Autoimmune aspects of depigmentation in vitiligo.

Autoimmune depigmentation of the skin, vitiligo, afflicts a considerable number of people, yet no effective therapeutic modalities have been developed to treat it. In part, this can be attributed to the obscure etiology of the disease, which has begun to reveal itself only recently. It is known that pigment is lost as a function of reduced melanocyte numbers in the epidermis, and that depigmentation is accompanied by T cell influx to the skin in the vast majority of patients. Characterizing such infiltrating T cells as type 1 proinflammatory cytokine-secreting cells reactive with melanocyte-sp

Deregulation of the expression of the fractalkine/fractalkine receptor complex in HIV-1-infected patients.

Fractalkine is the only member of the CX3C chemokine family. Polymorphism of the fractalkine receptor gene may influence the prognosis of human immunodeficiency virus (HIV) infection, but the nature of the cells expressing fractalkine or its receptor in HIV-infected patients remains unknown. We show that, in contrast to HIV-uninfected individuals, a large number of cells expressed fractalkine in T-cell zones of lymph nodes from HIV-infected patients. CD83(+) mature and CD123(+) plasmacytoid dendritic cells as well as plasma cells are involved in this increased expression of fractalkine. Increa

Angioimmunoblastic T-cell lymphoma with hyperplastic germinal centers: a clinicopathological and immunohistochemical study of 10 cases.

An absence of germinal centers is one of the histological characteristics of angioimmunoblastic T-cell lymphoma (AITL). We report here 10 unusual cases of AITL with hyperplastic germinal centers. The clinical presentation of each patient was characterized by generalized lymphadenopathy, constitutional symptoms and polyclonal hypergammaglobulinemia. The initial biopsy findings of each patient were similar and were characterized by hyperplastic germinal centers with ill-defined borders and a proliferation of high endothelial venules (HEV). In the paracortical area there was a mixed infiltrate in

Increased immunoglobulin E Fc receptor bearing cells in germinal centers of hyperimmunoglobulinemia E patients.

Lymph nodes from 6 patients with hyperimmunoglobulinemia E (hyper-IgE; 4 with Kimura's disease, 1 with atopic dermatitis, and 1 with immunoblastic lymphadenopathy-like T cell lymphoma) and from 7 patients with normal IgE levels were studied to determine the localization of dendritic reticulum cells and of cells bearing Fc epsilon and C3d receptors and immunoglobulin E. The avidin-biotin-glucose oxidase method was used for unfixed biopsy specimens. To identify the above-mentioned cells, H107, a murine monoclonal antibody specific to the Fc epsilon receptor molecule, and corresponding antibodies

Innervation of the iris by individual parasympathetic axons in the adult mouse.

Ciliary neurones were identified electrophysiologically in isolated ganglia and the projection of individual axons to the intrinsic muscles of the eye determined by labelling identified cells with horseradish peroxidase (HRP). The ciliary ganglion of the adult mouse contains 80-100 parasympathetic neurones clustered at the point where the oculomotor nerve branches to supply the medial and inferior rectus. These nerve cells show varying degrees of dendritic complexity (zero to six primary dendrites) and differing degrees of innervation by preganglionic axons (one to seven inputs). Cells with mo…

A quantitative analysis of the laminar distribution of synaptic boutons in field CA3 of the rat hippocampus.

We analyzed the laminar distribution of synaptic boutons in field CA3 of the rat hippocampus using a large montage electron micrograph. The size of boutons and synaptic vesicles was measured using a computer-assisted digitizing system. In all, 3353 synaptic boutons were observed in a 15 microm x 100 microm strip. Of these, 86.3% contained spherical vesicles (S-boutons), 12% contained flat vesicles (F-boutons), and 1.7% were mossy terminals (M-boutons). S-boutons were distributed widely in the strata moleculare (st. Mol), radiatum (st. Rad), and oriens (st. Ori), but there were only a few in th

[Ultrastructural relief of unaffected mucosa in recurrent oral ulcer patients. 2].

Electron microscopy was performed on clinically normal vestibular mucosa biopsies of two patients affected with recurrent oral ulcers. Apoptotic cells were absent from vestibular mucosa; this is in contrast with the pathogenetic theory proposed by Honma et al., 1985 (1). Only a few modifications in the keratinization process (intracytoplasmic desmosomes, electrondense granules in basal keratinocytes) and an unusually high number of dendritic cells were found; dendritic cells could be involved in local immunological iperreactivity, responsible for the ulcers of these subjects.

Purkinje cell intrinsic activity shapes cerebellar development and function.

The emergence of functional cerebellar circuits is heavily influenced by activity-dependent processes. However, the contribution of intrinsic Purkinje cell activity to cerebellar development remains less understood. Here, we demonstrate that before synaptic networks mature, Purkinje cell intrinsic activity is essential for regulating dendritic growth, establishing connections with cerebellar nuclei, and ensuring proper cerebellar function. Disrupting this activity during the postnatal period impairs motor function, with earlier perturbations causing more severe deficits. Importantly, only earl

Early chronic fasudil treatment rescues hippocampal alterations in the Ts65Dn model for down syndrome.

Down syndrome (DS) is the most common genetic disorder associated with intellectual disability. To study this syndrome, several mouse models have been developed. Among the most common is the Ts65Dn model, which mimics most of the alterations observed in DS. Ts65Dn mice, as humans with DS, show defects in the structure, density, and distribution of dendritic spines in the cerebral cortex and hippocampus. Fasudil is a potent inhibitor of the RhoA kinase pathway, which is involved in the formation and stabilization of dendritic spines. Our study analysed the effect of early chronic fasudil treatm

Intrinsic and synaptic determinants of receptive field plasticity in Purkinje cells of the mouse cerebellum.

