Oxidative stress and antioxidant pathways in the pathogenesis of periodontitis and peri-implantitis: mechanistic insights and therapeutic potentials.

BACKGROUND: Periodontitis and peri-implantitis are chronic, immune-mediated inflammatory diseases characterized by progressive destruction of tooth- and implant-supporting tissues. Although microbial dysbiosis initiates these conditions, accumulating evidence indicates that host-derived oxidative stress plays a central role in amplifying inflammation, impairing tissue repair, and driving irreversible bone loss. Excessive production of reactive oxygen species (ROS) disrupts redox homeostasis, induces molecular damage, and activates redox-sensitive signaling pathways that perpetuate tissue destr

Complementary Regulation of Inflammatory, Catabolic, and PPARγ Signaling by Dexamethasone and Decanoic Acid in Donor-Specific Human Chondrocyte Models.

Background/Objectives: This study examined whether dexamethasone (Dex) and decanoic acid (DA) exert complementary effects on inflammatory signaling, catabolic gene expression, and peroxisome proliferator-activated receptor gamma (PPARγ) activation in human chondrocytes. Methods: Two donor-specific primary human chondrocyte models, each derived from a single knee donor, were used, in which cells were treated with Dex (≤0.1 µM), DA (≤500 µM), or combinations thereof. Acute Interleukin-1β(IL-1β)-induced PGE2 release was evaluated in normal donor cells, whereas prolonged transcriptional responses

Phenotype switching in highly invasive resistant to vemurafenib and cobimetinib melanoma cells.

BACKGROUND: A mutation in the BRAF (serine/threonine-protein kinase B-raf) gene is most often responsible for the progression of melanoma. A breakthrough in its treatment was the application of BRAF and MEK (mitogen-activated protein kinase kinase) inhibitors. Unfortunately, the effectiveness of this therapy is limited due to rapidly emerging resistance to the drugs. We derived two melanoma cell lines resistant to vemurafenib (a BRAF inhibitor)/cobimetinib (an MEK inhibitor). Due to the significant impact of invasion on cancer progression, we focused our further research on this process. METH

Molecular mechanisms of 10-butyl ether minocycline, a novel nonantibiotic tetracycline, as a potential treatment for inflammatory and neuroimmune-related disorders.

The pleiotropy of minocycline (MINO), including anti-inflammatory, antioxidant, antimigratory, anti-matrix metalloproteinase (MMP), and neuroprotective effects, has been extensively reported. A novel nonantibiotic MINO derivative, 10-butyl ether minocycline (BEM), was synthesized to retain the pleiotropy of MINO while minimizing side effects such as antibiotic resistance and gut dysbiosis. Previously, we showed that BEM reduced alcohol consumption in dependent murine and porcine models of alcohol use disorder. In this study, we investigated the molecular mechanisms of BEM to determine its pote

Monotropein synergizes with methotrexate to attenuate synovial inflammation in adjuvant-induced arthritis mice and fibroblast-like synoviocyte via GSK-3β.

BACKGROUND: Methotrexate (MTX) is the preferred drug for the treatment of rheumatoid arthritis (RA), but the toxic effects of long-term use … with GSK-3β. Furthermore, GSK-3β knockdown suppressed TNF-α-induced FLSs proliferation, migration, matrix metalloproteinase 2 (MMP2) and MMP3 expression, and the activation of NF-κB and STAT3 signaling, while MON lost its inhibitory effects in GSK-3β knockdown FLSs. In addition, joint tissue-specific GSK-3β knockdown in AA mice alleviated synovial inflammation, decreased p …

Matrix metalloproteinase-responsive hydrogels with tunable retention for on-demand therapy of inflammatory bowel disease.

Therapeutic options for addressing inflammatory bowel disease (IBD) include the administration of an enema to reduce intestinal inflammation and alleviate associated symptoms. However, uncontrollable retention of enemas in the intestinal tract has posed a long-term challenge for improving their therapeutic efficacy and safety. Herein we have developed a protease-labile hydrogel system as an on-demand enema vehicle with tunable degradation and drug release rates in response to varying matrix metalloproteinase-9 (MMP-9) expression. The system, composed of three tailored hydrogel networks, is cro

Comparison of 16 Pediatric Acute Respiratory Distress Syndrome-Associated Plasma Biomarkers With Changing Lung Injury Severity.

