Wharton's Jelly-Mesenchymal Stem Cell-Engineered Conduit for Pulmonary Artery Reconstruction in Growing Piglets.

Surgical treatment of congenital heart defects affecting the right ventricular outflow tract often requires complex reconstruction and multiple reoperations. With a randomized controlled trial, we compared a novel tissue-engineered small intestine submucosa-based graft for pulmonary artery reconstruction (seeded with mesenchymal stem cells derived from Wharton's Jelly) with conventional small intestine submucosa in growing piglets. Six months after implantation, seeded grafts showed integration with host tissues at cellular level and exhibited growth potential on transthoracic echocardiography

An in-body networking system for communication between wearable and implantable therapeutics.

Networks of bioelectronic sensors and actuators enable closed-loop therapies that connect distant, related physiological dynamics. Currently, device interactions are limited by communication methods that inefficiently penetrate tissue and require bulky components. Inspired by the ionic signaling of the nervous system, we engineered a communication platform that uses tissue as a conductive medium to transmit signals among implants and wearables through epidermal, subcutaneous, intraperitoneal, and gastrointestinal spaces. Devices generate transient electric fields that selectively activate othe

Novel chitosan derivative based composite scaffolds with enhanced angiogenesis; potential candidates for healing chronic non-healing wounds.

The success of wound healing depends upon the proper growth of vascular system in time in the damaged tissues. Poor blood supply to wounded tissues or tissue engineered grafts leads to the failure of wound healing or rejection of grafts. In present paper, we report the synthesis of novel organosoluble and pro-angiogenic chitosan derivative (CSD) by the reaction of chitosan with 1,3-dimethylbarbituric acid and triethylorthoformate (TEOF). The synthesized material was characterized by FTIR and 13C-NMR to confirm the incorporated functional groups and new covalent connectivities. Biodegradability

Activin receptor type IIA/IIB blockade increases muscle mass and strength, but compromises glycemic control in mice.

PURPOSE: Blocking the Activin receptor type IIA and IIB (ActRIIA/IIB) has clinical potential to increase muscle mass and improve glycemic control in obesity, cancer, and aging. However, the impact of blocking ActRIIA/IIB on strength, metabolic regulation, and insulin action remains unclear. METHODS: Here, we investigated the effect of short- (10 mg kg-1 bw, once, 40h) or long-term (10 mg kg-1 bw, twice weekly, 21 days) antibody treatment targeting ActRIIA/IIB (αActRIIA/IIB) in lean and diet-induced obese mice and engineered human muscle tissue. RESULTS: Short-term α ActRIIA/IIB administratio

Bone Morphogenetic Protein-9 Is a Potent Chondrogenic and Morphogenic Factor for Articular Cartilage Chondroprogenitors.

Articular cartilage contains a subpopulation of tissue-specific progenitors that are an ideal cell type for cell therapies and generating neocartilage for tissue engineering applications. However, it is unclear whether the standard chondrogenic medium using transforming growth factor beta (TGFβ) isoforms is optimal to differentiate these cells. We therefore used pellet culture to screen progenitors from immature bovine articular cartilage with a number of chondrogenic factors and discovered that bone morphogenetic protein-9 (BMP9) precociously induces their differentiation. This difference was…

Investigating the foreign body response and regenerative mechanisms in medical-grade polycaprolactone scaffold guided breast reconstruction in a porcine model.

To enhance the clinical use of scaffold-guided breast reconstruction (SGBR), the porcine model provides a reliable means of assessing scaffold biocompatibility and tissue regeneration. Understanding the immunological processes triggered by the implantation of medical-grade polycaprolactone (mPCL) scaffolds is essential for elucidating the underlying biology and optimizing the regenerative potential of tissue engineering and regenerative medicine (TE&RM) technologies. This observational work aims to characterize these in vivo processes through systematic, comprehensive immunohistochemical (IHC)…

Influence of storing muscle tissue in the cold on in vitro myogenic cell line development from the black sea bream (Acanthopagrus schlegelii).

With advances in biotechnology and tissue engineering, the development of fish muscle cell lines from aquatic resources has gained increasing attention. However, most studies rely on immediate tissue processing after sampling, and few studies have investigated the potential of establishing cell lines from stored samples. In this study, we evaluated the potential for establishing skeletal muscle cell lines from black sea bream (Acanthopagrus schlegelii) tissue stored at 4 ℃ for 0, 2, 5, 10, 20, and 30 days. Primary culture was conducted using two methods: explant culture and dissociated culture…

Additive manufactured polyether-ether-ketone composite scaffolds with hydroxyapatite filler and porous structure promoted the integration with soft tissue.

