is also produced endogenously and is a potent vasodilator of large conduit arteries. The effects of NO- in the resistance vasculature remain unknown. This study examines the activity of NO- in rat small isolated mesenteric resistance-like arteries and characterizes its mechanism(s) of action. With the use of standard myographic techniques, the vasorelaxant properties of NO* (NO gas solution

Coronary vasodilator capacity and hypertension-induced increase in left ventricular mass.

An increase in left ventricular mass represents a compensatory response of hypertensive heart to augmented loading conditions. The concept of inappropriate mass has been proposed to define an increase in left ventricular mass higher than needed to compensate for increased workload. To assess whether inappropriate left ventricular mass is associated with more severe impairment of coronary vasodilator capacity, 64 untreated middle-aged hypertensive patients without significant coronary artery stenosis and 14 normotensive volunteers comparable for age and gender were studied by transthoracic and

New NO donors with antithrombotic and vasodilating activities, part 25. Hydroxylamine derivatives.

Twelve ethoxycarbonyl or phenylsulfonyl derivatives as prodrugs of hydroxylamine or phenylhydroxylamine were prepared and tested for antiplatelet (in vitro, Born test) antithrombotic (in vivo thrombosis model), and antihypertensive (in vivo, SHR rats) effects. In the Born test N,N-bisphenylsulfonylhydroxylamine (10) was most active (IC50 = 11 mumol/L). The N-ethoxycarbonyl-phenylhydroxylamine (7) was the most potent antithrombotic compound. It inhibited the thrombus formation in mesenteric arterioles of rats by 39% after a single p.o. dose of 60 mg/kg. Compound 7 lacked any antihypertensive ac

New NO donors with antithrombotic and vasodilating activities, Part 26. Amidoximes and their prodrugs.

Seventeen amidoximes (2a-q) comprising aliphatic (2a-d), aromatic (2e-n), and bis compounds (2o-q) have been synthesized. In the Born test 4-chlorophenylethenecarboxamidoxime (21) was most active and inhibited the blood platelet aggregation induced by collagen with an IC50 = 3 microM. After oral administration to rats (60 mg/kg) fourteen compounds significantly inhibited the formation of thrombi in arterioles and venules. The strongest effect was observed with ethene-bis-carboxamidoxime (2q) (31% in arterioles and 18% in venules). The O-ethoxycarbonylderivatives 3 and the corresponding 1,2,4-o

The effect of topical vasodilating agents on microvascular vessel diameter in the rat model.

Topical vasodilating agents maximize blood flow and prevent vasospasm in microvascular tissue-transfer procedures. Two commonly used agents … ere found to be superior to saline solution as vasodilating agents. Papaverine was found to sustain vasodilation longer than lidocaine, suggesting that it is a superior vasodilating agent in microvascular anastomoses.

Inhaled aerosolized prostacyclin as a selective pulmonary vasodilator for the treatment of severe hypoxaemia.

Two case reports are presented where inhaled aerosolized prostacyclin (IAP) was used to good effect as a selective pulmonary vasodilator. It was used in the treatment of a patient with severe hypoxaemia secondary to amniotic fluid embolism and for hypoxaemia secondary to the acute respiratory distress syndrome (ARDS) in a patient with acute on chronic liver failure and intra-abdominal sepsis. An apparent dose-response curve is demonstrated in the second case. A dose of IAP of 30-40 ng/kg/min produced an effect on oxygenation in the patient with liver failure equal to that seen at the maximal d

[The clinical and hemodynamic aspects of using peripheral vasodilators in hypertension in pregnant women].

The clinical efficacy of various vasodilators characterized by different direction of the hemodynamic action (arteriolar, mixed, venous) was assessed in therapy of the slight and medium-severe hypertensive syndrome in 92 pregnant women aged 19-36 at pregnancy terms of 24 to 39 weeks. The drugs were administered as monotherapy. Twenty-two healthy women with the normal gestation process made up the reference group. Echocardiograms have shown before the course of treatment the hyper-, hypo-, and eukinetic changes in the hemodynamics (26, 31, 35 cases, respectively). Venodilators (sustonit-mitte)

Endothelium-derived vasodilator responses to sympathetic stimulation of the submandibular gland in the cat.

1. The extent to which vasodilator responses to electrical stimulation of the sympathetic innervation, in the submandibular gland of the cat, depend upon release of endothelium-derived relaxing factor (EDRF) within the gland has been investigated in anaesthetized cats given N-nitro-L-arginine methyl ester (L-NAME) which specifically blocks the synthesis of EDRF from arginine. 2. Close intra-arterial infusions of L-NAME (> or = 100 mg kg-1) produced a steady and significant rise in mean aortic pressure together with a steady increase in basal submandibular vascular resistance over the next 20-3

The vasodilating effect of spinal dorsal column stimulation is mediated by sympathetic nerves.