OBJECTIVES: Pediatric acute respiratory distress syndrome (PARDS) is a source of substantial morbidity and mortality in the PICU, and different plasma biomarkers have identified different PARDS and ARDS subgroups. We have a poor understanding of how these biomarkers change over time and with changing lung injuries. We sought to determine how biomarker levels change over PARDS course, whether they are correlated, and whether they are different in critically ill non-PARDS patients. DESIGN: Two-center prospective observational study. SETTING: Two quaternary care academic children's hospitals.

Matrix Metalloproteinase-7 in Urinary Extracellular Vesicles Identifies Rapid Disease Progression in Autosomal Dominant Polycystic Kidney Disease.

SIGNIFICANCE STATEMENT: There is an unmet need for biomarkers of disease progression in autosomal dominant polycystic kidney disease (ADPKD). This study investigated urinary extracellular vesicles (uEVs) as a source of such biomarkers. Proteomic analysis of uEVs identified matrix metalloproteinase 7 (MMP-7) as a biomarker predictive of rapid disease progression. In validation studies, MMP-7 was predictive in uEVs but not in whole urine, possibly because uEVs are primarily secreted by tubular epithelial cells. Indeed, single-nucleus RNA sequencing showed that MMP-7 was especially increased in p

Amelioration of Posttraumatic Osteoarthritis in Mice Using Intraarticular Silencing of Periostin via Nanoparticle-Based Small Interfering RNA.

OBJECTIVE: Recent evidence delineates an emerging role of periostin in osteoarthritis (OA), since its expression after knee injury is detrimental to the articular cartilage. We undertook this study to examine whether intraarticular (IA) knockdown of periostin would ameliorate posttraumatic OA in a murine model. METHODS: Posttraumatic OA was induced in 10-week-old male C57BL/6J mice (n = 24) by destabilization of the medial meniscus (DMM), and mice were analyzed 8 weeks after surgery. Periostin expression was inhibited by small interfering RNA (siRNA) delivered IA using a novel peptide-nucleot

Lumbar spine surgery in patients with rheumatoid arthritis (RA): what affects the outcomes?

BACKGROUND CONTEXT: Although the cervical spine is only occasionally involved in rheumatoid arthritis (RA), involvement of the lumbar spine is even less common. A few reports on lumbar spinal stenosis in patients with RA have appeared. Although disc space narrowing occurs in aging, postoperative adjacent segment disease (ASD) in patients with RA has not been subject to much analysis. PURPOSE: The objective of this study was to investigate differences in ASD and clinical outcomes between lumbar spinal decompression with and without fusion in patients with RA. STUDY DESIGN/SETTING: This is a r

The inhibitory effect of mesenchymal stem cell on blood-brain barrier disruption following intracerebral hemorrhage in rats: contribution of TSG-6.

BACKGROUND: Mesenchymal stem cells (MSCs) are well known having beneficial effects on intracerebral hemorrhage (ICH) in previous studies. The therapeutic mechanisms are mainly to investigate proliferation, differentiation, and immunomodulation. However, few studies have used MSCs to treat blood-brain barrier (BBB) leakage after ICH. The influence of MSCs on the BBB and its related mechanisms were investigated when MSCs were transplanted into rat ICH model in this study. METHODS: Adult male Sprague-Dawley (SD) rats were randomly divided into sham-operated group, PBS-treated (ICH + PBS) group,

Netazepide, a gastrin receptor antagonist, normalises tumour biomarkers and causes regression of type 1 gastric neuroendocrine tumours in a nonrandomised trial of patients with chronic atrophic gastritis.

INTRODUCTION: Autoimmune chronic atrophic gastritis (CAG) causes hypochlorhydria and hypergastrinaemia, which can lead to enterochromaffin-like (ECL) cell hyperplasia and gastric neuroendocrine tumours (type 1 gastric NETs). Most behave indolently, but some larger tumours metastasise. Antrectomy, which removes the source of the hypergastrinaemia, usually causes tumour regression. Non-clinical and healthy-subject studies have shown that netazepide (YF476) is a potent, highly selective and orally-active gastrin/CCK-2 receptor antagonist. Also, it is effective in animal models of ECL-cell tumours

Molecular Imaging of Collagen Turnover in Myocardial Infarction.

Cardiac fibrosis is a key contributor to cardiomyopathy after myocardial infarction (MI). Existing imaging techniques can detect established fibrotic changes; however, they lack sensitivity for ongoing collagen turnover-a dynamic process involving the denaturation of collagen triple helix. Molecular imaging of this process could enhance risk assessment and aid in the development of antifibrotic treatments. This study aimed to evaluate 99mTc-(HE)3-(GPO)9, a radiotracer designed to target denatured collagen, as a biomarker of collagen turnover after MI. This tracer features a polyhistidine-gluta
