Bioactive: A new era of bioactive ingredients in topical formulations for inflammatory dermatoses.

Moisturization is the pillar of daily skin care and has significant benefit in improving hydration, strengthening the skin barrier, and ameliorating inflammatory dermatoses. Bioactive ingredients such as endocannabinoids, bioactive lipids, growth factors, microbiome modulators, and antioxidant enzymes are increasingly being incorporated into moisturizer formulations. These novel ingredients have been shown to improve skin barrier function, upregulate barrier lipid synthesis, decrease itch and inflammation, and have antioxidative properties. In this article, we highlight evidence supporting the

Apoptosis induced by Staphylococcus aureus toxins.

Apoptosis stimulated by bacterial toxins is common during infection and is now considered important in disease processes. As a major human pathogen, Staphylococcus aureus also causes apoptosis during infection. In some diseases such as atopic dermatitis and sepsis, the apoptosis induced by S. aureus influences the severity and outcome of diseases. S. aureus has various toxins, many of which have reportedly triggered apoptosis. In this review, we focused on the apoptosis-inducing toxins secreted by S. aureus, and their underlying mechanisms. Novel therapies for cancer that utilized the reconstr

Kaposi's varicelliform eruption in association with rosacea.

Kaposi's varicelliform eruption is characterized by disseminated vesiculopustules and erosions caused by a herpes virus infection superimposed on a pre-existing dermatosis. The eruption usually occurs in individuals with atopic dermatitis or other pre-existing dermatosis such as Darier's disease, pemphigus foliaceus, mycosis fungoides, Sezary syndrome, benign familial pemphigus, ichthyosis vulgaris, second-degree burns, multiple myeloma, and Grover's disease. We report here a new case of Kaposi's varicelliform eruption in a 38-year-old woman with rosacea. To our knowledge, this is the first ca

[Impaired daytime functions in children with sleep disorders].

Sleep in adequate amount and quality is essential for normal child development. Sleep disorders may affect the childs daytime functions resulting in behavioral problems such as attention deficit, aggressiveness and hyperactivity. Recent reports have suggested that chronic sleep disturbance can cause neurocognitive deficits and impaired learning abilities. Obstructive sleep apnea syndrome, asthma, atopic dermatitis and juvenile rheumatoid arthritis--are common childhood diseases that can impair normal sleep. This article reviews the sleep characteristics in these diseases and the relation betwe

Placental DNA methylation mediates the prenatal maternal stress-childhood allergy association: A cohort study.

BACKGROUND: Prenatal maternal stress is a common exposure implicated in childhood allergies. However, evidence for placental DNA methylation as a mediator is limited, with prior studies focusing narrowly on HPA-axis genes. This study aimed to examine the impact of prenatal maternal stress on offspring allergic diseases and placental DNA methylation's mediating role from an epigenome-wide perspective. METHODS: Within the Shanghai Parent-Child Cohort (n = 3883 pairs), maternal stress and childhood allergic diseases were assessed via questionnaires and medical records. Latent class analysis iden

Evaluation of Two Induction Protocols for Oclacitinib in Dogs.

BACKGROUND: Oclacitinib is approved for the treatment of canine atopic dermatitis (AD) and pruritus. The conventional administration protocol includes twice-daily oral dosing for 14 days with a subsequent reduction to once-daily administration. This reduction sometimes leads to an increase in pruritus. HYPOTHESIS/OBJECTIVES: The aim of this study was to compare the changes in skin lesions and pruritus following a shortened induction versus the conventional protocol for oclacitinib. ANIMALS: Forty-five client-owned dogs with AD were randomised into two groups. MATERIAL AND METHODS: The conve

Case Report: Treatment of trachyonychia in children with crisaborole: a retrospective case series.

BACKGROUND: Trachyonychia is a chronic nail disorder affecting children aged 3-12 years, characterized by nail plate roughness and longitudinal ridging. It exhibits pathological features similar to atopic dermatitis, including spongiotic edema and inflammatory cell infiltration of the nail epithelium. Most pediatric cases are idiopathic, and high-quality therapeutic evidence remains limited. Crisaborole, a novel nonsteroidal PDE4 inhibitor with potent anti-inflammatory activity, may represent a promising therapeutic strategy. METHODS: This retrospective study was conducted at the Department o

Prevalence of allergic diseases and evaluation of total IgE and blood eosinophils as diagnostic biomarkers in Colombia, a tropical country.

INTRODUCTION: The prevalence of allergic conditions and inflammatory T2 diseases has been widely studied in developed countries but less explored in developing countries located in the tropics. Total IgE (tIgE) and blood eosinophils count (BEC) are frequently used for assessing type 2 (T2) inflammation. However, their diagnostic performance information is limited in populations where different non-allergic factors could affect the interpretation of these biomarkers. OBJECTIVE: To determine the prevalence of asthma, rhinitis, conjunctivitis, rhinosinusitis, and atopic dermatitis, in Colombian

RNA velocity and beyond: Current advances in modeling single-cell transcriptional dynamics.

Single-cell RNA sequencing (scRNA-seq) has revolutionized biology through high-throughput quantification of gene expression at individual cell resolution. However, standard scRNA-seq provides only static cellular snapshots, obscuring dynamic processes that unfold temporally, such as differentiation, reprogramming, and disease progression. RNA Velocity, introduced in 2018, offers a groundbreaking solution. By leveraging unspliced pre-mRNA and spliced mRNA information, RNA Velocity models infer instantaneous gene expression change rates and effectively predict future transcriptional states over

Real-World Use of Ruxolitinib Cream: Safety Analysis at 1 Year.

BACKGROUND: Ruxolitinib cream is the first topical Janus kinase (JAK) inhibitor approved in the United States (US) for the treatment of mild to moderate atopic dermatitis and nonsegmental vitiligo. A postmarketing study with oral tofacitinib, approved for rheumatoid arthritis, triggered class warnings for JAK inhibitors, including risk of serious infections, mortality, malignancy, major adverse cardiovascular events, and thrombosis. Because ruxolitinib cream is indicated for inflammatory conditions, it is subject to the same warnings as oral JAK inhibitors in the US. Here, nearly 14,000 patien
