A vascularized tumoroid model for human glioblastoma angiogenesis.

Glioblastoma (GBM) angiogenesis is critical for tumor growth and recurrence, making it a compelling therapeutic target. Here, a disease-relevant, vascularized tumoroid in vitro model with stem-like features and stromal surrounds is reported. The model is used to recapitulate how individual components of the GBM's complex brain microenvironment such as hypoxia, vasculature-related stromal cells and growth factors support GBM angiogenesis. It is scalable, tractable, cost-effective and can be used with biologically-derived or biomimetic matrices. Patient-derived primary GBM cells are found to clo

TUG1 Promoted Tumor Progression by Sponging miR-335-5p and Regulating CXCR4-Mediated Infiltration of Pro-Tumor Immunocytes in CTNNB1-Mutated Hepatoblastoma.

INTRODUCTION: HB presents with the highest frequency of CTNNB1 mutations, resulting in activation of Wnt signaling pathway. A number of studies have demonstrated CTNNB1 mutation contributed to the development of HB. However, limited research explored the function of lncRNAs in HB with CTNNB1 mutation. METHODS: We screened lncRNA expression profiles in CTNNB1-mutated HB samples and identified lncRNAs associated with malignant phenotype in HB. The association between lncRNA and immune microenvironment was investigated. The biological function of lncRNA was further explored using in vitro experi

The direct and indirect effects of corticosterone and primary adipose tissue on MCF7 breast cancer cell cycle progression.

BACKGROUND: Breast cancer is the second leading cause of cancer-related mortality in women. Glucocorticoids (GCs) have the potential to directly affect breast cancer or indirectly via changes to the tumor growth microenvironment a breast cancer is exposed to. The role of GCs in breast cancer progression by direct and indirect means are not fully understood. AIM: To study the direct and indirect effects of GCs on breast cancer cell cycle regulation. METHODS: MCF7 breast cancer cells were incubated with increasing concentrations of corticosterone (CORT) to investigate the direct effects. In ad

T-cell/histiocyte-rich B-cell lymphoma: biology, diagnosis, and management.

T-cell/histiocyte-rich B-cell lymphoma (T/HRBCL) is an uncommon morphologic variant of diffuse large B-cell lymphoma (DLBCL). Pathologically, it is distinguished by <10% malignant B cells amid a majority population of reactive T lymphocytes and histiocytes. Diagnosis of this entity is occasionally difficult, as it may appear similar to other lymphoid diseases, such as nodular lymphocyte-predominant Hodgkin's lymphoma and classic Hodgkin's lymphoma. Accurate diagnosis therefore rests with careful immunohistochemical analysis of the tumor cells and the inflammatory microenvironment. Clinically,

[Evaluation of Inflammatory Cells (Tumor Infiltrating Lymphocytes) in Solid Tumors].

BACKGROUND: The tumor microenvironment plays an important role in tumorigenesis and the tumor-host relationship. An important part of the tumor microenvironment is inflammatory infiltration. Its evaluation in solid tumors has prognostic meaning and appears also to be predictive of outcome, which is particularly important for predicting responses to immune checkpoint inhibitors. However, the methodology used to assess inflammatory infiltration is problematic, because it has been standardized only for certain types of tumors. OBJECTIVE: The present study provides an overview of current issues r…

Exploiting Tumor Cell Invasiveness to Construct an Isolated Niche for Postoperative Immunotherapy.

Residual tumor cells persisting in the surgical bed are the principal drivers of postoperative tumor recurrence, yet the immunosuppressive wound healing microenvironment limits immune activation and compromises antigen enrichment and cross-presentation. To address this issue, we exploited tumor cell invasiveness to construct isolated niches that confine the cells in micron-scale chambers shielded from external microenvironmental perturbations. For proof of principle, we fabricated a hyaluronic acid-based double-network hydrogel comprising HAMA and PEGDA. Hyaluronidase-dependent invasive tumor

Living human lung slices for ex vivo modelling of lung cancer.