Non-synaptic ('intrinsic') plasticity of membrane excitability contributes to aspects of memory formation, but it remains unclear whether it merely facilitates synaptic long-term potentiation (LTP), or whether it plays a permissive role in determining the impact of synaptic weight increase. We use tactile stimulation and electrical activation of parallel fibers to probe intrinsic and synaptic contributions to receptive field (RF) plasticity in awake mice during two-photon calcium imaging of cerebellar Purkinje cells. Repetitive activation of both stimuli induced response potentiation that is i

Evaluation of various adjuvant nanoparticulate formulations for meningococcal capsular polysaccharide-based vaccine.

Neisseria meningitidis is a leading cause of bacterial meningitis and sepsis and its capsular polysaccharides (CPS) are a major virulence factor in meningococcal infections and form the basis for serogroup designation and preventive vaccines. We have formulated a novel meningococcal nanoparticulate vaccine formulation that does not require chemical conjugation, but encapsulates meningococcal CPS polymers in a biodegradable material that slowly release antigens, thereby has antigen depot effect to enhance antigenicity. The novel vaccine formulation is inexpensive and can be stored as a dry powd

Cytokeratin-positive interstitial reticulum cell sarcoma: extranodal presentations mimicking carcinoma.

Cytokeratin-positive interstitial reticulum cell (CIRC) sarcoma is a rare type of dendritic cell tumor derived from a subset of fibroblastic reticular cells. Expression of cytokeratins and extranodal location of these tumors can lead to a misdiagnosis of carcinoma. We report two cases of CIRC sarcomas primarily involving the scalp and breast. Patients were referred with an initial diagnosis of carcinoma. Case 1 underwent wide local excision of the scalp tumor with left posterolateral neck dissection. Case 2 had modified radical mastectomy for the tumor in left breast. Histopathological examina

Occurrence of giant cell arteritis...suddenly.

PURPOSE: To define the kinetics and mechanisms of frank arteritis onset in patients with giant cell arteritis. METHODS: Cytokines were analyzed from tissue of a patient before and after the development of arteritis. RESULTS: A temporal artery biopsy specimen from a patient with giant cell arteritis showed no pathologic changes on microscopic examination, but there was evidence of early tissue activation of inflammatory markers. A specimen from the contralateral artery 12 days later had high levels of IL-18 transcripts and abundant transcripts for CCL19. Also, CD83 and IL-1 were present, conf

Differential effects of nicotine and complex housing on subsequent experience-dependent structural plasticity in the nucleus accumbens.

Drugs and other forms of experience (e.g., complex housing) share the ability to alter the dendritic fields of cortical and subcortical neurons. Although such modifications are typically considered advantageous, recent research has demonstrated that psychomotor stimulants (cocaine and amphetamine) block subsequent experience-dependent structural plasticity in the nucleus accumbens (NAcc) and parietal neocortex. The authors investigated whether these findings generalize to another commonly used stimulant (nicotine) and further asked whether prior experience blocks subsequent nicotine-related st

Distribution of immunocompetent cells in various areas in the normal laryngeal mucosa of the rat.

The larynx can be divided into a supraglottic, a glottic and a subglottic area, each serving different functions. In many cases of laryngitis the site of infection is located in one area, leaving other areas unaffected. It seems reasonable to speculate that the underlying cause of the heterogeneous infection pattern in the larynx is the different processing of infectious agents. Therefore, the number and distribution of granulocytes, macrophages, dendritic cells, natural killer cells and T and B lymphocytes in the normal laryngeal mucosa of young rats were studied. The results show that, with

Ultrastructural studies of microtubules and microtubule organizing centers of the vertebrate olfactory neuron.

The olfactory neuron is specialized along its length into highly determined morphological regions. These regions include the dendritic cilia, dendritic vesicle, dendritic shaft proper, perikaryon, axon, zone of transition where the axon widens as it approaches its termination, and the axon terminal. Except for the zone of transition and the terminal, characteristic populations of microtubules occur in these compartments. In the olfactory vesicle, three discrete microtubule organizing centers (MTOCs) nucleate microtubules: the basal body, the lateral foot associated with the body, and dense mas

Altered stimulus-intensity encoding occurs in the primary somatosensory cortex of a central post-stroke pain rat model.

Central post-stroke pain (CPSP) is a chronic form of pain that develops after ventral posterolateral thalamus (VPL) lesions and may involve maladaptive plasticity within central somatosensory pathways. Although abnormalities in the primary somatosensory cortex (S1) are observed in patients and animal models, the mechanism by which thalamic lesions alter cortical processing of sensory stimuli remains unclear. Here, we investigated stimulus intensity encoding and dendritic remodeling in S1 using a VPL-lesion CPSP rat model. Behavioral assessments were conducted, and mechanical allodynia was obse

Nanostructured branched Y-DNA promotes antitumor immunity through dual activation of cGAS/STING and TLR9.

Effective activation of innate and adaptive immunity remains a major challenge in cancer immunotherapy. In this study, we aimed to develop an immunostimulatory oligonucleotides for antitumor immunotherapy. We designed branched Y-shaped DNA (Yb-DNA) nanoparticles complexed with a reducible polycationic carrier (RPC-bPEI0.8kDa), which we named YbNano, to enhance intracellular delivery and immune activation. The immunostimulatory activity of YbNano was determined in mouse bone marrow-derived dendritic cells. The efficacy of YbNano for antitumor immunotherapy was investigated using a melanoma allo
