FAS-controlled T cells drive lymphoproliferation through glycolysis without effector differentiation.

Lymphoproliferation in autoimmune lymphoproliferative syndrome (ALPS) due to FAS deficiency is driven by highly proliferative FAS-controlled T cells (FCT) with a distinct molecular signature. Activating signals and metabolic fuels of their proliferation are poorly understood. Lymphoproliferation caused by proliferative T cells is also a hallmark of acute EBV infection. In these antiviral T cells, a metabolic switch to glycolysis underpins effector differentiation and IFNγ translation. Here, we used EBV-induced CD8 effector T cells as a benchmark to characterize FCT metabolism. Metabolic assays

A Review on Lymphatic Uptake of Large Therapeutic Proteins after Subcutaneous Injection.

Subcutaneous (SC) administration has emerged as a promising route for delivering large therapeutic proteins, offering advantages such as improved patient convenience and reduced healthcare costs. However, the mechanisms that regulate the uptake of these macromolecules are not yet fully understood. A recent surge in computational and experimental studies has systematically analyzed the transport of large proteins from the SC injection site to the lymphatic system, advancing our understanding of how tissue environments and injection parameters affect lymphatic uptake kinetics. This review summar

Synthesis of Highly Borylated 2‑Boraindanes through Multiple Borylation of Diynes.

Boraindanes, composed of aromatic two-dimensional (2D) benzene and saturated three-dimensional (3D) boracycle moieties, are considered promising candidates for bioactive compounds/probes, agents for boron neutron capture therapy, and functional materials, as they combine the photophysical properties of the 2D aromatic and the high molecular recognition ability of the 3D boracycle. However, 2-boraindanes are difficult to access due to the difficulty of constructing two unstable C-(sp3)-B bonds. Herein, we present a straightforward synthesis of highly borylated, phenanthrene-fused 2-boraindane d

Integrated abundance analysis and artificial intelligence-assisted screening reveal novel antihypertensive milk-derived tripeptides.

Milk consumption has been associated with reduced blood pressure, yet most studies have focused on a limited number of peptides, such as IPP and VPP. Here, we evaluated the antihypertensive effect of a single serving of milk in spontaneously hypertensive rats (SHRs) and developed an integrated computational-experimental framework to identify milk-derived tripeptides with potential bioactivity. Major milk proteins were subjected to in silico gastrointestinal digestion to generate a virtual tripeptide library. Artificial intelligence-assisted structural modeling of angiotensin-converting enzyme-

Dihydrotanshinone as a Natural Product-Based CYP17A1 Lyase Inhibitor for Hyperandrogenic Disorders.

Selective inhibition of CYP17A1 17,20-lyase is critical for treating hyperandrogenic disorders without the cortisol-depleting side effects of non-selective drugs like abiraterone. We evaluated tanshinones from Salvia miltiorrhiza as potential selective inhibitors using biochemical assays and computational modeling. Dihydrotanshinone (DT) emerged as the superior candidate; at 10 µM, it inhibited 17,20-lyase activity by 56.6% while preserving >93% of 17α-hydroxylase activity. This yields a selectivity index of 8.67, drastically outperforming abiraterone (0.73). Furthermore, DT displayed minimal

From Energy Deprivation Stress to Catalytic Adaptation of Reductive Dehalogenase TmrA for 1,1-DCA Dechlorination: An Experimental and in Silico Study.

Organohalide-respiring bacteria capable of metabolizing multiple organohalogens represent valuable tools for bioremediation and offer intriguing evolutionary potential for adaptation to non-native substrates. In the present study, a chloroform respiring Dehalobacter restrictus strain was examined for its adaptability to a low affinity substrate (1,1-dichloroethane) over five subcultures (~28 generations). We obtained an enhanced 11,1-dichloroethane dechlorination rate by culturing with a 1:9 ratio of native (chloroform) to non-native (1,1-dichloroethane) substrates. We identified mutations cor

Failure to classically condition G. dorotocephala and S. mediterranea flatworms using historical and contemporary light-shock paradigms.

Planarian flatworms represent one of the most evolutionarily informative nervous systems for an account of ancient bilaterian brains. Likewise, the unparalleled regenerative ability of planarians makes possible certain investigations of neural development, memory, and behavior that are simply impossible with other model organisms. Despite these facts, learning and memory are today underexplored in planarians, likely due in part to the controversial legacy of 20th-century experiments on the transfer of memories between individual flatworms. Here, we attempted to replicate and extend the classic

[Research progress on the biomechanical effects of lumbar spondylolysis on adjacent segment intervertebral discs and treatment strategies].

This study systematically reviews the biomechanical effects of lumbar spondylolysis on adjacent intervertebral discs and surgical intervention strategies. By integrating biomechanical experiments, finite element analyses, and clinical data,we found that lumbar spondylolysis significantly increased the biomechanical burden on adjacent discs, leading to reduced stiffness, abnormal stress distribution, and expanded range of motion. Comparative analysis of surgical methods revealed that direct repair techniques effectively reduced adjacent segment stress, while fusion surgery stabilized the affected seg

Molecular dynamics modeling of the Candida albicans membrane for the discovery of membrane-binding antifungal peptides.

Fungal infections caused by Candida albicans (C. albicans) pose a significant clinical challenge, especially due to the rise of multidrug-resistant strains. Antifungal peptides (AFPs) represent promising alternatives because of their high efficacy, low toxicity, and reduced risk of resistance. In this study, we integrated computational modeling and experimental validation to identify novel AFPs targeting the fungal membrane. Four candidate peptides-HIV-F9170 (GWEALKYLWNLLQYW), PinA (WRWWKWWK), PinB (WPTKWWK), and XYP1 (KIKWFKAMKSIAKFIAKDQLKKHL)-were evaluated using AlphaFold2 structure predict

A rare twist in the abdomen: diagnostic and therapeutic approaches to omental infarction - a case report and literature review.

