Hepatitis B viremia manifesting as polyarteritis nodosa and secondary membranous nephropathy.

Renal involvement in hepatitis B-polyarteritis nodosa (HBV-PAN) usually occurs in the form of hypertension, microscopic hematuria, proteinuria but nephrotic range proteinuria or renal failure is very uncommon. A 60-year-old man had abdominal pain for 15 days which was followed by bilateral pedal edema in a day and in next three days he had sudden onset bilateral foot drop with numbness. He had weight loss of 10 kg in last 20 days. Pedal edema was pitting, bilateral. Macular skin rashes around both ankles were present. Serum creatinine was 2.4 mg/dl and blood urea nitrogen was 102 mg/dl.24 hour

Mechanisms of bacterial resistance to chromium compounds.

Chromium is a non-essential and well-known toxic metal for microorganisms and plants. The widespread industrial use of this heavy metal has caused it to be considered as a serious environmental pollutant. Chromium exists in nature as two main species, the trivalent form, Cr(III), which is relatively innocuous, and the hexavalent form, Cr(VI), considered a more toxic species. At the intracellular level, however, Cr(III) seems to be responsible for most toxic effects of chromium. Cr(VI) is usually present as the oxyanion chromate. Inhibition of sulfate membrane transport and oxidative damage to

Exercise intolerance due to mutations in the cytochrome b gene of mitochondrial DNA.

BACKGROUND: The mitochondrial myopathies typically affect many organ systems and are associated with mutations in mitochondrial DNA (mtDNA) that are maternally inherited. However, there is also a sporadic form of mitochondrial myopathy in which exercise intolerance is the predominant symptom. We studied the biochemical and molecular characteristics of this sporadic myopathy. METHODS: We sequenced the mtDNA cytochrome b gene in blood and muscle specimens from five patients with severe exercise intolerance, lactic acidosis in the resting state (in four patients), and biochemical evidence of com

p53 antigen in cervical condylomata, intraepithelial neoplasia, and carcinoma: relationship to HPV infection and integration.

It has been proposed that wild-type p53 cell-regulating functions are annulled in human cervical carcinomas, either by mutations in the human papillomavirus (HPV)-negative cases or as a consequence of their complexing with HPV E6. The aim of this study was to test this hypothesis on 39 fresh cervical biopsies by p53 immunocytochemistry (ICC) with antibody PAb 240 and with NISH (non-isotopic in situ hybridization) and PCR (polymerase chain reaction) for HPV detection. p53 protein was present in the basal layer of pure wart virus infection; the basal to middle third of CIN (cervical intraepithel

Molecular Characterization of Atypical Hepatitis B Serological Profiles in HBsAg-Negative Women of Childbearing Age in Gabon.

Occult hepatitis B infection (OBI) and mutated forms of the hepatitis B virus (HBV) represent diagnostic challenges, especially in individuals with atypical serological profiles. This study explores the molecular characteristics of HBV in HBsAg-negative women of childbearing age exhibiting atypical serological markers. We selected 100 HBsAg-negative sera from a cohort of 433 women aged 15-45 years. Additional HBV serological markers (anti-HBc, anti-HBs, HBeAg, anti-HBe) were assessed. Real-time PCR targeting the HBV S gene was performed on samples presenting atypical profiles. Socio-demographi

A Programmable Toolbox for Fragment-Specific DNA Labeling by Coupling a Chimeric γ‑Peptide Nucleic Acid-Peptide Invasion Probe with T7 Endonuclease I.

Site-specific and fragment-selective internal labeling of double-stranded DNA (dsDNA) remains a fundamental challenge, as most existing strategies rely on thermal denaturation, predefined sequence motifs, or enzymatic incorporation or are limited to end-labeling. Concurrently, conventional peptide nucleic acids (PNAs) exhibit poor invasion efficiency in mixed-sequence and GC-rich dsDNA due to insufficient binding energy and limited conformational preorganization. Here, we report a chimeric γPNA-peptide invasion probe that enables programmable, site-specific invasion and internal fluorescence l

Targeting mutant p53 in cancer: from mechanistic insights to therapeutic strategies.