Spinal dorsal column stimulation has been used in the treatment of a patient with a painful vasospastic condition in the right arm following surgical sympathectomy on the left side. After sympathectomy the left arm became constantly dry and warm and consistently lacked skin vasomotor (laser Doppler flowmetry) responses to arousing stimuli, indicating a complete loss of sympathetic vasomotor innervation. The return of minimal sudomotor (skin resistance) responses to mental stress 2 years after sympathectomy indicated a partial reinnervation of sweat glands. Immediately after sympathectomy, the

Vasodilators in persistent pulmonary hypertension of the newborn: a need for optimal appraisal of efficacy.

Tolazoline hydrochloride is usually the first choice pulmonary vasodilator in persistent pulmonary hypertension of the neonate (PPHN). The analysis of 26 articles including 467 tolazoline-treated infants has been hindered by many methodological drawbacks. Tolazoline has always been administered to infants suffering refractory hypoxemia, but, unfortunately, pulmonary hypertension has not usually been investigated. Moreover, 80% of the tolazoline-treated neonates had an underlying pulmonary parenchymal disease as a potential cause of severe hypoxemia. Noteworthy is that similar comments apply to

Echo/Doppler and hemodynamic correlates of vasodilator responsiveness in primary pulmonary hypertension.

To determine correlates of acute vasodilator responsiveness in primary pulmonary hypertension, we retrospectively studied 25 patients, comparing 41 resting echo/Doppler and nine resting catheterization variables with the maximal reduction in pulmonary vascular resistance achieved during vasodilator trials. Twelve vasodilators were tested (mean, 5.6 drugs per patient; range, three to eight). Eight patients were vasodilator responsive, as defined by a reduction in pulmonary vascular resistance greater than or equal to 30 percent in response to at least one agent. Univariate and multivariate anal

Direct vasodilator effect of hyperventilation-induced hypocarbia in autonomic failure patients.

Hyperventilation produces small decreases in blood pressure in normal subjects and larger decreases in patients with autonomic failure. The authors studied the mechanism for this observation by measuring mean arterial pressure (MAP) and arterial blood gas (ABG) changes in eight patients with severe primary autonomic failure after various maneuvers designed to alter PaCO2, PaO2, and pH. Maneuvers included voluntary hyperventilation, breathing a 5% CO2/95% O2 mixture, breathing 12% O2, breathing through a 1 meter tube to increase dead space, breathing 100% O2, and infusion of 120 mEq NaHCO3 over

The interaction of vasodilating drugs and sympathetic blockade in normal and ischemic canine hindlimbs.

The purpose of this study was to determine if intraarterial vasodilating drugs could augment the vasodilation produced by sympathetic blockade, such as occurs during epidural anesthesia. Papaverine (2 mg/min), nitroglycerin (1 microgram/kg/min), aand saline were infused into the femoral artery before and after lumbar sympathectomy in six arterially isolated canine hindlimbs. Femoral blood flow was controlled with a perfusion circuit at baseline (80 ml/min), half-baseline (40 ml/min), and low (5 ml/min) flow rates so that hindlimb mean arterial pressure served as an index of peripheral vasodila…

Peptidergic vasodilator nerves in the peripheral circulation and in the vascular beds of the heart and brain.

This overview focusses on the ubiquitous presence of immunohistochemically visible peptidergic nerves with vasodilatory function. The nerve fibres are primarily related to the parasympathetic system (vasoactive intestinal polypeptide or VIP), the sympathetic system including the adrenal medulla (enkephalins) and to the sensory system (substance P as well as calcitonin gene-related peptide, CGRP). Substance P and probably also CGRP seem to be the mediators of antidromic vasodilatation. Enkephalins appear to be released both from nerve endings and from the adrenomedullary cells. The vasodilatory

Medical management of chronic heart failure: inotropic, vasodilator, or inodilator drugs?

On the basis of pathophysiologic mechanisms, the medical therapy of today for chronic heart failure is reviewed. The advantages and disadvantages of the vasodilator drugs and the inotropic drugs are presented. Finally, the therapeutic value of the inodilator drugs, which combine the central myocardial effects of positive inotropic agents with those of peripheral vasodilators, is discussed. In particular, the orally available dopaminergic agents, such as ibopamine, which interact with beta-receptors in the heart (mediating a positive inotropic effect) as well as with dopaminergic receptors in t

Reversibility of primary pulmonary hypertension with vasodilator, anticoagulant and nocturnal oxygen therapy.

Successful vasodilator therapy of a 19 year old woman with primary pulmonary hypertension is described. Long-term home oxygen therapy during sleep (2 1/min, 8 hours) in combination with vasodilator drugs (prazosin 2 mg and long-acting ISDN 20 mg bid) and an anticoagulant (ticlopidine 100 mg bid) brought significant reduction of pulmonary artery pressure and a rise of cardiac output 3 years later. This combination therapy may be effective in selected patients with primary pulmonary hypertension.