Intraperitoneal focal fat infarction (IFFI) encompasses a group of rare conditions that are clinically and radiologically similar, arising from focal fatty tissue necrosis. These entities often mimic other acute abdominal conditions such as acute appendicitis or cholecystitis. We present the case of a 65-year-old female with progressive abdominal pain, ultimately diagnosed with IFFI using contrast-enhanced computed tomography (CT). Omental infarction (OI) was the leading diagnosis, though a definitive distinction from epiploic appendagitis (EA) could not be made on imaging. Conservative manage

Lung Adenocarcinoma Presenting as Bilateral Nonresolving Pneumonia Mimicking Pulmonary Infection.

Lung adenocarcinoma may occasionally present with radiological features that closely mimic pulmonary infection, leading to diagnostic delay. We report the case of a 63-year-old nonsmoking woman who presented with chronic cough, low-grade fever, progressive weight loss, and worsening breathlessness. Computed tomography of the thorax demonstrated bilateral peribronchial nodules and multifocal consolidations with air bronchograms, initially raising suspicion for pulmonary tuberculosis and subsequently fungal infection. Despite prolonged antimicrobial therapy, her symptoms persisted. Extensive mic

A digital twin for Escherichia coli K1 neonatal meningitis.

Escherichia coli K1 is a major Gram-negative pathogen responsible for neonatal meningitis. Despite significant progress in antimicrobial therapy and neonatal intensive care, clinical outcomes remain problematic due to delayed diagnosis, rapid disease progression and a lack of precision tools for personalized management. Here, we propose a technical and translational digital twin framework for E. coli K1 infection in neonates that integrates clinical, microbiological, physiological and molecular data within a continuously adaptive computational model. This twin would simulate bloodstream invasi

Hearing loss as the initial manifestation of Chlamydia psittaci pneumonia in an older adult: A case report and literature review.

Chlamydia psittaci (C. psittaci) pneumonia is a zoonotic infection that typically presents with fever, cough, and fatigue. Sudden sensorineural hearing loss is an uncommon manifestation and may lead to early misdiagnosis. We report a 67-year-old woman admitted with fever for 1 week and hearing loss for 3 days. Chest computed tomography revealed extensive consolidation in the right lung, and empirical anti-infective therapy was ineffective. Targeted next-generation sequencing (tNGS) of bronchoalveolar lavage fluid (BALF) detected C. psittaci. Following confirmation of the diagnosis, antimicrobi

Drug Search and Design Considering Cell Specificity of Chemically Induced Gene Expression Profiles for Disease-Associated Tissues.

The use of omics data, including gene expression profiles, has recently gained increasing attention in drug discovery. Omics-based drug searches and designs are often based on the correlations between chemically induced and disease-induced gene expression profiles; however, the cell specificity has not been considered. In this study, we designed a novel computational method for drug search and design using cell-specific correlations between drugs and diseases. A data completion technique allowed the characterization of cell-specific gene expression patterns in diseased cells. This proposed met

Elucidating the Sigmoidal Adsorption Behavior of Xenon in Flexible Hofmann-Type MOFs Through Experiments and Molecular Dynamics with Machine Learning Potentials.

Sigmoidal adsorption in flexible metal-organic frameworks (MOFs) can be achieved through a dramatic phase transition from narrow to large pores; however, such a unique phenomenon, which is not induced by abrupt phase changes, has rarely been reported. We report a series of flexible Hofmann-type MOFs, CoNi-Pipe, NiNi-Pipe, and CoPd-Pipe, pillared with a dimensional (3D) piperazine linker. Owing to the rotational flexibility and 3D dimensionality of the piperazine linker, CoNi-Pipe and CoPd-Pipe exhibited Xe-induced gate opening and the corresponding sigmoidal adsorption isotherms. CoNi-Pipe rev

50 Years of Progress in NSCLC: A New Fellow's Guide in the Clinic.

In the past 50 years, we have seen a dramatic evolution in thoracic oncology. We have gone from a time of limited understanding of the biology of lung cancer, no demonstrated benefits for screening, and marginal benefits from therapy to an era of molecular profiling, survival benefits from low-dose computed tomography screening, and a steadily expanding list of therapeutic options. Coupled with these advances have been substantial improvements in approaches related to supportive care and end of life. In this state-of-the-art article, the Communications Committee of the International Associatio

Probabilistic-graph-based spatial context-aware framework for interpretable spatial omics denoising and augmentation.

Spatially resolved omics technologies offer unprecedented insight into tissue organization, yet current analytical methods face challenges in effectively handling technical noise while preserving biological heterogeneity. We present CadaST, an interpretable and unified computational framework that integrates spatially aware feature selection and an adaptive imputation strategy to address this limitation. By inferring the spatial molecular pattern for each feature and conducting pattern-guided aggregation, CadaST effectively denoises and augments spatial omics data while preserving sharp biolog

Searching for Novel Antiviral Agents as COVID19 Treatments: Guanidino Diaryl Thioureas.

The COVID-19 pandemic highlighted the urgent need for effective antiviral treatments targeting SARS-CoV-2. TMPRSS2, a serine protease essential for viral entry into host cells, represents a promising therapeutic target, and this study explores guanidino diaryl thioureas as potential TMPRSS2 inhibitors. Initial screening identified a "hit-compound" (1) with reversible inhibitory activity against TMPRSS2. Computational studies, including docking and molecular dynamics simulations, were conducted to optimize derivatives of compound 1. Twenty-five derivatives were synthesized, and their pharmacoki