The tumor suppressor p53, often referred to as the “guardian of the genome,” is mutated in more than half of human cancers. These mutations have a significant impact on cancer biology and response to therapy. p53 mutations involve a range of changes that interfere with DNA binding, eliminate tumor suppressor functions, and, in many cases, confer dominant-negative or gain-of-function characteristics that actively promote oncogenesis. These mutations are closely linked to poor clinical outcomes and resistance to standard treatments. This review examines the impact of p53 mutations on cancer ther

ETfinder: harnessing conserved C-terminal tails of single-stranded DNA-binding proteins for mining and engineering RecET systems in non-model microbial chassis.

Nonmodel microorganisms offer substantial potential as next-generation microbial chassis (NGMCs), yet most lack efficient and broadly transferable genome-editing systems. Here we present ETfinder, a framework that uses the conserved C-terminal tail of host single-stranded DNA-binding proteins (SSB-Ct) as a biochemical constraint to guide the discovery of RecET recombineering systems. Applied to Rhodobacter sphaeroides, ETfinder identified 91 candidates from 18 841 α-proteobacterial genomes, and all five experimentally tested RecT homologs supported measurable double-stranded DNA (dsDNA) recomb

Efficacy of Ultraviolet Light Disinfection of the Waterborne Microsporidian Pathogen Ecytonucleospora hepatopenaei (EHP).

The microsporidian Ecytonucleospora hepatopenaei (EHP) is an environmentally persistent, waterborne pathogen that threatens global shrimp aquaculture. This study evaluated the effectiveness of medium-pressure ultraviolet (MPUV) irradiation for disinfecting the unculturable EHP spores in water. Spore viability was assessed using a modified dual-fluorescence staining method. MPUV-induced DNA damage of spores was evaluated through both conventional PCR and quantitative PCR (qPCR) targeting the spore wall protein (SWP1) and small subunit ribosomal RNA (SSU) genes. Results showed that MPUV irradiat

The role of transcription-coupled nucleotide excision repair (TC-NER) during mammalian forebrain development.

Mechanisms that maintain genome integrity are crucial for coordinating transcription that drives mammalian forebrain development. Neural progenitor cells and differentiating neurons in the developing forebrain sustain high transcriptional activity and metabolic demand and are therefore vulnerable to DNA damage. Transcription-coupled nucleotide excision repair is a specialized DNA repair pathway that helps to mitigate damage induced by 'bulky' adducts such as UV-induced pyrimidine dimers and monoadducts formed by reactive oxygen species. TC-NER factors, which include CSB/CSA, UVSSA-USP7, ELOF1,

Clomiphene citrate induces changes in human fallopian tube secretory epithelial cells: An in vitro study.

The fallopian tube microenvironment supports gamete transport, fertilization and early embryo development. Any disturbances in this microenvironment can lead to fertilization failure, infertility or ectopic pregnancy. In this study, we systematically investigated the effects of clomiphene citrate (CC) on human PAX8-positive fallopian tube secretory epithelial cells (hPFTSECs) to assess CC-induced alterations in the tubal microenvironment. Human fallopian tube tissues obtained from women undergoing postpartum tubectomy were enzymatically digested, and the isolated hPFTSECs were cultured with CC

Recent Advances in Therapy for the Neurodegenerative Disorder Ataxia-Telangiectasia.

At present, there is no cure for the human genetic disorder ataxia-telangiectasia (A-T), which is managed by supportive care. This disorder arises due to mutations in the ATM (ataxia-telangiectasia mutated) gene and is characterised by a defect in the response to DNA damage, oxidative stress, mitochondrial dysfunction and immune deficiency. The ATM protein is activated by DNA damage, reactive oxygen species (ROS), and a variety of other stimuli, which leads to the phosphorylation or altered cellular localisation of multiple protein substrates that participate in cellular defence pathways. ATM

Multilocus, morphological, and ecological data reveal the presence of three new species of the genus Neodiplostomum Railliet 1919 (Digenea: Diplostomidae), endoparasites of birds from the Neotropical region of Mexico.

