Modulation of the metabolism of beta-L-(-)-2',3'-dideoxy-3'-thiacytidine by thymidine, fludarabine, and nitrobenzylthioinosine.

beta-L-(-)-2',3'-Dideoxy-3'-thiacytidine (3TC) is a cytosine nucleoside analog that potently inhibits the replication of human and duck hepatitis B viruses and human immunodeficiency virus through the activity of its 5'-triphosphate ester metabolite. The present study examined the intracellular decay of 3TC 5'-phosphates and tested strategies for modulating the cellular content of those nucleotides in primary cultures of duck hepatocytes and in human hepatoma 2.2.15 cells and CCRF-CEM T lymphoblasts. Inhibition by deoxycytidine of the 5'-phosphorylation of 3TC in duck hepatocytes confirmed tha

Is antrum or corpus the best site for culture of Helicobacter pylori?

BACKGROUND: Isolating Helicobacter pylori on culture media and performing antibiotic susceptibility testing is potentially the most useful tool for guiding antibiotic therapy, especially when antimicrobial resistance is suspected. The aim of this study was to determine whether the yield of H. pylori culture was related to the site from which the gastric specimen was obtained either before or after therapy. METHODS: Gastric mucosal biopsies from the antrum and the corpus of the stomach were cultured. H. pylori status was determined by histological assessment using the Genta stain. RESULTS: Fi

Analysis of residual amino acid--DNA crosslinks induced in intact cells by nickel and chromium compounds.

Chinese hamster ovary cells were incubated with radioactive amino acids, the DNA was isolated by standard proteinase K/phenol/chloroform extraction and residual amino acids complexed to the DNA were examined as an index of metal induced DNA-protein crosslinks. Using this method, both chromate and nickel caused residual histidine and cysteine to be complexed with the DNA isolated from metal-treated cells. In the case of chromate, a number of amino acids were studied and Tyr, Thr and Cys were found to be complexed to DNA at a level (above the untreated control) that was statistically significant

[The effect on the cerebral microcirculation of in vivo stains used in biomicroscopy].

Vasoactive properties of semi-colloidal acid stains of the Evan blue and tripane blue, were revealed in rabbits and rats. The microphoto-technique revealed a dose-depending vasodilating effect of the stains. The response of the arteries had sometimes a biphasic character: constriction--dilation. I. v. administration of the stains induced a dose-dependent significant reduction of local blood flow in the cortex, subcortical white substance and other parts of the brain, of total cerebral blood flow in the venous sinus; a drop of systemic arterial pressure in normo- and hypertensive animals; a cha

Chromium-associated 32P-labeling of proteins.

The incubation of proteins with chromium (Cr3+ or Cr6+) in the presence of 32P ([gamma-32P]ATP or H3(32)PO4) at room temperature for 10-30 min resulted in the labeling of these proteins with 32P. The 32P-labeled proteins could be separated by SDS-polyacrylamide gel electrophoresis and identified by exposure to X-ray film. The characteristics of this procedure included: the optimal chromium concentration was 100 microM; the minimum requirement of each protein was 1 microgram; the optimal pH value was between 6 and 8; metal ions such as V5+, Mn2+ and Fe3+ strongly inhibited the effect of chromiu

[Retinal protective effects of zinc-loaded magnesium oxide nanoparticles in a glutamate-excitotoxicity glaucoma model].

OBJECTIVES: Glaucoma is pathologically characterized by the progressive loss of retinal ganglion cells (RGCs). Currently, effective strategies for protection of RGCs in glaucoma remain lacking, and nanomaterials represent promising drug-delivery carriers. This study aims to investigate the effects of zinc-loaded magnesium oxide nanoparticles (MgO-Zn²⁺ nanoparticles, MgO-Zn NPs) on glutamate-induced RGC injury, and to evaluate their in vivo and in vitro biocompatibility and neuroprotective potential. METHODS: MgO-Zn NPs were prepared and characterized by transmission electron microscope and en

Pharmacovigilance analysis and real-world adverse event profile (safety signals and product-related issues) of potassium citrate: a US FDA adverse event reporting system (FAERS) based analysis.

Potassium citrate is widely used as oral alkali therapy for the prevention of recurrent nephrolithiasis. Despite its long-standing clinical use, systematically collected real-world safety data remain limited. We performed a pharmacovigilance analysis to characterize adverse event reports associated with potassium citrate using the U.S. Food and Drug Administration Adverse Event Reporting System (FAERS). FAERS reports submitted between 2014 and 2025 listing potassium citrate as the primary suspect drug were analyzed. Duplicate records were removed, and adverse events were coded using the Medica

Activity-based chemoproteomic profiling reveals the active kinome of Leishmania.

BACKGROUND: Leishmania parasites cause neglected tropical diseases such as cutaneous and visceral leishmaniasis, which have limited treatment options and rising drug resistance. Protein kinases are pivotal in Leishmania biology and attractive drug targets, but their functional status in the parasite remains largely unexplored. METHODS: We applied activity-based protein profiling (ABPP) with custom in-house cell-permeable ATP-site directed probes to map the ligandable, "active kinome" of Leishmania mexicana. Three related covalent probes featuring an ATP-mimetic scaffold, electrophilic warhead

Evaluation of Artesunate-mefloquine as a Novel Alternative Treatment for Schistosomiasis in African Children (SchistoSAM): protocol of a proof-of-concept, open-label, two-arm, individually-randomised controlled trial.

INTRODUCTION: Alternative drugs and diagnostics are needed for the treatment and control of schistosomiasis. The exclusive use of praziquantel (PZQ) in mass drug administration programmes may result in the emergence of drug resistance. PZQ has little activity against Schistosoma larvae, thus reinfection remains a problem in high-risk communities. Furthermore, the insufficient sensitivity of conventional microscopy hinders therapeutic response assessment. Evaluation of artesunate-mefloquine (AM) as a Novel Alternative Treatment for Schistosomiasis in African Children (SchistoSAM) aims to evalua
