[Binding sites for A2 and A24 monoclonal antibodies on the alpha-latrotoxin molecule].

alpha-Latrotoxin (alpha-LTX) binding sites to functionally active monoclonal antibodies (MA) A4 and A24 were localized using three approaches: hydrolysis of the toxin followed by the N-terminal sequencing of immunoreactive peptides; the study of antibody interaction with several recombinant alpha-LTX fragments; and Western immunoblotting of synthetic overlapping peptides (6-8 aa) whose structures correspond to that of the immunoreactive alpha-LTX fragment. It was shown that the MA A4 epitope is located within the F234-M294 protein fragment and that of MA A24 interacts with the fragment 347FDKD

Spectrum of virus inhibition by consensus interferon YM643.

The spectrum of viruses inhibited by a genetically engineered consensus interferon (IFN) YM643 (interferon alfacon-1) was evaluated using a cytopathic effect inhibition assay or plaque inhibition assay for five DNA viruses and 12 RNA viruses. This activity was compared to that of natural IFN-alpha derived from Namalwa lymphoblastoid cell line [IFN-alpha (Namalwa)]. The viruses inhibited by both IFNs were herpesvirus types 1 and 2, human cytomegalovirus, varicella-zoster virus, vesicular stomatitis virus, yellow fever virus, bovine viral diarrhoea virus, Semliki Forest virus, western equine enc

A 5'-splice site mutation in the cytochrome P450 steroid 17alpha-hydroxylase gene in 17alpha-hydroxylase deficiency.

17alpha-Hydroxylase deficiency (17OHD) is an autosomal recessive disorder that produces an excess of mineralocorticoids and sexual differentiation abnormalities. Using DNA sequencing analysis of the 17alpha-hydroxylase (CYP17) gene from a Japanese patient with 17OHD, we identified a new type of genetic abnormality in this disease, a G to A transition at position +5 in the splice donor site of intron 7 of the CYP17 gene. In vitro expression analysis of an allelic minigene that consists of exons 6-8 of the patient's CYP17 gene showed that the transition causes the skipping of exon 7. This exon s

HTLV-I-associated myelopathy in a Californian: diagnosis by reactivity to a viral recombinant antigen.

A male patient developed leg numbness and weakness, and bowel, bladder, and erectile dysfunction. Examination revealed an isolated thoracic myelopathy, with lower-extremity spasticity, decreased vibration and position sense, hyperreflexia, and Babinski's signs. Serum and CSF showed antibody reactivity to human T-cell lymphotropic virus type I or II (HTLV-I/II), suggesting HTLV-I-associated myelopathy. Antibody reactivity to a unique HTLV-I recombinant protein provided definitive diagnosis of HTLV-I infection.

Increased biological activity of a recombinant factor IX variant carrying alanine at position +1.

In attempts to improve the post-translational modification and processing of recombinant factor IX (FIX) we have altered the cDNA sequence encoding pre-pro-FIX using site-directed mutagenesis and have expressed the variant cDNAs in BHK21 cells using a vaccinia-virus-derived vector. We find that substitution of the tyrosine residue at +1 for an alanine increases the biological activity of the recombinant molecules 2-fold. On the other hand, substitution of the proline at -3 for a valine results in no significant change to the specific activity of the protein. Other alterations to the N-terminus

Intron-mediated recombination may cause a deletion in an alpha 1 type I collagen chain in a lethal form of osteogenesis imperfecta.

To understand the nature of the mutation in type I collagen genes in cells from an infant with the perinatal lethal form of osteogenesis imperfecta (type II), we cloned and sequenced almost 2 kilobases of a normal alpha 1(I) collagen gene and the corresponding region of a mutant alpha 1(I) gene from cell strain CRL 1262. The mutant gene had undergone recombination between two non-homologous introns, which resulted in the loss of three exons coding for 84 amino acids in the triple-helical domain. The deletion predicted the loss of amino acid residues surrounding and including the methionine at

Advances in Feline Panleukopenia Virus Vaccines: Immunological Mechanisms, Current Challenges, and Future Perspectives.

