Elucidating relationships between P.falciparum prevalence and measures of genetic diversity with a combined genetic-epidemiological model of malaria.

There is an abundance of malaria genetic data being collected from the field, yet using these data to understand the drivers of regional epidemiology remains a challenge. A key issue is the lack of models that relate parasite genetic diversity to epidemiological parameters. Classical models in population genetics characterize changes in genetic diversity in relation to demographic parameters, but fail to account for the unique features of the malaria life cycle. In contrast, epidemiological models, such as the Ross-Macdonald model, capture malaria transmission dynamics but do not consider gene

Machine learning application in colon cancer treatment outcome prediction.

Colon cancer represents a significant global health burden, accounting for a substantial portion of cancer-related morbidity and mortality worldwide. Many studies have been conducted to predict survival outcomes; however, most of these analyses have been performed predominantly via basic statistical methods. The aim of this study was to perform machine learning techniques to build models for survival prediction in patients with colon cancer. A retrospective review of 764 colon cancer patients treated over a 10-year period facilitated the construction of a detailed dataset containing 44 predict

Testing the robustness of daily associations of affect with alcohol and cannabis use.

Etiological models of alcohol and cannabis use disorders hypothesize that people are more likely to use substances when experiencing heightened negative affect, yet recent Ecological Momentary Assessment (EMA) studies found no evidence for this daily association. To provide a robust understanding of whether and when affect regulation is supported in EMA, we tested within-person associations between affect and substance use across hundreds of statistical models in a diverse sample of young adults (n = 496) recruited from both college and community sources, aged 18-22 years (55.8% assigned femal

DNA-aware evaluation and debiasing of sequence-to-function models.

MOTIVATION: Genome sequence-to-function (S2F) models are widely used to interpret base-resolution functional genomics assays. Most S2F models are trained and evaluated against observed counts and profile-shapes using statistical objectives and fidelity metrics. These choices are well motivated, but they are DNA-independent. At the same time, experimental measurements arise from DNA-dependent assays with distinct characteristics. This mismatch motivates a complementary DNA-aware evaluation of S2F-predicted and experimental functional genomic tracks. RESULTS: We study DNA-dependency of experime

Nitric oxide inhibition ameliorates cortical proteomic changes in the Cntnap2-/- and Shank3Δ4-22 mouse models of autism spectrum disorder.

BACKGROUND: Autism spectrum disorder (ASD) is a neurodevelopmental disorder with a strong genetic component, and over a thousand associated genes have been identified, including CNTNAP2 and SHANK3. Our previous work using Cntnap2-/- and Shank3Δ4–22 ASD mouse models implicated dysregulated nitric oxide (NO) signaling in ASD-related behaviors, which were improved by inhibition of neuronal nitric oxide synthase (nNOS) with 7-Nitroindazole (7-NI). However, the molecular mechanisms linking NO signaling to ASD pathology remain poorly defined. METHODS: We performed mass spectrometry-based global prot

Influence of subchondral bone density on intra-articular stresses due to fixation hardware instrumentation and removal: A biomechanical cadaver study.

PURPOSE: Tibial plateau fractures are often surgically treated to restore native joint congruity and articular alignment. While these injuries portend an increased risk for end stage knee osteoarthritis, it is unknown whether the fixation constructs contribute to the development of osteoarthritis by influencing articular stress distribution following instrumentation. METHODS: We conducted a cadaver study measuring resultant intra-articular stresses of the native knee due to physiological levels of ex-vivo loading, after instrumentation with plate and screw fixation, and after implant removal.

Development and validation of interpretable machine learning models to predict 30-day mortality in patients with intracerebral hemorrhage.

Intracerebral hemorrhage (ICH) carries high early mortality. To enable personalized decision-making, we developed and validated interpretable machine learning models for 30-day mortality prediction. This retrospective cohort study included patients with ICH extracted from the Medical Information Mart for Intensive Care (MIMIC) clinical database. Model development was performed using the MIMIC-IV (v3.1), while temporal validation was conducted on a distinct, non-overlapping cohort from the MIMIC-III CareVue subset (v1.4). Multiple imaging-free machine learning models were developed using routin

A shape completion model for corrective osteotomy of distal radius malunion.

PURPOSE: When performing 3D planning for osteotomies in patients with distal radius malunion, the contralateral radius is commonly used as a template for reconstruction. However, in approximately 10% of the cases, the contralateral radius is not suitable for use. A shape completion model may provide an alternative by generating a healthy radius model based on the proximal part of the malunited bone. The aim of this study is to develop and clinically evaluate such a shape completion model. METHOD: A total of 100 segmented CT scans of healthy radii were used, with 80 scans used to train a stati
