Synthesis of Isochromans From Anacardic Acids Obtained From Cashew Nutshell Liquid (CNSL) and Their Antimicrobial Activity.

The synthesis of racemic isochromans from cashew nut shell liquid (CNSL) derived anacardic acids is described. Two key steps for the synthesis of the isochromans were utilized. (i) The benzylic bromination of methyl 2-methoxy-6-pentadecylbenzoate was accomplished with N-bromosuccinimide in the green solvent tert-butyl acetate to afford methyl 2-methoxy-6-(pentadec-1-en-1-yl)benzoate. (ii) Exposure of the secondary alcohol 1-(2-methoxy-6-(pentadec-1-en-1-yl)phenyl)ethanol to potassium tert-butoxide resulted in the formation of the isochroman, trans-8-methoxy-1-methyl-2-tridecylisochroman. Gut-derived biofilm-forming bacteria as a source of catheter-associated infections: inhibitory effects of O-alkyl naringenin derivatives.

The capacity for biofilm formation is a fundamental defense mechanism among antimicrobial-resistant pathogenic strains. In addition, its persistence may result in chronic colonization of host systems, and uncontrolled growth may lead to dangerous flow blockages, particularly in catheter-associated urinary tract infections. This warrants the search for novel anti-biofilm compounds to combat these pathogens. Naringenin is an example of such a structure, widely known for its multifunctional activity, targeting the synthesis of exopolysaccharide, the expression of biofilm-relevant genes, or efflux

Valorization of Turnip Greens (Brassica rapa subsp. sylvestris) Wastes: Investigation on the Sustainable Recovery of Bioactive Extracts with Antioxidant and Antibiofilm Properties.

The valorization of agri-food residues is crucial for advancing circular bioeconomy strategies and mitigating environmental impacts. Turnip greens (Brassica rapa subsp. sylvestris) are a traditional vegetable cultivated in southern Italy. While the edible portions include flower sprouts, buds, and young leaves, the more leathery leaves and stems are typically discarded. These wastes represent valuable sources of compounds with antioxidant and antimicrobial potential. This study aims to develop the extraction of phenolic compounds from turnip green residues using two techniques: silent macerati

Visible-light-responsive supramolecular enzyme mimics for combating antimicrobial resistance.

The global rise of antimicrobial resistance has created an urgent need for alternative strategies that can overcome the limitations of conventional drug-resistance mechanisms. Light-assisted antimicrobial therapy, particularly photodynamic inactivation, offers spatial and temporal control while minimizing the likelihood of resistance. Since biomedical applications demand visible-light-responsive catalysts that enable deeper tissue penetration and minimize photodamage, designing systems that avoid reliance on UV excitation is essential. This work reports a supramolecular oxidase mimic "suprazym

Siderophore-Porphyrin conjugates for bacteria-selective antimicrobial photodynamic therapy.

Targeted delivery of photosensitizers represents a key strategy for improving the selectivity and efficacy of antimicrobial photodynamic therapy (aPDT). Herein, we describe the synthesis and biological evaluation of siderophore-conjugated photosensitizers designed to exploit bacterial iron acquisition pathways. Four distinct photosensitizers-neutral porphyrin (TPP), cationic porphyrin (TMPyP), metalloporphyrin (PdTMPyP), and chlorin (Ce6)-were covalently linked to deferoxamine B (DFO), a clinically approved hydroxamate siderophore. The resulting conjugates retained their intrinsic photophysica

The Paternò-Büchi Reaction Diversifies Rifamycin Scaffolds and Enhances Antibacterial Activity.

Rifamycins display structural diversity and broad bioactivities, with the intrinsic reactivity of their polyketide chain and quinone ring providing a foundation for photochemical reactions. Herein, we constructed the mutant strain Δrif-orf3_16::rifR+rif-orf19 by deleting post-PKS modification genes of rifamycin, yielding 34a-deoxyrifamycin W (1a) and its derivatives (2a-4a). Subsequent sunlight irradiation of these compounds triggered an intramolecular Paternò-Büchi reaction coupled with Michael addition, producing oxetane-rifamycins A-D (1b-4b). They feature a unique 4/6/6/6/6/17 hexacyclic s

Narrow-spectrum drug repurposing: targeting Gardnerella vaginalis biofilms associated with bacterial vaginosis.

AIM: Bacterial vaginosis (BV) is the most common vaginal disorder in women of reproductive age. Current therapies are limited by poor activity against biofilms and high recurrence rates (>50%), demonstrating that new antimicrobials are required. Drug repurposing is an attractive approach for the discovery of new antimicrobials, so we aimed to screen repurposed libraries for activity against the key BV pathobiont Gardnerella vaginalis. METHODS AND RESULTS: Two drug libraries from Medicines for Malaria Venture comprising 640 compounds were screened against G. vaginalis and various Lactobacilli