Feline panleukopenia is a highly contagious and often fatal disease in cats caused by the feline panleukopenia virus (FPV), a member of the Parvoviridae family. Despite the widespread use of vaccination, FPV remains a significant threat to both domestic and wild felid populations worldwide, particularly in young or unvaccinated animals. Effective vaccination strategies are therefore essential for controlling the disease and reducing mortality. Current vaccines, including modified live and inactivated vaccines, have demonstrated substantial efficacy in inducing protective immunity; however, sev

In vitro validation of a CRISPR/Cas9 guide RNA targeting COL1A1 gene by cleavage assay.

The CRISPR/Cas9 system has become a widely used tool for genome editing; however, editing efficiency is largely determined by the design and functional activity of the guide RNA (gRNA). Experimental validation prior to cellular applications is particularly important when targeting genes associated with genetic disorders, such as COL1A1, which is frequently mutated in Osteogenesis Imperfecta. This study performed an in vitro functional validation of a CRISPR/Cas9 gRNA designed to target exon 2 of the COL1A1 gene, using a cell-free cleavage assay as a preliminary step before cellular genome edit

Efficacy Analysis of Paiteling in Treating Persistent High-Risk Human Papillomavirus after Cervical Cancer Surgery.

Persistent infection with high-risk human papillomavirus (HR-HPV) following cervical cancer surgery poses a significant risk for disease recurrence and progression to vaginal intraepithelial neoplasia. This retrospective cohort study evaluated the comparative efficacy of Paiteling, a Traditional Chinese Medicine compound, versus recombinant human interferon gel in treating persistent HR-HPV infection post-hysterectomy. Thirty-six patients with persistent HR-HPV positivity were allocated to either Paiteling (n = 17) or interferon (n = 19) treatment groups. Comprehensive assessment included HPV

Recombinant MAM from Faecalibacterium duncaniae exhibits a protective effect in DNBS-induced colitis.

BACKGROUND: Microbial anti-inflammatory molecule (MAM) is a key effector of the next-generation probiotic Faecalibacterium duncaniae A2-165, a species whose depletion in the gut microbiota is strongly linked to inflammatory bowel disease (IBD) and other conditions. Despite its importance, the direct anti-inflammatory effects of purified MAM have never been evaluated in vitro or in intestinal inflammation models. Prior studies have relied on bacterial supernatants, synthetic peptides, or DNA delivery systems, each with inherent limitations. RESULTS: In this study, we produced and purified reco

Cross-continental transmission and host adaptation of Mycobacterium tuberculosis in China unveiled by population history reconstruction and adaptive evolution signal detection.

The transmission history and adaptive evolution of Mycobacterium tuberculosis complex (MTBC) in China remain underexplored despite its remarkably low diversity and enduring public health burden. Here, we analyzed 23,873 whole genome sequences of MTBC to reconstruct its spread timeline and routes, population dynamics, and host adaptation patterns in China. The Bayesian coalescent models revealed the recurrent introductions of four MTBC sub-lineages (L2.2, L4.2, L4.4, and L4.5) during 1000-400 years ago, by European-Chinese transmission networks, which might have triggered the formation of their

In vivo and in vitro Models of Hepatitis B Virus Infection.

Hepatitis B virus (Orthohepadnavirus hominoidei, HBV) is a hepatotropic virus from the Hepadnaviridae family and the causative agent of both acute and chronic hepatitis B (CHB). The possible outcomes of CHB include liver cirrhosis and hepatocellular carcinoma (HCC) that pose a significant burden on the healthcare systems worldwide. In the nuclei of infected hepatocytes of patients with CHB, the HBV genome persists as a pool of covalently closed circular DNA (cccDNA) molecules. Current therapeutic strategies cannot directly target cccDNA. Instead, the available treatments focus on long-term sup

Candidate correlates of protection in the HVTN505 HIV-1 vaccine efficacy trial identified by positive-unlabeled learning.

