Extracellular Vesicles Derived from Elaeocarpus braceanus Alleviate DSS-Induced Ulcerative Colitis in Mice Through Multiple Pathways.

AIM OF THE STUDY: This study aims to isolate extracellular vesicles derived from Elaeocarpus braceanus fruits (EBDEVs) and evaluate their alleviating efficacy as nature nanoparticles against dextran sulfate sodium (DSS)-induced ulcerative colitis (UC). METHODS: EBDEVs were isolated by differential and density gradient ultracentrifugation, then characterized for morphology, size, stability, and composition. Their anti-inflammatory activity was assessed in LPS-stimulated RAW264.7 macrophages. In vivo, acute UC was induced in C57BL/6 mice by 2.5% DSS. Disease severity, intestinal barrier integri

Androgen loss accelerates brain tumour growth via HPA axis activation.

Many cancers, including glioblastoma (GBM), show a male-biased incidence and associated worse outcomes1. The mechanisms that underlie this sex difference remain unclear but may involve an immune response2 that is partly driven by sex hormones such as androgens. Such hormones are thought to suppress antitumour T cell immunity and to promote tumour progression3,4. However, here we report a previously unreported tumour-suppressive role for androgens in brain tumours. Using mouse models, we demonstrate that androgen loss via castration accelerates intracranial tumour growth, whereas the opposite e

The Role of Metals and Trace Elements in the Pathogenesis of Osteoarthritis and Other Rheumatic Diseases.

Osteoarthritis (OA) and other rheumatic diseases are among the leading causes of chronic pain, disability, and reduced quality of life worldwide. Increasing evidence indicates that disturbances in metal homeostasis contribute to the pathogenesis of these disorders through their effects on oxidative stress, immune regulation, cartilage metabolism, bone remodeling, and extracellular matrix degradation. This review summarizes current knowledge regarding the role of essential metals and trace elements, including zinc, iron, copper, magnesium, selenium, and manganese, as well as toxic metals such a

Pituitary adenylate cyclase-activating polypeptide and mitochondrial homeostasis in neuronal injury: mechanisms and translational prospects.

Pituitary adenylate cyclase-activating peptide (PACAP) is a pleiotropic neuropeptide widely distributed in the nervous system, exhibiting potent cytoprotective effects across a spectrum of neurological disorders. Its neuroprotection is largely mediated through three G protein-coupled receptors (PAC1, VPAC1, VPAC2), activating downstream pathways that converge on preserving mitochondrial integrity. Mitochondrial dysfunction, characterized by bioenergetic failure, oxidative stress, perturbed dynamics (such as fusion and fission), and impaired quality control, is a hallmark of traumatic nerve inj

Glyphosate induces intestinal inflammatory injury associated with mitochondrial-lysosomal dysfunction, cGAS-STING activation, and pyroptosis-related responses in common carp (Cyprinus carpio L.).

Glyphosate (GLY) poses a potential risk to fish health, yet the molecular mechanisms underlying GLY-induced intestinal injury remain unclear. This study established in vivo (common carp midgut) and in vitro (EPC cells) models to investigate the toxic effects of GLY. We evaluated ROS production, mitochondrial and lysosomal function, autophagic flux, cGAS-STING signaling, and pyroptosis via histology, fluorescence staining, RT-qPCR, and western blotting. Both in vivo and in vitro results showed that GLY exposure markedly increased ROS production, induced mitochondrial damage and lysosomal dysfun

Inflammation and Oxidative Stress in Male Infertility: A Narrative Review.

BACKGROUND: Male infertility affects 8%-15% of couples, with male factors implicated in about half of cases; many remain idiopathic despite improved diagnostics. This narrative review addresses this gap by examining chronic inflammation as an underappreciated driver of sperm dysfunction that forms a bidirectional, self-reinforcing cycle with oxidative stress (OS). METHODS: We synthesized studies from 2015 to 2025 identified via PubMed, Scopus, and Web of Science, examining links between inflammation and OS, key immune pathways, and related risk factors through qualitative narrative synthesis.

Lactylation-driven PDLIM1/PDAP1 axis remodels the inflammatory landscape of acute lung injury: mechanistic insights and precision intervention.