Neodiplostomum Railliet, 1919, is a cosmopolitan genus that parasitises mainly the intestines of birds of prey, and occasionally those of passerines, cuculiforms, and mammals. Adults of three undescribed species of Neodiplostomum were obtained from five bird species from three families (Accipitridae, Cuculidae, and Tyrannidae) in a single locality. Four molecular markers, namely, the small (SSU) and large (LSU) subunits and internal transcribed spacers (ITS1 and ITS2 plus 5.8S) from nuclear ribosomal DNA, and cytochrome c oxidase subunit 1 (cox 1) from mitochondrial DNA, were sequenced from sp

Role of pks + Escherichia coli in colorectal cancer: Mechanisms, clinical evidence, and therapeutic perspective.

Colorectal cancer (CRC) is a multifactorial disease influenced by genetic, environmental, and microbial factors. Within the gut microbiota, pks + Escherichia coli has emerged as a key contributor to colorectal carcinogenesis through the production of colibactin, a genotoxic secondary metabolite. Colibactin induces DNA damage by forming interstrand crosslinks and promoting replication-associated double-strand breaks, thereby triggering host DNA damage response pathways and genomic instability. Recent advances in genomic profiling have identified distinct mutational signatures, such as SBS88 and

[BRCA1 Gene's Mutations And Hereditary Breast Cancer: Genetic, Biological, And Clinical Aspects].

INTRODUCTION: Hereditary breast cancer accounts for approximately 5 to 10% of all breast cancer cases. Mutations in the BRCA1 gene, which plays a central role in DNA repair and cell cycle regulation, are the main cause of these familial forms and are strongly associated with aggressive subtypes, particularly triple-negative breast cancer. METHODS: A narrative literature review was conducted using biomedical databases (PubMed, Scopus, Web of Science, Google Scholar) between January 2024 and June 2025. Eligible publications addressed the genetic, biological, epidemiological, and clinical aspect

Cancer Cell-Derived Large Extracellular Vesicles Promote Venous Thromboembolism by Activating NETosis Through Delivering CYBA.

Venous thromboembolism (VTE) is the second-leading cause of cancer-associated mortality. Neutrophil extracellular trap formation (i.e., NETosis) is a crucial process in forming VTE in cancer patients. Nevertheless, how cancer cells contribute to NETosis remains unclear. This study investigated the potential activation effects of cancer cell-derived extracellular vesicles (CC-EVs) on neutrophils. Both small and large EVs (sEVs and lEVs) released from cancer cells are found to significantly induce NETosis in neutrophil-like HL-60 (dHL-60) cells. Following an in-depth exploration of EV-induced NE

A synthetic benzoxazine dimer derivative targets c-Myc to inhibit colorectal cancer progression.

The c-Myc protein is a well-known oncoprotein that plays a crucial role in regulating cell growth, proliferation, and differentiation. The overexpression or dysregulation of c-Myc is commonly associated with tumorigenesis in several cancers, including colorectal cancer (CRC). c-Myc forms a heterodimer with its partner MAX to activate the expression of various genes. Here, we synthesized a novel c-Myc-targeting small molecule, 2,2'-((cyclohexylazanedyl)bis(methylene))bis(4-ethylphenol), or ECD, and demonstrate ECD's anticancer activity via interference with the c-Myc/MAX dimer to promote c-Myc

Methylation analysis as a diagnostic tool for sweat gland tumours classification with emphasis on the distinction between digital papillary adenocarcinoma and mimickers.

AIMS: Digital papillary adenocarcinoma (DPA) is a rare sweat gland carcinoma arising on acral sites. The main differential diagnosis included tubular adenoma, hidradenoma, poroid hidradenoma, and mixed tumours, distinction between DPA and these mimickers being crucial for therapeutic management. Recently, HPV42 was identified as the main oncogenic driver of most DPA. Controversially, a few sweat gland tumour cases diagnosed as "DPA" but lacking the HPV42 genome and harbouring instead a BRAFV600E mutation, a genetic hallmark of tubular adenomas, have been recently described. Methylation analysi
