Beyond the Clinic: A Social Media Network Analysis of 5466 Posts Reveals the Interconnected Challenges Facing Sarcoma Patients.

OBJECTIVES: Social media platforms have increasingly been investigated as a source of patient perspectives that may not emerge in clinical settings. This study aimed to explore how disease status, treatment, and the patient experience interconnect for sarcoma patients posting on a major social media platform. METHODS: We conducted a systematic thematic analysis of 5466 posts across 7 health-related subreddits using a framework of 6 major categories: physical symptoms, disease status, treatments, psychosocial impact, support systems, and daily life impact. Theme detection utilized regular expr

Antibiotic therapy for severe bacterial infections.

BACKGROUND: Early antibiotic therapy for patients with severe infections is essential to improve outcomes. Conversely, use of overly broad antibiotic therapy for susceptible pathogens or unnecessary antibiotics in patients without bacterial infections is associated with adverse life-threatening events and superinfections. Antibiotics-induced changes in the human microbiota alter both immune and metabolic systems. Uncontrolled antibiotic use encourages emergence of antibiotic-resistant organisms. Around 50% of ICU patients receiving antibiotic therapy do not have confirmed infections, whilst de

Does exposure to zinc, methomyl, and perfluorooctanoic acid alter feeding behavior on MUG® in the freshwater amphipod Gammarus fossarum?

Feeding rate alteration is one of the first observed responses when animals are exposed to toxic stress and is recognized as a relevant tool for studying chemical compounds toxicity. However, food substrates that are currently used for ecotoxicity tests are not always easily available compared with referenced products. Using the European freshwater amphipod Gammarus fossarum, we here propose a standardized food substrate fabricated with referenced ingredients: the MUG® (meal unit for gammarid) for ecotoxicity tests. To investigate the suitability of using MUG to study behavioral response of am…

Neuroprotective effects of ceftriaxone after severe traumatic brain injury in male rats: a behavioral, biochemical, and histological study.

BACKGROUND: Only a limited number of animal studies have demonstrated the neuroprotective effects of single dose of ceftriaxone (CTX) in traumatic brain injury (TBI). Consequently, the present study seeks to fill these research gaps by examining the impact of CTX on neurological and motor performance, brain edema, and blood-brain barrier (BBB) permeability in a rat model of severe diffuse TBI. METHODS: Ninety-eight male Albino Wistar rats were subjected to TBI using the Marmarou method. The rats were divided into seven groups, each consisting of 14 animals: Intact, Sham, TBI, Vehicle (TBI + s

New is not always better: Toxicity of novel copper based algaecides to Daphnia magna.

In this study, the effects of traditional copper (CuSO4.5H2O) and novel copper algaecides (Captain XTR, SeClear and Lake Guard Blue) were tested on Daphnia magna under acute (48 h) and chronic (21 d) exposure scenarios. The EC50 values calculated in the acute tests were between 0.5 and 0.6 mg Cu L-1 for all four compounds. Lake Guard Blue and CuSO4.5 H2O were more toxic than SeClear and Captain XTR. During the chronic test, the effects of SeClear (EC50: 0.274 mg Cu L-1) on reproduction and body length were larger than the effects of the other three copper-based algaecides (EC50: 0.436 mg Cu L-

Tuberculosis and HIV-An Update on the "Cursed Duet" in Children.

HIV and tuberculosis (TB) often occur together with each exacerbating the other. Improvements in vertical transmission prevention has reduced the number of HIV-infected children being born and early antiretroviral therapy (ART) protects against tuberculosis. However, with delayed HIV diagnosis, HIV-infected infants often present with tuberculosis co-infection. The number of HIV exposed uninfected children has increased and these infants have high exposure to TB and may be more immunologically vulnerable due to HIV exposure in utero. Bacillus Calmette-Guérin (BCG) immunization shortly after bir

Thermogelling chitosan lactate hydrogel improves functional recovery after a C2 spinal cord hemisection in rat.

The present study was designed to provide an appropriate micro-environment for regenerating axotomized neurons and proliferating/migrating cells. Because of its intrinsic permissive properties, biocompatibility and biodegradability, we chose to evaluate the therapeutic effectiveness of a chitosan-based biopolymer. The biomaterial toxicity was measured through in vitro test based on fibroblast cell survival on thermogelling chitosan lactate hydrogel substrate and then polymer was implanted into a C2 hemisection of the rat spinal cord. Animals were randomized into three experimental groups (Cont

Clinical efficacy and management of monoclonal antibodies targeting CD38 and SLAMF7 in multiple myeloma.

Immunotherapeutic strategies are emerging as promising therapeutic approaches in multiple myeloma (MM), with several monoclonal antibodies in advanced stages of clinical development. Of these agents, CD38-targeting antibodies have marked single agent activity in extensively pretreated MM, and preliminary results from studies with relapsed/refractory patients have shown enhanced therapeutic efficacy when daratumumab and isatuximab are combined with other agents. Furthermore, although elotuzumab (anti-SLAMF7) has no single agent activity in advanced MM, randomized trials in relapsed/refractory M

Chromium removal from aqueous solution by the ferrite process.

This research summarises the results of the study on the removal of chromium by applying the ferrite process to the solutions obtained from two different Cr(VI) reduction processes utilising sodium sulphite and ferrous sulphate as reducing agents. For both solutions containing trivalent chromium ions, the optimum treatment conditions were determined. The generated sludges were characterised by XRD analysis and physical tests. In addition, to explore the dissolution properties of the sludges obtained, they were contacted with the solutions of sulphuric, citric, tartaric, oxalic and ascorbic aci

Monitoring acrylic fiber workers for liver toxicity and exposure to dimethylacetamide. 2. Serum clinical chemistry results of dimethylacetamide-exposed workers.