INTRODUCTION: Acute lung injury (ALI) remains a lethal clinical challenge driven by an uncontrolled "cytokine storm" resulting from dysregulated inflammatory networks. The metabolic and molecular mechanisms orchestrating this process remain incompletely understood. METHODS: We integrated multi-omics profiling with functional analyses in murine models and human iPSC-derived alveolar organoids. Key molecular players were identified through loss-of-function and pharmacological interventions, and the effects of a dual-target strategy using progesterone (PT) and ethyl methanesulfonate (EMS) were e

The potential connection between inflammatory bowel disease and Alzheimer's disease: mechanistic insights and therapeutic implications.

Inflammatory bowel disease (IBD) and Alzheimer's disease (AD) are two chronic conditions previously considered pathologically unrelated. However, accumulating epidemiological evidence suggests that patients with IBD may have an elevated risk. This review systematically analyzes the mechanistic links between IBD and AD, emphasizing the roles of gut microbiota dysbiosis, systemic and neuroinflammation, and glymphatic dysfunction. We propose a putative conceptual framework incorporating the 'gut-glymphatic axis' as a potential conduit through which IBD-associated microbial and immune perturbation

Role of Nod-like Receptors in Helicobacter pylori Infection: Insights into Innate Immune Signaling Pathways.

Helicobacter pylori is a prevalent gastric pathogen that establishes chronic infection and contributes to gastritis, peptic ulcer disease, and gastric cancer. Its persistence depends on immune evasion strategies that promote sustained low-grade inflammation in the gastric mucosa. Nucleotide-binding oligomerization domain-like receptors (NLRs) are cytosolic pattern recognition receptors that play key roles in innate immune responses against H. pylori. Nod1 and Nod2 detect bacterial peptidoglycan delivered via the type IV secretion system or outer membrane vesicles, activating NF-κB, MAPK, and i

MSC-EVs in Cartilage Regeneration and Immunomodulation: Mechanisms and Therapeutic Prospects for Osteoarthritis, Rheumatoid Arthritis and Intervertebral Disc Degeneration.

Mesenchymal stem cell-derived extracellular vesicles (MSC-EVs) represent a promising cell-free therapeutic strategy for cartilage regeneration and inflammation modulation in degenerative and inflammatory musculoskeletal disorders, including osteoarthritis (OA), rheumatoid arthritis (RA) and intervertebral disc degeneration (IVDD). MSC-EVs enhance cartilage repair by promoting chondrocyte proliferation, migration, survival and extracellular matrix (ECM) synthesis to maintain cartilage homeostasis. In parallel, they exert anti-inflammatory and anti-catabolic effects by suppressing inflammatory c

Liver Fibrosis and Purinergic Signaling: Autocrine-Paracrine Role of ATP in Liver Damage.

Fibrosis is a common extracellular matrix pathology characterized by increased scarring, representing a critical checkpoint toward cirrhosis and hepatocellular carcinoma. Its onset involves coordinated interplay among hepatocytes, Kupffer, and hepatic stellate cells (HSCs). Extracellular ATP and its derivates act as crucial damage-associated molecular patterns when released by injured liver cells, binding to specific purinergic receptors (P2X, P2Y, and P1) to establish an autocrine-paracrine signaling loop. The hepatic fibrotic response underlies the activation of ATP receptors that generate s

NDUFA4L2 regulates the progression and chemotherapy sensitivity of HNSCC by inhibiting PANoptosis.

Head and neck squamous cell carcinoma (HNSCC) represents a leading global malignancy among head and neck cancers. While chemotherapy serves as a standard adjuvant treatment, cisplatin resistance frequently compromises therapeutic outcomes. PANoptosis is an integrated inflammatory cell death pathway governed by PANoptosome complexes. It critically influences chemotherapy response, though its regulatory mechanisms remain incompletely characterized. NADH dehydrogenase (ubiquinone) 1 alpha subcomplex subunit 4-like 2 (NDUFA4L2), a subunit of respiratory chain complex I, has been identified as a cr

Immunomodulatory Nanozymes as Programmable Redox-Immune Set-Point Regulators.

