Nickel-Catalyzed Cross-Dehydrogenative Allylation of Aldehydes.

Nucleophilic additions to aldehydes play a central role in synthetic chemistry. While additions of organometallic nucleophiles are commonly employed, alternative catalytic processes such as cross-electrophile couplings and reductive couplings involving various π-systems have also been developed and extensively used. In this study, we describe a complementary approach to aldehyde-alkene couplings via direct nickel-catalyzed cross-dehydrogenative coupling of aldehydes and alkenes. The process utilizes a combination of zinc powder and di-tert-butyl peroxide, enabling cross-dehydrogenative couplin

H2S-Activable BiFeO3-x Nanocatalysts for Ferroptosis-Driven Cancer Immunotherapy.

Controlling ferroptosis through material redox dynamics represents a frontier in the field of catalytic nanomedicine. Here, we introduce nonstoichiometric bismuth ferrite (BiFeO3-x) as an H2S-activable semiconductor that couples oxygen-vacancy-enabled charge modulation with endogenous sulfur chemistry to achieve spatiotemporally controlled immunogenic ferroptosis. Upon exposure to tumor-enriched gasotransmitter H2S, BiFeO3-x undergoes a transformation into Bi2S3 while releasing Fe2+ ions to trigger Fenton-like lipid peroxidation. The material's mixed-valence Fe redox pair amplifies reactive ox

Antioxidant properties of tauroursodeoxycholic acid towards free radicals revisited.

Antioxidant properties of tauroursodeoxycholic acid (TUDCA), a cytoprotective bile acid, toward free radicals-induced oxidation of lipids were studied in different model systems using EPR oximetry, EPR spin trapping and the FOX-2 method. Various generators of free radicals were tested: Victoria Blue as a type I sensitizer with minimal generation of singlet oxygen (ΦΔ ≈ 0.2%), hydrophilic (Fe(III)-ADP) and hydrophobic (Fe(III)-8HQ) ferric ion complexes and thermolabile, hydrophilic azo compound (AAPH). The rate of free radicals-induced lipid peroxidation was shown to be reduced by TUDCA in the

Targeting Ferroptosis in Cardiovascular Diseases: The Emerging Role of Traditional Chinese Medicine.

Ferroptosis is a distinct form of regulated cell death (RCD) characterized primarily by iron dysregulation and lipid peroxidation. Emerging evidence indicates ferroptosis critically contributes to the initiation and progression of multiple cardiovascular diseases (CVDs), including atherosclerosis (AS), myocardial ischemia/reperfusion (MI/R) injury, myocardial infarction (MI), cardiomyopathy, and heart failure (HF). With growing recognition of ferroptosis's role in CVD pathogenesis, targeting ferroptosis has become a promising therapeutic strategy for these diseases. Traditional Chinese medicin

A novel microfluidic multichannel electrochemical cell for multiplexed monitoring of water pollutants.

A novel microfluidic multichannel (4×) electrochemical cell (MMEC) was developed and used for multiplexed determination of compounds related to water quality. These include heavy metals (lead and mercury ions), catechol and hydrogen peroxide. No crosstalk between the channels of the MMEC was observed. This enabled the specific and independent modification of each MMEC channel with respect to the targeted analyte. Namely, the mercury ions were determined at the bare gold (Au) electrode, lead ions were determined at the Au electrode coated with a thin mercury film (MF), H2O2 was determined at th

Ferroptosis-Centered Mitochondria-ROS Loop Drives PFOS-Induced Renal Dysfunction, from Epidemiological Evidence to Mechanistic Insights with Mice Model.

