Influenza A and influenza B viruses are continuing causes of morbidity and mortality on an annual basis. Influenza A viruses have historically caused periodic pandemics in the human population, sometimes with devastating consequences, such as in 1918. Fears of a new pandemic have increased in recent years because of continuing outbreaks of highly pathogenic H5N1 avian influenza viruses in birds with occasional, but often lethal infection of humans. Despite their importance as human pathogens, the antiviral drugs approved to treat influenza virus infections are currently limited to two targets,…

Although adeno-associated virus (AAV) infection is common in humans, the biology of natural infection is poorly understood. Since it is like … pisomal AAV DNA and isolate infectious molecular clones from several human tissues. Serotype 2-like viruses were generated from these DNA clones and interestingly, failed to bind to a heparin sulfate column. Inspection of the capsid sequence from these two clones (and the other six AAV2-like isolates) revealed that they lacked arginine residues at positio …

Following infection, a virus must battle against the host's immune response. Viruses have developed many ways to escape immune surveillance and downregulate the host's immune response. Some viruses cause a generalized immunosuppression, thereby inhibiting or depressing the immune response towards themselves as well as towards unrelated pathogens. This review will focus on the mechanisms involved in the three main human viral infections causing immunosuppression: measles, human immunodeficiency virus and cytomegalovirus. We will also discuss what has been learned from the extensively studied mo…

The cutaneous or mucosal DC-T cell environments seem extremely supportive of immunodeficiency virus replication. Apart from very early after SIV infection, similar virus producing cells have been difficult to detect in the lymphoid tissues where DCs and T cells are also known to interact. Large amounts of virus can be visualized in the germinal centers of the lymph nodes, much of which represents immune complexed virus that is trapped on the follicular dendritic cell surface. However, whether these virus-carrying cells actually make virus or even virus proteins requires further investigation.

Attempts to isolate viruses from 178,181 unengorged female mosquitoes collected from different ecologic areas of Sri Lanka yielded 31 isolates: 17 of Japanese encephalitis (JE) virus, nine of Getah virus, three of a Batai-related bunyavirus, and two of Arkonam virus. Culex tritaeniorhynchus and Mansonia uniformis mosquitoes were found to carry JE virus in a dry zone nonepidemic area, and Cx. pseudovishnui was found to carry it in a wet zone nonepidemic area. Japanese encephalitis virus was isolated from Cx. tritaeniorhynchus, Cx. gelidus, Cx. fuscocephala, and Cx. whitmorei during a human epid…

We have re-evaluated the relationships among the polypeptides of eastern equine encephalitis (EEE) viruses using SDS-PAGE and peptide mapping of individual virion proteins. Four to five distinct polypeptide bands were detected upon SDS-PAGE analysis of viruses: the E1, E2 and C proteins normally associated with alphavirus virions, as well as an additional more rapidly-migrating E2-associated protein and a high M(r) (HMW) protein. In contrast with previous findings by others, the electrophoretic profiles of the virion proteins of EEE viruses displayed a marked correlation with serotype. The pro…

Two hundred eighty-five nucleotides from the capsid/premembrane gene region of 16 isolates of Japanese encephalitis (JE) virus from the southern and northern regions of Vietnam obtained during the period 1964-1988 were sequenced and compared with each other and with wild JE virus strains from China and Japan. The 16 Vietnamese isolates showed low rates of genetic evolution (< or = 3.2%) and were classified into one genotype with an overall similarity of > or = 95.4%, including the Japanese JaArOS982 strain taken as reference, and earlier isolated strains from Japan (Nakayama 1935) and China (B

Many different viruses belonging to several genera have the potential to damage beta cells. The mechanisms they employ are varied, and infection may result in either a direct destruction of islets and rapid insulin deficiency, or in a more gradual loss of functioning islets with the onset of diabetes many years later. Several case histories involving extensive cytolysis of beta cells can be directly linked to viral infection, whilst an example of diabetes occurring many years after viral infection is found in individuals who had a congenital infection with rubella virus. Here, the virus induce…

There are several viral infections which are known to cause lymphoma among animals; all establish latency in lymphoid cells. The human T-lymphotropic virus type I is a human virus which causes lymphomas among a subset of carriers. However, this virus is very restricted in its distribution and as such, is unlikely to play a role in the increase of non-Hodgkin's lymphoma (NHL). A highly prevalent infection, the Epstein-Barr virus (EBV) is known to play a role in the etiology of NHL among persons with acquired or inherited immune suppression. However, whether it is involved with "spontaneous" NHL