Nanozymes are engineered nanomaterials possessing intrinsic enzyme-mimetic catalytic activities that enable regulation of reactive oxygen species (ROS). Initially developed for superoxide dismutase-, catalase-, and peroxidase-like antioxidant functions, nanozymes are increasingly recognized as modulators of immune responses through redox-dependent mechanisms. Given that ROS participate in immune activation, metabolic regulation, and inflammatory resolution, controlled catalytic modulation offers a strategy for disease-specific immune regulation. This review summarizes recent advances in nanozy

Natural Bioactive Compounds Targeting Gut Barrier Integrity and Metabolic Endotoxemia in Cardiometabolic Disease: Mechanistic Insights and Translational Perspectives.

Cardiometabolic diseases are increasingly recognized as disorders of chronic low-grade systemic inflammation and gut barrier dysfunction that mutually reinforce one another. Each condition amplifies the other through progressive injury to the intestinal epithelium. Compromise of the mucus layer, altered tight junction dynamics, dysbiosis, and impaired epithelial restitution promote intestinal permeability and enable the translocation of lipopolysaccharide and other microbial products into the circulation, thereby inducing metabolic endotoxemia. This gut derived inflammatory signal activates To

Circadian clock-gut microbiota axis in pulmonary hypertension: linking gut-lung crosstalk, immune timing and vascular remodeling.

Pulmonary hypertension (PH) is a severe disorder characterized by progressive pulmonary vascular remodeling, increased pulmonary arterial pressure, and right heart failure. Beyond local abnormalities within the pulmonary vasculature, growing evidence suggests that PH is shaped by systemic inflammation, metabolic dysregulation, gut microbiota imbalance, and disruption of host circadian organization. In this review, we examine the circadian clock-gut microbiota-gut-lung axis as a time-dependent regulatory network in PH. We focus on core clock gene dysfunction, impaired intestinal barrier integri

Mitochondrial Quality Control Links Exercise to Sterile Inflammation in the Cardiovascular System: A Narrative Review.

Preservation of mitochondrial integrity has emerged as a central hub in the anti-inflammatory effect of exercise. This narrative review advances a framework in which mitochondrial damage-associated molecular patterns (mtDAMPs) serve as the mechanistic bridge between exercise and inflammation. Mitochondrial dysfunction releases mtDAMPs, including mitochondrial DNA (mtDNA), reactive oxygen species, cardiolipin, N-formyl peptides, and ATP, which activate cGAS-STING, the NLRP3 inflammasome, TLR9, AIM2, ZBP1, and NF-κB signaling. Crosstalk among these pathways allows mild mitochondrial damage to es

Thapsigargin-induced autophagic flux impairment and inflammation are potentiated by CLN3 deficiency and alleviated by 5-aminoimidazole-4-carboxamide ribonucleoside (AICAR) in human ARPE-19 cells.

Juvenile neuronal lipofuscinosis (JNCL) is a rare disease caused by mutations in the CLN3 gene. It leads to early vision loss mediated by retinal degeneration. Impaired autophagosomal-lysosomal degradation is a major hallmark of JNCL pathology, and neuroinflammation has also been postulated to play a role in its pathogenesis. Thapsigargin, a selective inhibitor of sarco/endoplasmic reticulum Ca2+-ATPase, inhibits autophagy, leading to an accumulation of autophagosomes/autophagophores in cells. Cells with defective CLN3 protein function have been found to be particularly sensitive to the anti-a

Therapeutic Potential of Berberine in Obesity-Associated Neuroinflammation Through Shared Molecular Pathways: TLR4/NF-κB/MAPK, ROS/NRF2 and NLRP3.

Obesity is recognized as a pathological condition that induces chronic low-grade systemic inflammation capable of affecting multiple organs, including the central nervous system, thereby promoting neuroinflammation. Although adipose and neural tissues differ in their structural and functional characteristics, they share common inflammatory mechanisms involving the TLR4/NF-κB/MAPK signaling pathways, the ROS/NRF2 axis, and the NLRP3 inflammasome. This review summarizes current evidence demonstrating the ability of berberine (BBR) to modulate these shared molecular pathways across different expe

Key Inflammatory Pathways, Biomarkers, and Targeted Management Strategies in Primary Total Joint Arthroplasty: A Narrative Review.

Total joint arthroplasty is a surgical procedure with rising global incidence. Although a strong postoperative inflammatory response is necessary for tissue repair following primary arthroplasty, it may prove to be harmful if excessive or prolonged. This could compromise osseointegration, increase pain, and delay the detection of periprosthetic joint infection. This narrative review examines the principal inflammatory pathways activated by primary arthroplasty.
