Rapid Phenotypic Drug Susceptibility Testing of Mycobacterium tuberculosis Using Raman-Deuterium Isotope Probing.

Accurate and timely drug susceptibility testing (DST) is essential for tuberculosis (TB) management, especially given the global rise of multidrug-resistant Mycobacterium tuberculosis (MTB). However, conventional phenotypic DST (pDST) is reliable but time-consuming, while molecular assays may fail to detect resistance not associated with known mutations. Here, we report a rapid pDST method using single-cell Raman spectroscopy coupled with deuterium isotope probing (Raman-DIP) to assess MTB metabolic activity when exposed to different antibiotics. The Raman-DIP workflow was specifically optimiz

C-BODIPY Dyads Based on Fused-Ring Organoboron Scaffolds for Efficient Photodynamic Therapy Targeting Candida albicans Yeast and Biofilms.

A series of heavy-atom-free C-BODIPY photosensitizers based on five different types of fused-ring organoboron scaffolds was designed for the application in antifungal photodynamic therapy. The dyes were functionalized with a positively charged quaternary ammonium group or zwitterionic 3-(ethyldimethylammonio)propane-1-sulfonate unit. Spectroscopic, electrochemical, and photocatalytic studies supported by DFT calculations revealed the crucial role of the organoboron structure on triplet-state generation. Most of the BODIPY complexes display high ROS generation with a type I/type II ROS ratio de

Azure A as a promising candidate for antimicrobial photodynamic therapy of acne-related bacteria.

Antimicrobial photodynamic inactivation (aPDT) is an effective technique for treating acne vulgaris, due to its efficiency in inactivating multiresistant bacteria and the absence of systemic side effects. This study investigated the potential of the dye Azure A (AZA), belonging to the phenothiazine class, as a photosensitizer (PS) in aPDT to combat the gram-positive bacteria Staphylococcus epidermidis and Cutibacterium acnes, both associated with inflammatory acne vulgaris. The efficiency of AZA was compared to Orthotoluidine Blue (OTB), evaluating the absorbance spectra, singlet oxygen genera

Pharmacokinetic-pharmacodynamic modeling to evaluate the relative impact of immune response and meropenem on bacterial killing in vivo.

In vivo antibiotic pharmacokinetic-pharmacodynamic (PKPD) properties are typically studied in neutropenic infection models, limiting the understanding of interactions between immune cells and antibiotics. This study aimed to characterize the impact of immune status on meropenem PKPD and dose-response in a mouse lung infection model, and to quantify the relative contribution of immune response and meropenem to bacterial killing. Meropenem PK was analyzed in plasma and epithelial lung fluid, and bacterial counts were monitored over 24 h following 40 or 300 mg/kg meropenem doses every 4 h in a mo

Papiliotrema laurentii fungemia in a neutropenic patient with limited echinocandin susceptibility: a case report.

Papiliotrema laurentii is a rare yeast species and an uncommon cause of human infection, and optimal management strategies have not been established. We report a case of P. laurentii fungemia in a 77-year-old male farmer who developed febrile neutropenia during chemotherapy for pancreatic cancer. Blood cultures obtained on two separate occasions yielded yeast in one of four bottles; a single isolate was identified as P. laurentii by a reference mycology laboratory using ITS and D1/D2 rRNA gene sequencing. The minimum inhibitory concentration of micafungin (MIC) was ≥16 μg/mL. The patient was i

Synthesis of Furan and Benzene Derivatives Containing Unsaturated Substituents with Electron-Withdrawing Groups, and Analysis of Their Antimicrobial Activity.

Furan-2-yl substituted ethenes and conjugated butadienes containing two geminal electron-withdrawing groups, as well as phenylbutadienes with two electron acceptors, were synthesized from the corresponding aldehydes and various β-dicarbonyl compounds via aldol condensation. Reactions of these ethenylfurans and methyl 3-(benzofuran-2-yl)propenoate with arenes in triflic acid (TfOH) yield arylated 2,3-dihydrofurans and 2,3-dihydrobenzofurans, representing a novel synthetic approach to these dihydro derivatives. In contrast, analogous reactions of furanyl- and phenylbutadienes result in carbon-ca

Emergence of difficult-to-treat lymphocutaneous sporotrichosis? Analysis based on three cases with high and low MIC Sporothrix brasiliensis isolates.

Sporotrichosis, caused by a Sporothrix schenckii complex species, is historically viewed as an easily manageable mycosis. However, the current zoonotic epidemic in Brazil due to the more virulent Sporothrix brasiliensis poses some challenges. We describe three cases of lymphocutaneous sporotrichosis (LC) in non-immunocompromised patients, which proved refractory to standard treatment, and suggest that closer epidemic surveillance would be advisable to detect whether more difficult-to-treat cases are --emerging. All cases required prolonged treatment, escalating doses, and therapeutic changes f

Biofilm formation by clinical Clostridium perfringens isolates and its suppression by thymol.

Necrotic enteritis (NE) caused by Clostridium perfringens imposes major health and economic losses in broiler production, especially with coccidiosis co-infection. Beyond toxin secretion, C. perfringens forms biofilms that enhance persistence and reduce treatment efficacy. Thymol, a phenol from thyme, has broad antimicrobial and antibiofilm activities, but its effects on C. perfringens biofilms are not well defined. We profiled 23 clinical isolates, genotyped them (all cpa⁺, most cpβ2⁺; eight netB⁺; tpeL detected only in LLY_TpeL17), quantified biofilm biomass by 0.1% crystal violet staining (

Linalool exhibits potent antibacterial and anti-biofilm activity against carbapenem-resistant Acinetobacter baumannii.

Acinetobacter baumannii poses an urgent clinical challenge due to its extensive antimicrobial resistance and enhanced capacity for biofilm development. These biofilm structures act as protective barriers that impede antibiotic penetration and shield bacterial cells from immune-mediated clearance, often leading to persistent and hard-to-treat infections. This investigation explored the therapeutic potential of linalool, a naturally occurring monoterpene alcohol, against clinical carbapenem-resistant Acinetobacter baumannii (CRAB) isolates. Linalool demonstrated robust inhibitory effects on both

Biochemical and Structural Characterization of Two-domain Glycoside Hydrolase PgaB from Serratia marcescens and Its Application for S. aureus Biofilm Degradation.

Antimicrobial resistance (AMR) is a critical global health threat, with projections estimating up to 10 million deaths annually by 2050. One of the strategies for developing bacterial AMR is the formation of microbial biofilms (BFs). Thus, enzymes capable of degrading BF exopolysaccharides represent potential tools for BF disruption. In this work, we characterize β-1,6-N-acetylglucosaminidase from Serratia marcescens (SmPgaB), a two-domain enzyme with covalently attached GH153 and CE4 modules. Small-angle scattering data demonstrate that SmPgaB is monomeric in solution. We also demonstrate tha

Pyocyanin produced by Pseudomonas aeruginosa creates legacy effects that promote antibiotic resistance evolution in enterococci.

Polymicrobial infections are small communities of multiple interacting bacterial species. Interactions among constituent species may modify the growth of community members in the presence of antibiotics, for example, via degradation of the antibiotic, the induction of specific resistance mechanisms, or increased mutation supply rates. However, for most polymicrobial infections, the nature of such interactions is opaque, while they may affect both treatment efficacy and the evolution of antibiotic resistance. Here, we describe that past interaction of enterococci with Pseudomonas aeruginosa cre