UNLABELLED: Digital PCR (dPCR) systems offer high sensitivity and reproducibility without requiring external control standards. However, their performance against real-time reverse transcription-PCR (rRT-PCR) for detecting respiratory viruses remains unexplored in Ghana. We therefore evaluated the performance of a novel dPCR, Lab-On-An-Array (LOAA), for the detection of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), respiratory syncytial virus (RSV), and influenza viruses type A (Flu A) and B (Flu B). A cross-sectional hospital-based study was conducted between August 2022 and J…

During surveillance for avian influenza viruses, three H5N6 viruses were isolated in chickens obtained from live bird markets in eastern China, between January 2015 and April 2016. Sequence analysis revealed a high genomic homology between these poultry isolates and recent human H5N6 variants whose internal genes were derived from genotype S H9N2 avian influenza viruses. Glycan binding assays revealed that all avian H5N6 viruses were capable of binding to both human-type SAα-2,6Gal receptors and avian-type SAα-2,3Gal receptors. Their biological characteristics were further studied in BALB/c mi…

Protein-based nanocompartments are ubiquitous in nature. They include both viruses as well as protein cages from non-viral origin. To broaden our knowledge on viruses and non-viral protein nanoshells, a profound insight into their mechanical properties is required. Nanoindentation measurements with an atomic force microscope (AFM) are increasingly being performed to probe such material properties. This single-particle approach allows for determining the shell's spring constant, their Young's modulus, as well as the force and deformation at which failure occurs. The experimental procedures for …

Macrophages are frontline effectors of innate immunity, acting as essential elements for clearing pathogens. However, viruses have continuously evolved sophisticated mechanisms to subvert these critical defenses. This review comprehensively decodes how viral pathogens systematically dismantle macrophage microbicidal capacities. It delineates the evasion of pattern recognition receptor surveillance, the hijacking of endocytic trafficking, and the active arrest of phagolysosomal maturation to secure intracellular replication niches. Furthermore, the review explores the paradoxical viral manipula…

BACKGROUND: Mosquitoes are the main vectors of arboviruses, which infect millions of people every year. These viruses depend on host factors, such as the proprotein convertase furin, for replication. While the interactions between arboviruses and furin have been widely studied in mammals, little is known about furin homologs and their role in virus replication in mosquitoes.
METHODS: We performed a comparative analysis of the sequences and predicted structures of human and other dipteran furin with their mosquito homologs. We used RT-qPCR to determine the mRNA expression of the identified fur…

ZBP1 is a nucleic acid receptor that restricts virus infection by activating an inflammatory gene response and by inducing host cell death. Left-handed Z-form nucleic acids accumulating in infected cells activate ZBP1 by binding to its N-terminal Zα domains. Signal transduction relies on the presence of RIP homotypic interaction motifs (RHIMs) that recruit the RIPK1 and RIPK3 signalling kinases to induce NF-κB activation or cell death under the forms of apoptosis or necroptosis. The expression of Zα domain proteins by poxviruses and RHIM-containing proteins by herpesviruses that antagonise ZBP…

Contamination of food and water by viruses is a major public health issue worldwide. Several viruses are associated with foodborne outbreaks, with norovirus and hepatitis A virus being the primary causes of foodborne outbreaks, followed by hepatitis E virus and rotavirus. These viruses are responsible for the majority of outbreaks reported globally, representing a significant challenge to food safety; therefore, effective viral inactivation processes are needed. This review presents and discusses recent research involving emerging technologies used for the inactivation of foodborne viruses, em…

Viruses contribute to approximately 15-20% of global cancer cases, yet the full spectrum of their oncogenic mechanisms continues to be uncovered. Beyond the classical roles of genome integration, chronic inflammation, and immune evasion, mounting evidence reveals that oncogenic viruses-including the human papillomavirus (HPV), Epstein-Barr virus (EBV), hepatitis B virus (HBV), hepatitis C virus (HCV), and Human T-cell leukemia virus type 1 (HTLV-1)-profoundly reshape the host epigenome to establish persistent infection and promote tumorigenesis. These viruses orchestrate widespread and durable…