Additive Manufactured (AM) Polyether-ether-ketone (PEEK) orthopaedic implants offer new opportunities for bone substitutes. However, owing to its chemical inertness, the integration between PEEK implants and soft tissue represents a major challenge threatening the early success of the PEEK implants. Here we investigated the influence of hydroxyapatite (HA) fillers and porous structure of AM HA/PEEK scaffolds on the integration with soft tissue through in-vitro cellular experiments and in-vivo rabbit experiments. Among the animal experiments, HA/PEEK composite scaffolds with HA contents of 0, 2

Nondestructive quality control across the life cycle of bioprinted products.

Bioprinting is a transformative technology paving the way toward complex tissue-engineered products, yet its translation is constrained by major quality control challenges. Variability in cells, bioinks, bioprinting parameters, and tissue maturation can compromise reproducibility and product consistency. Addressing such variability requires repeated measurements across the multistage, multifactorial bioprinting pipeline that conventional destructive assays cannot provide. In this opinion article, we argue that nondestructive testing (NDT) should become an enabling component of reliable bioprin

Use of dermal-like tissue in the management of chronic and acute full-thickness cutaneous wounds.

This paper presents a dermal replacement technique for cutaneous wounds of diverse origin. An autologous tissue-engineered dermal-like structure was implanted in one patient with an acute surgical excision of a nevus and in another patient with a chronic post-traumatic ulcer. The dermal constructs were well integrated with the wound area, and, at 3 weeks, an epidermal covering was implanted. In both cases, 3 to 8 weeks after autografting, good integration of the transplanted epidermis with the surrounding native skin and a good take over the dermal-like tissue occurred. The use of this technol

Protocol for the Implantation of Scaffolds in a Humanized Mouse Cutaneous Excisional Wound Healing Model.

Tissue-engineered constructs combine the mechanical properties of biomaterials with biological agents to serve as scaffolds that direct the wound-healing process and promote tissue regeneration. A limitation to studying wound healing in vivo is that mouse skin contracts to heal rather than exhibiting granulation tissue formation and epithelialization like human skin. Therefore, it became necessary to develop a mouse model to better recapitulate human wound healing. The first splinted excisional wound healing model in mice, described in 2004, utilized silicone splints to prevent skin contractur

The allogeneic response to cultured human skin equivalent in the hu-PBL-SCID mouse model of skin rejection.

BACKGROUND: Engineered tissues have been proposed for the treatment of a variety of conditions including the partial or complete replacement of human organs. To determine the basis for the rejection of these tissues, we analyzed the immune response to allogeneic human skin equivalent (HSE, also called Apligraf) in the humanized SCID mouse (hu-PBL-SCID). METHODS: Two models of hu-PBL-SCID were used for these studies. In one model, human skin or HSE was transplanted onto humanized mice so that graft survival could be analyzed. In the other model, skin grafts were allowed to heal on naive mice b

Structural and Molecular Changes of Human Chondrocytes Exposed to the Rotating Wall Vessel Bioreactor.

Over the last 30 years, the prevalence of osteoarthritis (OA), a disease characterized by a loss of articular cartilage, has more than doubled worldwide. Patients suffer from pain and progressive loss of joint function. Cartilage is an avascular tissue mostly consisting of extracellular matrix with embedded chondrocytes. As such, it does not regenerate naturally, which makes an early onset of OA prevention and treatment a necessity to sustain the patients' quality of life. In recent years, tissue engineering strategies for the regeneration of cartilage lesions have gained more and more momentu…

Pre-cultivation of adipose tissue-derived microvascular fragments in porous scaffolds does not improve their in vivo vascularisation potential.

Adipose tissue-derived microvascular fragments represent promising vascularisation units for implanted tissue constructs. However, their reassembly into functional microvascular networks takes several days, during which the cells inside the implants are exposed to hypoxia. In the present study, we analysed whether this critical phase may be overcome by pre-cultivation of fragment-seeded scaffolds prior to their implantation. Green fluorescent protein (GFP)-positive microvascular fragments were isolated from epididymal fat pads of male C57BL/6-TgN (ACTB-EGFP) 1Osb/J mice. Nano-size hydroxyapati