The tumor microenvironment (TME) markedly affects cancer progression, yet traditional animal models do not fully recapitulate the situation in humans. To address this, we developed tumor-derived precision lung slices (TD-PCLS), an ex vivo platform for studying the lung TME and evaluating therapies. TD-PCLS, viable for 8-10 days, preserve the heterogeneity and metabolic activity of primary tumors, as confirmed by seahorse analysis. Using multispectral FACS and phenocycler multiplex imaging, we spatially profiled TME components and cancer cell functionality. Additionally, TD-PCLS revealed patien…

Quantitative assessment of extravasation of IL-15-secreting MSLN-CAR-NK-92 cells using tumor transparency imaging.

BACKGROUND: The extravasation of anticancer immune cells is a very important issue that must be understood to improve the anticancer effect of chimeric antigen receptor (CAR)-expressing anticancer immune cell therapy. To date, no study has been reported to quantitatively evaluate the degree of extravasation of anticancer immune cells escaping from tumor blood vessels to the tumor microenvironment (TME) at the microscopic level. METHODS: In this study, for the first time, using tumor transparency imaging, the extent of extravasation of CAR-NK and NK cells in pancreatic tumors was determined. w…

Co-delivery of IL-12/IL-15/IL-18 engineered DC vaccines with anti-IL-10R and nanoconjugated methotrexate in melanoma.

BACKGROUND: In an immunosuppressive microenvironment created by melanoma cells, interleukin (IL)-10 can promote tumor growth and impair the function of antigen-presenting cells, particularly dendritic cells. One of the leading strategies to counteract IL-10's action is the administration of antibodies against its receptor. The tumor microenvironment can also be modulated by cytokines and/or cellular vaccines such as modified dendritic cells that overproduce IL-12, IL-15/IL-15Rα, and IL-18. These cellular vaccines serve as a source of cytokines and stimulate the immune system by presenting tumo…

Increased expression of IRF8 in tumor cells inhibits the generation of Th17 cells and predicts unfavorable survival of diffuse large B cell lymphoma patients.

The immunological pathogenesis of diffuse large B cell lymphoma (DLBCL) remains elusive. Searching for new prognostic markers of DLBCL is a crucial focal point for clinical scientists. The aim of the present study was to examine the prognostic value of interferon regulatory factor 8 (IRF8) expression and its effect on the development of Th17 cells in the tumor microenvironment of DLBCL patients. Flow cytometry, immunohistochemistry, and quantitative real-time PCR were used to detect the distribution of Th17 cells and related cytokines and IRF8 in tumor tissues from DLBCL patients. Two DLBCL ce…

Macrophage type 2 differentiation in a patient with laryngeal squamous cell carcinoma and metastatic prostate adenocarcinoma to the cervical lymph nodes.

BACKGROUND: The tumor microenvironment often polarizes infiltrating macrophages towards a type 2, or M2 phenotype, that is characterized by expression of various cysteine-rich, scavenger receptors, including CD163. The primary function of M2 macrophages is to facilitate wound healing. As such, they are capable of providing metabolic support to a growing tumor, neovascularization, as well as protection from cytotoxic T cells. The tumor microenvironment contains a milieu of secreted factors and vesicles, which in certain circumstances can gain access to lymphatic vessels that drain to local lymp…

Mimicking tumor hypoxia in a glioblastoma-on-a-chip.

Glioblastoma (GBM) is one of the most aggressive and lethal brain tumors, characterized by limited therapeutic options and poor prognosis. This condition is strongly linked to the limitations of current preclinical models: traditional two-dimensional cell cultures and animal models often fail to recapitulate the complexity of the tumor microenvironment (TME), resulting in limited predictive value for clinical outcomes. Here, a biomimetic three-dimensional (3D) in vitro culture system is presented, designed to incorporate key components of the TME and to mimic human GBM biology by: (i) employin…

Elevated Lactate in the AML Bone Marrow Microenvironment Polarizes Leukemia-Associated Macrophages via GPR81 Signaling.

Interactions between acute myeloid leukemia (AML) and the bone marrow microenvironment (BMME) are critical to leukemia progression and chemoresistance. In the solid tumor microenvironment, altered metabolite levels contribute to cancer progression. We performed a metabolomic analysis of AML patient bone marrow serum, revealing increased metabolites compared to age- and sex-matched controls. The most highly elevated metabolite in the AML BMME was lactate. Lactate signaling in solid tumors induces immunosuppressive tumor-associated macrophages and correlates with poor prognosis. This has not yet…
