The terminal complement proteins C5b-9 augment binding of high density lipoprotein and its apolipoproteins A-I and A-II to human endothelial cells.

Terminal complement protein complexes C5b-9 have been found in human atherosclerotic lesions. Insertion of C5b-9 in the endothelial cell membrane alters permeability, induces membrane vesiculation, and activates secretion. We hypothesized that complement might also alter interactions of the endothelial surface with lipoproteins, particularly high density lipoprotein (HDL), which is reported to inhibit C5b-9-induced hemolysis. We now demonstrate that exposure to C5b-9 increases (by 2- to 50-fold) specific binding of HDL and its apolipoproteins (apo) A-I and A-II to endothelial cells. Binding to

Molecular forms of serum insulin-like growth factor (IGF)-binding proteins in man: relationships with growth hormone and IGFs and physiological significance.

Insulin-like growth factor-I (IGF-I) and IGF-II are associated in the blood with specific binding proteins (BPs), forming complexes that elute in gel filtration with estimated mol wt around 40 and 150 kD. The latter appears to be under GH control. Five molecular forms of BP (41.5, 38.5, 34, 30, and 24 kD) have been identified by Western blotting using 125I-labeled IGF. All five forms are present in the smaller complexes, but only the 41.5- and 38.5-kD forms are found in the larger complexes. In this study immunoblotting showed that the 41.5- and 38.5-kD forms were recognized by antibodies dire

Binding of a monoclonal anti-DNA autoantibody to identical protein(s) present at the surface of several human cell types involved in lupus pathogenesis.

A monoclonal anti-DNA antibody PME77, spontaneously produced in autoimmune B/W mice, has been found to recognize identical protein(s) present at the surface of several human cell types involved in the pathogenesis of systemic lupus erythematosus: glomeruli, platelets, erythrocytes, T and B cells, and neuronal tissue. Data indicate that protein(s) could represent a major stimulus or the target of anti-DNA autoimmunity and could account for tissue lesions observed in this disease.

Studies on the pharmacological bases of fetal toxicity of drugs (IV). Effect of endotoxin and starvation on serum protein binding of salicylic acid in pregnant rats.

Our previous paper reported that the fetotoxic effects of aspirin (ASA) were enhanced by bacterial endotoxin (LPS), and the effects of ASA w … A was removed from serum protein by charcoal treatment. These data suggested the decrease of the SA-protein binding in serum by the increased level of FFA, an inhibitor of the binding, and the decreased level of albumin as a possible mechanism for the potentiation of the fetotoxicities of ASA by LPS.

Decreased binding of drugs and dyes to plasma proteins from rats with acute renal failure: effects of ureter ligation and intramuscular injection of glycerol.

1 The decreased binding of drugs and dyes to plasma proteins from male and female rats with acute renal failure has been investigated using equilibrium dialysis at 37 degrees C. 2 Acute renal failure induced by bilateral ligation of the ureters produced a greater than threefold increase in the unbound fraction of o-methyl red relative to normal rat plasma. Unbound dye concentration in plasma from sham-operated control rats was also significantly increased but to a lesser extent. 3 Glycerol-induced acute renal failure produced a significant increase in the unbound fractions of o-methyl red, met

Serum vitamin B12, serum and red cell folates, vitamin B12 and folic acid binding proteins in women taking oral contraceptives.

Serum and red cell folate, FABP, serum vitamin B12 and vitamin B12 binding proteins were determined in 20 women taking oral contraceptives continuously for at least one year. Studies were also performed on 50 apparently healthy women with history of never taken or not taking oral contraceptives for at least one year as a control group. Serum vitamin B12 level, UBBC and TBBC in women taking oral contraceptives were slightly but not significantly lower than those of the control group. TCI and TCIII increased with a decreased TCII, so that UBBC was within the normal limit. Serum folate was non-si

Inhibitory binding to plastoquinone B site on D1 protein of photosystem II leading to decreased population growth rate via disruption of cellular energy metabolism.

Photosystem II (PSII) is essential for photosynthesis in primary producers, facilitating the primary photochemical reaction to oxidize water and drive the production of ATP and NADPH. A critical interaction within PSII occurs at the plastoquinone B (QB) site on the D1 protein, where electron transfer from QA to QB ensures the continuity of photosynthetic electron flow and energy transduction. PSII-inhibitors, such as diuron, can directly bind to the QB site of the D1 protein, thus hindering the flow of electrons and reducing the synthesis of ATP and other compounds (e.g. NADP) in chloroplasts.

Larger Acute Phase Reactions Are Associated with Immunogenicity of an Adjuvanted Recombinant Receptor Binding Domain Protein Vaccine Against SARS-CoV-2 in Rhesus Monkeys.