With a goal of unveiling mechanisms by which vaccines can provide protection against HIV-1 acquisition, several studies have explored correlates of risk of HIV-1 acquisition in HVTN 505, which was a phase IIb trial conducted to assess the safety and efficacy of a DNA plasmid and recombinant adenovirus serotype 5-vectored HIV vaccine regimen among individuals in the United States who were vulnerable to acquiring HIV. While this trial failed to meet its predetermined efficacy criteria, both immunological and virological correlates of reduced risk of acquisition have been reported, suggesting tha

F116I mutation in CYP9A25 associated with resistance to emamectin benzoate in Spodoptera litura.

BACKGROUND: The F116V mutation in the substrate recognition site 1 (SRS1) of Spodoptera exigua CYP9A186 has been demonstrated to confer ~200-fold resistance to emamectin benzoate (EB). In this study, a novel mutation (F116I) in CYP9A25, orthologous to CYP9A186, was detected in a field population of Spodoptera litura (YJ22) collected from Yuanjiang, Yunnan province, China in 2022. The association of this mutation with EB resistance was investigated. RESULTS: Two homozygous strains, YJ22-116F (wild-type at 116 position of CYP9A25) and YJ22-116I (mutant) were isolated from YJ22 through two round

User-Friendly Replication-Competent MAdV-1 Vector System with a Cloning Capacity of 3.3 Kilobases.

Mouse adenoviruses (MAdV) play important roles in studying host-adenovirus interaction. However, easy-to-use reverse genetics systems are still lacking for MAdV. An infectious plasmid pKRMAV1 was constructed by ligating genomic DNA of wild-type MAdV-1 with a PCR product containing a plasmid backbone through Gibson assembly. A fragment was excised from pKRMAV1 by restriction digestion and used to generate intermediate plasmid pKMAV1-ER, which contained E3, fiber, E4, and E1 regions of MAdV-1. CMV promoter-controlled GFP expression cassette was inserted downstream of the pIX gene in pKMAV1-ER an

Molecular Epidemiology of HIV-1 among Prisoners in Central Brazil and Evidence of Transmission Clusters.

Higher rates of human immunodeficiency virus (HIV) infection have been detected in prisoners when compared with the general population, but research into HIV molecular epidemiology and its transmission network has been lacking among them. Thus, this study aimed to verify potential HIV molecular transmission networks among prisoners. In addition, we aimed to describe the mutations related to antiretroviral resistance in these isolates. Thus, we conducted a cross-sectional survey from 2013 to 2018 in prisons in Central-Western Brazil, and the final sampling composed of 84 prisoners. Proviral DNA

The active form of the influenza cap-snatching endonuclease inhibitor baloxavir marboxil is a tight binding inhibitor.

Baloxavir marboxil (BXM) is an FDA-approved antiviral prodrug for the treatment of influenza A and B infection and postexposure prophylaxis. The active form, baloxavir acid (BXA), targets the cap-snatching endonuclease (PA) of the influenza virus polymerase complex. The nuclease activity delivers the primer for transcription, and previous reports have shown that BXA blocks the nuclease activity with high potency. However, biochemical studies on the mechanism of action are lacking. Structural data have shown that BXA chelates the two divalent metal ions at the active site, like inhibitors of th

NETosis Markers in Pregnancy: Effects Differ According to Histone Subtypes.

NETosis is an innate immune response occurring after infection or inflammation: activated neutrophils expel decondensed DNA in complex with histones into the extracellular environment in a controlled manner. It activates coagulation and fuels the risk of thrombosis. Human pregnancy is associated with a mild proinflammatory state characterized by circulatory neutrophil activation which is further increased in complicated pregnancies, placenta-mediated complications being associated with an increased thrombotic risk. This aberrant activation leads to an increased release of nucleosomes in the bl

TDP-43 maintains chondrocyte homeostasis and alleviates cartilage degradation in osteoarthritis.

OBJECTIVE: Osteoarthritis (OA) is the most prevalent age-related disorder due to cartilage degradation. Previous studies have identified aberrant chondrocyte homeostasis under extracellular stress as a key pathological mechanism behind cartilage degradation in OA. TDP-43, a DNA/RNA-binding protein has been demonstrated to participate in processing many extracellular stress responses; however, understanding of the role of TDP-43 in OA is limited. This study aims to investigate the role of TDP-43 in chondrocyte homeostasis and cartilage degradation in OA. METHODS: The role of TDP-43 during degr