Worker exposure to N,N-dimethylacetamide (DMAC) in an acrylic fiber manufacturing facility was measured, over a 1-year study period, by full-shift (12 hours) personal air monitoring for DMAC and biological monitoring for levels of DMAC, N-methylacetamide (MMAC) and acetamide in post-shift spot urine samples. Evidence of liver toxicity was assessed by serum clinical chemistry tests (serum levels of total bilirubin, aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase, and gamma-glutamyl transpeptase) at least once during the study period for all 127 male workers in the two

Correlations between common tests for assessment of liver damage: indices of the hepatoprotective activity of promethazine in carbon tetrachloride hepatotoxicity.

The effects of promethazine (PM) on different aspects of the hepatotoxic action of CCl4 in the rat were investigated with the objective of finding rapid and reliable indicators of hepatoprotective effects. The study was based on definitive histological assessment of liver damage caused by CCl4 in the presence and absence of PM: PM (78 mumol kg-1, i.p.) protected against CCl4-induced hepatic necrosis 24 h after a low dose of CCl4 (1.3 mmol kg-1) but not against a higher dose (13.0 mmol kg-1). The large increases in plasma activities of GOT, GPT and LDH produced by dosing with CCl4 were partiall

Survey of genes encoding staphylococcal enterotoxins, toxic shock syndrome toxin 1, and exfoliative toxins in members of the Staphylococcus sciuri group.

Genes encoding staphylococcal enterotoxins (sea to see, seg, and seh), toxic shock syndrome toxin 1 (tst), and exfoliative toxins (eta and etb) were not detected in a large panel of 48 Staphylococcus sciuri group isolates tested. This strongly suggests that production of the staphylococcal exotoxins by these bacteria is highly unlikely.

Protective role of TGF-β1 (-509C>T, rs1800469) single-nucleotide polymorphism against radiation toxicities in locally advanced head and neck cancer.

BACKGROUND: Head and neck cancer (HNC), predominantly head and neck squamous cell carcinoma (HNSCC), poses a significant health burden globally. Intensity-modulated radiation therapy (IMRT) has improved outcomes in HNC; however, radiation-induced toxicities, particularly oral mucositis and dermatitis, remain critical concerns. Genetic variations, such as single-nucleotide polymorphisms (SNPs) in the TGF-β1 gene (-509C>T, rs1800469), may modulate these toxicities and impact treatment response. MATERIALS AND METHODS: This observational analytical study included 50 patients with locally advanced

Natural Language Processing to Automatically Extract the Presence and Severity of Esophagitis in Notes of Patients Undergoing Radiotherapy.

PURPOSE: Radiotherapy (RT) toxicities can impair survival and quality of life, yet remain understudied. Real-world evidence holds potential to improve our understanding of toxicities, but toxicity information is often only in clinical notes. We developed natural language processing (NLP) models to identify the presence and severity of esophagitis from notes of patients treated with thoracic RT. METHODS: Our corpus consisted of a gold-labeled data set of 1,524 clinical notes from 124 patients with lung cancer treated with RT, manually annotated for Common Terminology Criteria for Adverse Event

Safety and tolerability of isoniazid preventive therapy for tuberculosis for persons with HIV with and without alcohol use.

OBJECTIVE: Isoniazid (INH) preventive therapy is recommended to prevent tuberculosis (TB) disease for persons with HIV (PWH), except for those with regular and heavy alcohol consumption, due to hepatotoxicity concerns. We aimed to quantify the incidence of severe INH-related toxicity among PWH with and without recent alcohol consumption. DESIGN: A prospective study of PWH receiving INH. METHODS: We included PWH in southwest Uganda with recent (prior 3 months) ( n  = 200) or no (prior year) self-reported alcohol consumption ( n  = 101), on antiretroviral therapy, TB infected (≥5 mm on tubercu

Quantitative in vitro to in vivo extrapolation for developmental toxicity potency of valproic acid analogues.

BACKGROUND: The developmental toxicity potential (dTP) concentration from the devTOX quickPredict (devTOXqP ) assay, a metabolomics-based human induced pluripotent stem cell assay, predicts a chemical's developmental toxicity potency. Here, in vitro to in vivo extrapolation (IVIVE) approaches were applied to address whether the devTOXqP assay could quantitatively predict in vivo developmental toxicity lowest effect levels (LELs) for the prototypical teratogen valproic acid (VPA) and a group of structural analogues. METHODS: VPA and a series of structural analogues were tested with the devTOXq

International Survey on the Use of Complementary and Alternative Medicines for Common Toxicities of Radiation Therapy.

PURPOSE: Complementary and alternative medicines (CAMs) are widely used by patients with cancer. However, little is known about the extent to which these potential remedies are used internationally to treat the most common toxicities of radiation therapy. We report on the results of an international survey that assessed the use of CAMs. METHODS AND MATERIALS: Surveys were distributed to 1174 practicing radiation oncologists. Questions evaluated the perceptions of CAMs and specific practice patterns for the use of CAM remedies in the treatment of common radiation-induced toxicities (eg, skin,

Evaluation of dicloran phototoxicity using primary cardiomyocyte culture from Crassostrea virginica.

Dicloran is a commonly used fungicide throughout the Southern and Western United States. Runoff of dicloran from agriculture systems to nearby waterbodies can accumulate in the organisms that inhabit those areas. Although severe damage of dicloran to ecological systems has not been reported, its toxicity has been modified by photodegradation. The objective of this study is to assess the changes of dicloran toxicities during photo exposure using a reliable in vitro biological model. In the present investigation, the photodegradation of dicloran in vitro showed over 90% of dicloran was degraded