BACKGROUND: Although prolonged inflammatory symptoms are an infrequent and problematic adverse effect of vaccination that can occur in some people, the transient activation of acute phase reactants (APRs) is expected with adjuvanted vaccines and helps to potentiate immune responses. METHODS: This experiment examined the association between vaccine reactogenicity and immunogenicity in monkeys immunized with an adjuvanted recombinant protein including a receptor binding domain-human IgG1-Fc fusion protein (RBD-Fc) sequenced from the ancestral Wuhan strain of SARS-CoV-2. The acute inflammatory r

Repurposing the angiotensin II receptor blocker valsartan to inhibit penicillin-binding protein 3 and its mutants in Haemophilus influenzae: a comprehensive in silico approach.

INTRODUCTION: Ampicillin-resistant Haemophilus influenzae (H. influenzae) has been recently designated as a medium-priority bacterial pathogen in 2024 by the World Health Organization (WHO). This pathogen is responsible for a wide range of infections, including sinusitis, acute otitis media, and pneumonia, as well as severe and life-threatening conditions such as bacteremia, meningitis, and epiglottitis. In this context, drug repurposing has emerged as an effective strategy, as the pharmacokinetic properties and safety profiles of approved drugs are already well established, allowing for faste

Proteomic profiling of Juniperus oxycedrus and Cupressus arizonica pollen reveals quantitative interspecies differences in the IgE-binding protein repertoire associated with Mediterranean Cupressaceae allergy.

BACKGROUND: Diagnosis and allergen-specific immunotherapy for Mediterranean allergy rely on extracts standardized to group-1 allergens, such as Cup a 1 from Cupressus arizonica, potentially overlooking interspecies variability. The native species Juniperus oxycedrus, widespread in Spain, may contribute to sensitization, but its clinical relevance and allergen repertoire remain poorly characterized. OBJECTIVE: To characterize and compare the allergen and IgE-binding protein composition of J. oxycedrus and C. arizonica pollen extracts and assess their sensitizing relevance in Spain. METHODS: P

Cysteine desulfurase protects against intracerebral hemorrhage by inhibiting neuronal ferroptosis through the suppression of iron-responsive element-binding protein 2-mediated iron-starvation responses.

BACKGROUND: Ferroptosis is emerging as a crucial type of cellular demise involved in intracerebral hemorrhage (ICH). Cysteine desulfurase (NFS1) is a key gene involved in the regulation of ferroptosis; however, its role in modulating neuronal ferroptosis in ICH remains unclear. This work aimed to explore whether NFS1 influences neuronal ferroptosis in ICH and to explore the underlying molecular mechanisms. METHODS: We established an ICH animal model using collagenase injection and a cellular model by treating neurons with hemin. Neurons with NFS1 overexpression or knockdown were generated usi

Immunoinformatic Prediction of HIV-1 Glycoprotein gp120 and Nef Epitopes Conjugated to HBsAg-Binding Protein (SBP) to Induce the Humoral and Cellular Immune Response.

Acquired Immunodeficiency Syndrome (AIDS) is caused by Human Immunodeficiency Virus (HIV), and continues to be responsible for a substantial number of deaths worldwide each year. Development of a robust and efficient HIV-1 vaccine remains a critical priority. Structural analysis of viral proteins provides a foundational approach to designing peptide-based immunogenic vaccines. In the current experiment, we used computational prediction approaches alongside molecular docking and molecular dynamics (MD) simulations to identify potential epitopes within gp120 and Nef proteins. The selected co-epi

Integrated cortical and plasma proteomic analysis of Cntnap2 knockout mice and human models of autism spectrum disorder: Potential involvement of galectin-3-binding protein.

Despite decades of research, the etiology of autism spectrum disorder (ASD) remains largely uncomprehended, probably due to its clinical and phenotypic heterogeneity. Animal models, particularly contactin-associated protein-like 2 (Cntnap2) knockout (KO) mice, have been instrumental in elucidating ASD-related neurobiological mechanisms, as they exhibit ASD-like phenotypes, such as impaired social interactions in sociability paradigms. This provides the possibility for identifying detectable protein-based biomarkers that may assist in ASD diagnosis. Herein, we implemented an integrated approach

Aptamer Development for SARS-CoV-2 and Omicron Variants Using the Spike Protein Receptor Binding Domain as a Potential Diagnostic Tool and Therapeutic Agent.

Despite various methods for detecting and treating SARS-CoV-2, affordable and easily applicable solutions are still needed. Aptamers can potentially fill this gap. Here, we establish a workflow to identify aptamers that bind to the spike proteins of SARS-CoV-2, a process applicable to other targets as well. The spike protein is crucial for the virus's entry into host cells. The aptamer development process for the spike protein's receptor binding domain (RBD) begins with splitting the SARS-CoV-2's genome into 40 nucleotide-long sequences, predicting their two-dimensional structure, and sorting