Perfluorooctanesulfonate (PFOS) exhibits systemic toxicity, yet its renal impact and mechanism remains unclear. In 296 matched pairs, serum PFOS, perfluorooctanoic acid (PFOA), and five other polyfluoroalkyl substances (PFAS) were significantly elevated and positively correlated with the risk of reduced estimated glomerular filtration rate (eGFR). In vivo, PFOS caused pathological alterations in mouse renal tissue with inflammatory changes, collagen fibrillar hyperplasia, and decreased renal function. PFOS resulted in damaged mitochondria-mediated oxidative stress and lipid peroxidation, enhan

SLC7A11-FUNDC1 axis drives Cr(VI)-Induced renal injury through mitophagy-ferroptosis crosstalk.

Hexavalent chromium [Cr(VI)] is a widespread environmental contaminant known to cause severe organ damage, with acute exposure leading to significant nephrotoxicity. To elucidate the underlying mechanisms, this study investigated the role of the mitophagy-ferroptosis axis in Cr(VI)-induced renal injury using mouse models and renal tubular epithelial cells (mRTECs). We found that Cr(VI) exposure disrupted mitochondrial iron homeostasis in mRTECs, leading to Mito-Fe2+ accumulation and mitochondrial damage. Consequently, this triggered an overproduction of mitochondrial and total reactive oxygen

MYBL2 Knockdown Enhances Ferroptosis and Bortezomib Sensitivity by Transcriptionally Regulating CDKN3 and Inactivating PI3K/Akt Signaling in Multiple Myeloma.

MYBL2 is an oncogene that has been found to be upregulated in diverse cancers. However, the role of MYBL2 in multiple myeloma (MM) remains unclear. Here we used bioinformatic analysis to identify the differentially expressed genes in MM. In this study, western blot was conducted to detect protein levels. Ferroptosis was evaluated by detecting reactive oxygen species (ROS), ferrous iron (Fe2+), lipid peroxidation, and glutathione (GSH) levels. Cell viability and apoptosis were detected by the cell counting kit-8 assay and flow cytometry, respectively. Luciferase reporter assay was performed to

Discovery of Trolox amide derivatives as potent membrane-targeted ferroptosis inhibitors for corneal injury therapy.

Vitamin E and its water-soluble analogue Trolox are potent radical scavengers but require high concentrations to inhibit ferroptosis effectively. The discovery of a Trolox amide derivative 1 showing 95-fold greater potency inspired the systematic synthesis of a focused library. Most amide derivatives demonstrated significantly enhanced potency, with IC50 values in the sub-micromolar range. Notably, compound 20, which features an indazole moiety, emerged as the most potent inhibitor, exhibiting a 556-fold improvement in activity (IC50 = 0.036 μM) over Trolox and a favorable safety index. 4-HDHA, a DHA metabolite screened by targeted lipidomic, alleviates DSS-induced colitis by inhibiting apoptosis and reducing inflammation.

Ulcerative colitis (UC) remains a global health challenge, with current therapeutic strategies limited by efficacy and adverse effects. In this study, we observed that the lipid peroxide scavenger Liproxstatin-1 unexpectedly promoted TNF-α-induced apoptosis in HT-29 cells and murine colonic organoids, in contrast to the protective effects previously reported for typical peroxide scavengers in UC. To identify alternative protective metabolites, we performed targeted lipidomics, which revealed 4-hydroxy-docosahexaenoic acid (4-HDHA), a metabolite of DHA, as a candidate with pronounced anti-apopt

SCD1 protects acinar cells from ferroptosis during severe acute pancreatitis by balancing lipid and redox homeostasis.

During severe acute pancreatitis (SAP), the circulating free fatty acid levels are elevated, and this elevation correlates with a poor prognosis. However, the precise mechanisms by which lipid metabolism influences the progression of SAP remain unclear. Considering that stearoyl-CoA desaturase (SCD1) is a key lipid-modifying enzyme, we investigated its role in SAP. Using caerulein and ligation-induced SAP models along with RNA sequencing, we found that SCD1 is upregulated in SAP models. Through studies with Scd1 knockout mice and acinar cells, we determined that the absence of SCD1 exacerbates

Targeting JMJD6/PPARγ/GPX4 axis overcomes ferroptosis resistance and enhances therapeutic efficacy in hepatocellular carcinoma.

Despite ferroptosis induction being a promising strategy for hepatocellular carcinoma (HCC), its clinical application is limited by intrinsic resistance mechanisms. Through CRISPR-Cas9 screening of epigenetic regulators, we identified JMJD6 as a critical mediator of ferroptosis resistance in HCC. JMJD6 knockdown or pharmacological inhibition (iJMJD6) enhanced ferroptosis induced by ferroptosis inducers (erastin and RSL3), as indicated by decreased cell viability, reduced intracellular glutathione levels, increased lipid peroxidation, and disrupted mitochondrial cristae morphology, thereby prom

Tannic Acid-Based Injectable Hydrogel Promotes the Recovery of Traumatic Brain Injury by Anti-Ferroptosis.

Traumatic brain injury (TBI) can trigger a series of complex physiological responses, with iron overload and neuronal ferroptosis playing particularly pivotal roles. These processes exacerbate secondary brain injury and significantly deteriorate neurological function. To address this challenge, this study developed an innovative local drug delivery strategy: an injectable, post-traumatic microenvironment-responsive hydrogel. The hydrogel, composed of tannic acid (TA) and quaternized chitosan (QCS), is designed to alleviate neurological deficits secondary brain injury following TBI through its

Natural polyphenol rutin inhibits ferroptosis in ulcerative colitis by regulating lipid metabolism via blocking the LCN2-ALOX15 axis.

Ulcerative colitis (UC), a chronic inflammatory disorder with elevated carcinogenic risk, urgently requires multi-target therapeutic strategies. Ferroptosis, characterized by iron-dependent lipid peroxidation, emerges as a promising intervention target for intestinal barrier protection. Rutin, a natural polyphenol with established anti-inflammatory properties, holds therapeutic potential for UC, yet its mechanisms involving ferroptosis regulation and barrier enhancement remain unexplored. This study elucidates rutin's novel role in mitigating UC through ferroptosis inhibition via lipid metabol

Strategy of "Controllable Ions Interference" for Boosting MRI-Guided Ferroptosis Therapy of Tumors.

Chemotherapy for oral squamous cell carcinoma (OSCC) is often marred by the development of multidrug resistance and systemic adverse effects. Metal ion interference therapy (MIIT) has risen as an innovative strategy to disrupt the intracellular metal ion equilibrium in tumor cells, potentially overcoming drug resistance. However, the effectiveness of cancer treatment that relies on delivering single metal ions to tumor site is often constrained. To address this, we have developed a therapeutic nanoplatform employing hollow mesoporous manganese dioxide nanoparticles (HMON) which harness the che

Pathophysiology, biological models and new therapeutic approaches in β-Propeller Associated Neurodegeneration.

Neurodegeneration with brain iron accumulation (NBIA) encompasses a set of rare disorders that present a diagnostic challenge due to their genetic and clinical diversity, with treatment options currently limited to symptom alleviation. One subtype, β-Propeller Associated Neurodegeneration (BPAN), results from pathogenic variants in the WDR45 gene on the X chromosome. This condition is marked by iron accumulation in the globus pallidus and substantia nigra, alongside early onset developmental delays, seizures, and motor impairments. The WDR45 gene produces the WDR45 protein, which plays a key r

Lipid Oxidation at the Crossroads: Oxidative Stress and Neurodegeneration Explored in Caenorhabditis elegans.

Lipid metabolism plays a critical role in maintaining cellular integrity, especially within the nervous system, where lipids support neuronal structure, function, and synaptic plasticity. However, this essential metabolic pathway is highly susceptible to oxidative stress, which can lead to lipid peroxidation, a damaging process induced by reactive oxygen species. Lipid peroxidation generates by-products that disrupt many cellular functions, with a strong impact on proteostasis. In this review, we explore the role of lipid oxidation in protein folding and its associated pathological implication
